    Evaluation of the analgesic activity of quercetin and chrysin in animal models

    Pain is relieved by analgesics that acts via modulating the central and peripheral pain mediators. Some of the analgesic has been reported to have a serious adverse effects when used in long-term treatments. Therefore, there is a need of developing a potent analgesic with minimal of no adverse effects. In the current study, quercetin and chrysin were evaluated for the analgesic activity in mice and rat by hyperalgesia (Eddy’s hot plate and Tail flick analgesiometer). Diclofenac and tramadol was used as a standard reference drugs. Male Swiss Albino mice and rats are divided into six groups and treated with vehicle (1% acacia), quercetin (40 mg/kg), chrysin (100 mg/kg), chrysin (200 mg/kg), diclofenac (10 mg/kg) and tramadol (10 mg/kg) orally. In the present study, diclofenac, tramadol, quercetin and chrysin significantly increased the tail flick latency in the tail flick pain model compared to control group. Chrysin showed remarkable analgesic activity at 200 mg/kg compared to 100 mg/kg in Tail Flick and Eddy’s hot plate model. The current study also reported that diclofenac has more analgesic activity than tramadol on both 1st and 7th day of treatment.

    Diabetes is a non-communicable disease which is attaining increasing importance among the adult population in both developed and developing countries. Pre-diabetes is a precursor condition for type 2 Diabetes mellitus. Although in many cases it is reversible, Pre-diabetes frequently remains undiagnosed and therefore risk of developing type 2 Diabetes Mellitus is increased. Lifestyle modification is more effective mode of preventing diabetes and reduction of 40%-70% with pre-diabetes. While there is increasing evidence to prove the efficacy of pharmacotherapy in prevention of diabetes in adults with pre-diabetes, pharmaceutical treatment options other than metformin are associated with adverse effects that limit their use for pre-diabetes. It is considered to be an at risk state, with high chances of developing diabetes. While, pre-diabetes is commonly an asymptomatic condition, there is always presence of pre-diabetes before the onset of diabetes. This aim of this study is to describe the challenges associated with diagnosis of pre-diabetes, the possible adverse medical outcomes associated with pre-diabetes and the treatment options and rationale for their use in context of pre-diabetes.     Assessment of Nephroprotective Activity of PippalyadiAgad on Diclofenac Sodium Induced Nephrotoxicity in Albino Mice: An Experimental Study

    Background: Nephrotoxicity due to Diclofenac Sodium can be correlated with this concept of Dushi Visha and nephroprotective drugs mentioned in Ayurveda texts may play an important role in this area. Pippalyadi Agad is the herbo-mineral medication stated by Yogaratnakara for treatment of Dushi Visha. Some of the drugs in this formulation have shown efficacy in correcting nephrotoxicity. Objective: To study the nephroprotective activity of Pippalyadi Agad on diclofenac sodium induced nephrotoxicity in albino mice. Methods: The experimental study was carried out in albino mice as per CPCSEA guidelines after getting approval from the Institutional Animal Ethics Committee (IAEC). 30 albino mice, each weighing about 20-30 gm, were procured from Bharat serum and vaccines. The mice were housed in the institutional animal house in standard conditions. 30 albino mice were equally divided into five groups having six animals in each group as Trial Drug (low dose), Trial Drug (medium dose), Standard Control, Disease Control and Vehicle Control. For assessment of nephrotoxicity, the animals were monitored on Day 0, Day 15 and Day 45 by carrying out blood investigations such as BUN, Serum Creatinine, Body weight and CBC. The Data were subjected to appropriate statistical tests to derive results. Results: Pippalyadi Agad treatment at low and medium doses significantly lowered BUN and Serum Creatinine in comparison with Disease and Standard Control at Day 45. Pippalyadi Agad treatment at low and medium doses significantly (p < 0.001 and p < 0.001) increased body weights of the animals in comparison with Disease and Standard Control at Day 45. No significant difference in CBC parameters was observed in any of the Groups except the Disease Control Group on Day 15 and Day 45. Conclusion: Pippalyadi Agad has shown nephroprotective activity based on biochemical changes observed in Serum Creatinine, BUN and CBC.

