In Vitro Immunomodulatory Activity of Sagalanoi Chooranam (Poly Herbal Formulation) in RAW Macrophage Cell Line
Within the realm of Siddha medicine, an ancient and venerable traditional healing system hailing from India, a comprehensive methodology is deployed to combat liver disorders. This approach is characterised by the utilisation of herbal remedies, accentuating the reinstatement of equilibrium in the body\u27s vital life force, known as "Vatham," "Pitham," and "Kapham." The utilisation of cell line studies is crucial in assessing the immunomodulatory properties of herbal medicine, offering invaluable insights into their capacity to influence the immune system and advancing our comprehension of their therapeutic benefits. The primary objective of this investigation was to elucidate the potential immunomodulatory properties of sagalanoi chooranam, a polyherbal Siddha formulation, through the execution of anti-proliferative assays conducted. On the RAW 264.7 macrophage cell line. Various test solutions were precisely prepared, encompassing a spectrum of concentrations (50, 100 and 200 μg/ml). The RAW 264.7 cells were diligently cultured and then carefully subjected to stimulation with lipopolysaccharide (LPS) to initiate cellular activation. Following this activation, the test formulation was introduced at varying concentrations, after which the cells underwent a 24-hour incubation period. The resultant nitrite levels, serving as indicators of immunomodulatory response, were evaluated within the cell lysate. The outcomes unveiled a notable decline in nitrite levels, correlating with the dosage of the test formulation during the incubation with RAW 264.7 cells. The outcomes of the study elucidate a conspicuous decline in nitrite levels in direct correlation with escalating concentrations of the test formulation, contrasting starkly with the control group exclusively subjected to LPS. Furthermore, the investigation meticulously probed the ramifications of the test formulation on cellular viability. The vitality of RAW 264.7 cells exhibited a discernible downward trajectory as the concentration of the test formulation ascended. Noteworthy is the fact that, at the concentration of 200 μg/ml, cellular viability registered at a mere 55.53 ± 3.567%. Collectively, these findings lend credence to the hypothesis that the test formulation possesses a dose-dependent capacity to attenuate both nitrite levels and cellular viability within the RAW 264.7 cell milieu, underscoring its auspicious immunomodulatory attributes. This research expands our insights into the potential immunological ramifications of the test formulation and underscores its plausible utility within the realm of immunomodulation.
Case report of Shwetapradara in Ayurveda w.s.r. to Leucorrhoea
Ayurveda is an ancient science, which deals with the various treatments, its prevention and various principles for management of diseases are described in Ayurveda. Ayurveda has mentioned Ashtanga Ayurveda in which Balaroga under which Streeroga and Prasruti Tantra was mentioned. Shwetapradara is one of the important disorder found in gynaecological practice. It can be correlated with Leucorrhoea in modern science. Shwetapradara is a condition which is characterized by Vaginal discharge (Yoni Srava) without any pain, discomfort and burning sensation, Vaginal Itching (Yoni kandu) etc. It is mentioned as one of the symptom presents in many gynaecological disorders. Ayurveda has mentioned various treatments for the management of Shwetapradara in which Shamana Chikitsa is important line of treatment. In-Silico Anti-Diarrhoeal Evaluation of Lagu Gangathara Chooranam
Background: In siddha system of medicine diarrhoea can be compared with kazhichal. Diarrhoea results from several factors like intestinal hyper-motility, malabsorbtion of water, inflammation of GIT or use of laxatives. In siddha literature, the preparation named lagu gangathara chooranam is indicated for kazhichal. Aim: The study aims to perform the in-silico computational studies of phytoconstituents of the siddha formulation lagu gangathara chooranam targeted against m3 muscarinic acetyl choline receptor for anti-diarrhoeal activity. Methods: The retrieved phytocomponents were subjected for docking calculations against target enzyme m3 muscarinic acetylcholine receptor. Results: The computational analysis retrived 11 bio-active compounds from herbal formulation. It includes limonene, α-humulene, cyperolone, lupeol, beta amyrin, betulinic acid, mangiferin, quercetin, caryophyllene and luteolin. These compounds significantly bind against the target m3 muscarinic acetylcholine receptor. Hence these phytocompounds may produce significant anti-diarrheal activity by hindering the activity of m3 muscarinic acetylcholine receptor in the intestinal region which mediates the diarrhoea. Conclusion: Further clinical trials or experimental studies could be conducted to confirm the effectiveness of this siddha formulation lagu gangathara chooranam against diarrhoea.
A comparative clinical study on Matra Basti with Kshirabala Taila and Ketakadya Taila in Janu Sandhigata Vata
Osteoarthritis is a prevailing form of Arthritis, also known as Degenerative Joint Disease, which occurs due to the gradual deterioration of joints, including the articular cartilage and subchondral bone. Common symptoms of osteoarthritis include joint pain, stiffness, and reduced range of motion, leading to a loss of flexibility in the affected joints. In modern medical science, NSAIDs are commonly prescribed to manage the symptoms of Osteoarthritis which help in reducing pain, inflammation, and stiffness. NSAIDs only provide symptomatic relief to the patients. Indeed, Sandhigatavata and Osteoarthritis are closely correlated terms, with Sandhigatavata being the Ayurvedic term for Osteoarthritis. Ayurvedic texts suggest various treatments for this condition, with the most common ones being repeated application of Snehana and Swedana, along with Basti and Mriduvirechana. Among these treatments, Basti is considered a crucial part of the overall therapy, and the Matra Basti, a specific form of Basti, is relatively easier to administer without any complications. Here an attempt was made through Matra Basti with Kshira Bala Taila and Ketakadya Taila mentioned in Vata Vyadhi Chikitsa specifically. This is an open-label, observational, and clinical research study, a total of 30 patients diagnosed with Sandhigata Vata were randomly selected from the OPD and IPD of Parul Ayurved Hospital and Khemdas Ayurved Hospital in Waghodia, Vadodara. These patients were divided into two groups i.e., in Group A, 15 patients were given Abhyanga, Baspa Swedana followed by Matra Basti with Kshira Bala Taila, and in Group B, 15 patients were given Abhyanga, Baspa Swedana followed by Matra Basti with Ketakadya Taila for 15 days. The assessment of patients was carried out before treatment, on the 8th day, after treatment, and on the 30th day of treatment. Randomised Controlled Trial on the