    A clinical study on the management of Arshas (Prolapsed Piles) by Jaloukavacharana

    Background: Arshas is one among the Mahagadas (major diseases) which causes pain and obstruction in the anal passage. It is a rakta and mamsa doshaja vyadhi (disease related to blood and muscle). Arshas can be correlated to hemorrhoids. It is caused due to the varicosity of hemorrhoidal plexus which makes it a surgically curable disease. In Ayurveda, various treatments have been mentioned for the management of arshas (hemorrhoid) such as chedhana (excision), sweda (sudation), etc among which Visravana or raktamokshana (blood-letting therapy) plays an important role. Jalaukavacharana (leech therapy), a type of Asashtra krita Visravana (non-invasive blood-letting) has been mentioned as an alternate, non-surgical measure in the management of arshas (hemorrhoid). The arshas (hemorrhoids) which are swollen, prolapsed and containing more quantum of stagnated blood inside can be managed by leech therapy. The saliva of leech contains an anti-coagulant factor – Hirudin and a spreading factor – Hyaluronidase, which act as anesthetic and anti-inflammatory, thereby helping in the treatment of arshas (hemorrhoid). In the present study, raktamokshana (blood letting) was performed with jalauka or leech in patients with prolapsed pile mass. Materials and method: 30 patients fulfilling the assessment criteria were selected for the study. Result and conclusion: In the present study 26 patients were completely cured, 3 patients were cured with presence of mild symptoms and 1 patient had improvement from severe symptoms to moderate symptoms. Therefore, Jalaukavacharana (leech therapy) can give promising results in the management of prolapsed piles

    Validation of hospital environmental disinfection efficiency of herbal fumigants

    Background: Healthcare associated infections includes infestation by bacteria, viruses and fungi. As per WHO estimates, approximately 15% of all hospitalized patients suffer from these nosocomial infections caused by these micro-organisms. Outbreaks of such infections increases the use of antibiotics, vitamins and immonomodulator medicines. During ancient times, to prevent spread of infections; fumigation with herbs was a practice and it is still seen continued. Material & methods: Good quality herbal fumigants like Guggulu gum-resin, turmeric powder, spikenard rhizome, white mustard fruits, caraway fruits, camphor resins and cow’s ghee were used for fumigation of various hospital rooms like out-patient department room, dental clinic, gynecological procedure room, physiotherapy and operation theatre of Community Healthcare Center, Sarsa. Swabs from these units were tested for bacterial and fungal load before and after experiment by using culture media’s like nutrient broth and potato dextrose agar respectively. Results: The bacteria culture reported organisms namely Bacilus subtilis, Bacilus megaterium, Staphylococcus aureus, Micrococcus luteus and fungi like Aspergillus niger, mucor and rhizophus. After 30 minutes of fumigation procedure, significant reduction in bacterial colonies was found in gynecological procedure room, dental clinic, OPD and physiotherapy room while statistically non-significant results were observed in operation theatre. Two hours after fumigation, statistically significant result was observed in bacterial count in all studied units except operation theatre while fungal colonies reduction was achieved in all studied units. Conclusion: Fumigation shall be encouraged in hospital units like out patients department, wards and minor procedure rooms but may not be perfectly suitable for operative theatres always. The study encourages daily herbal fumigation of home and surroundings for hygienic and pleasant environment

    An Insilico Computational Screening of  Siddha Formulation Kalingathy Thailam against Cyp- 17α-Hydroxylase in PCOS

    Background: PCOS is characterized by ovulatory failure, hirsutism, obesity, glucose intolerance, resistance to insulin, dyslipidemia and infertility. Siddha system of medicine has a worthwhile therapeutic effect in treating PCOS. The bioactive compounds of Kalingathy Thailam mentioned in Siddha literature was selected for evaluating their ability to inhibit CYP- 17α-hydroxylase. Objective:  The study is aimed to perform the In Silico computational screening of the formulation Kalingathy Thailam against the target enzyme CYP-17 α -hydroxylase in PCOS. Methods: Autodock program was used against the target enzyme CYP-17 α -hydroxylase  for the phytocomponents such as Sinapic acid, Cucurbitacin B of Citrullus colocynthis (L. ) Schrader, Aromadendrene, Linolenic acid of Melia dubia Linn. , Onionin A, Protocatechuic acid of Allium cepa Linn. , Ascorbic acid, Limonene of Citrus limon (L. )Burm. f. , Ricinine of Ricinus communis Linn. Results: A total of 9 components were screened, out of these Protocatechuic acid, Ascorbic acid, Linolenic acid and Cucurbitacin B reveals maximum of 2 to 3 interactions accounting for 75-100% binding efficacy with the core active amino acid residues present on the target enzyme CYP- 17α-hydroxylase. Conclusion: Based on the results of the computational analysis it was concluded that the bio-active compounds present in the herbal ingredients possess significant binding against the target enzyme CYP-17α-hydroxylase by interacting with active amino acids. Hence these phytocomponents which inhibit the target enzyme may act as a potential therapeutic agent for management of PCOD.

    Ayurveda Management in Diabetic Neuropathy vis-à-vis Prameha Upadrava–A Case Study

    Background- Diabetes mellitus (DM) in India has a higher prevalence (4.3%) than in the west (1-2%) and its complication rate is also high.  Diabetic neuropathy symptoms include pain, numbness, reduced sensation of touch or oversensitivity, uncomfortable tingling and burning, skin ulcer and reduced reflexes. Diabetic neuropathies are seen in approximately 50% of individuals with long standing type 1 and type 2 DM and it often goes undiagnosed. This article presents a case study of patient with signs and symptoms of neuropathy who came with the complaint of lower back pain, bilateral lower limb pain, pricking sensation and burning sensation. Diabetic Neuropathy Examination score (DNE) along with other neuropathy screening parameters was more than 60% indicating functional loss. The Nerve conduction study also revealed demyelinating polyneuropathy. Observations and Results: The patient was treated with ayurvedic oral medication, Abhyanga (~application of medicated oils), swedana (~fomentation), and basti (~medicated enema). After ayurvedic treatment the patient showed good remission in pain, burning sensation and DNE along with all neuropathy screening were less than 20%. Patient got marked relief in all the symptoms

    The existence of life on Earth is sustained by the act of reproduction. The ultimate blessing for a couple in their lifetime is to give birth to a healthy progeny, which is only possible by the union of healthy beeja in the favorable conditions of Ritu, kshetra and ambhu. Female reproductive health plays a key role in all these four factors. Good reproductive health ensures a female to have a satisfactory and safe sexual life both physically and mentally. Rasayana prayoga during the course of dhatu utpatti in balya and madhyamavastha helps to attain the rasadi dhatu at its supreme quality. Acharya Sharangadhara quotes that the development of certain bodily characteristics attained in its pinnacle in every decade of life, starts to decline gradually after that certain period of years naturally. Rasayana therapy administered in each reproductive stage of female life may improve her reproductive health qualitatively which intern helps them to prevent various common reproductive aliments from balyatara avastha (pre-menarche) to Jara (complications after menopause). In this article, an attempt is made to collect and analyze the relevant facts from Samhitas, contemporary textbooks and e-resources and tried to conceptualize the significance of administering decade-wise Rasayana in female for improving their reproductive health.

    Madhuca longifolia var. latifolia (Roxb.) A.Chev: A plant with medicinal boon

    Throughout human history, people used to obtain drugs from natural resources like plants to cure their ailments and improve their health. Nowadays human beings rely more on herbal medicines for the treatment of various ailments because they show better therapeutic activity and have very little to zero side effects on the human body. Madhuca longifolia is also one of the plants used in this category. The plant has been used by people for its medicinal properties since the Ayurveda period (before B.C 300). The plant possesses various medicinal activities including anti-proliferative, anti-inflammatory, anti-bacterial, anti-cancer, anti-ulcer and antidiabetic, and antioxidant activity. In various parts of India, the plant is used in the treatment of different diseases in ethnomedicinal practices. The plant possesses several phytochemicals which are responsible for its medicinal properties.  The primary focus of this manuscript is to emphasize the significance of diverse ethnomedical characteristics, multiple preclinical investigations, and clinical trials conducted on this particular plant.     Standardization of Dooshivishahari Agada through HPTLC

    Ayurveda medicines are a time-tested and valuable resource for healing, even today, globally. Hence the quality control standards for the Ayurvedic formulations is the need of time because of commercialization. Despite the availability of modern equipment and techniques, not all  Ayurvedic formulations have been standardized as per modern day protocol. Dooshivishari agada (DVA), a herbo-mineral preparation; mentioned in the text of Asthangha hrudaya in the context of treatment of poisoning, specifically for dooshivisha (cumulative poison). High-performance thin-layer chromatography (HPTLC) is a validated analytical tool for qualitative and quantitative analysis of chromatographic information. In this present study, an effort has been made to standardize DVA through HPTLC. HPTLC photo documentation of DVA showed 8, 11, and 10 spots under short UV, long UV, and under white light after derivatization respectively. Densitometric scan at 254 nm revealed 3 high peaks corresponding to 3 different compounds in the ethanol extract, compounds, at 366 nm and 620 nm there were three high peaks. These Physico-chemical constants, TLC photo documentation, the unique Rf values, and densitogram obtained at different wavelengths can be used as a fingerprint to identify DVA
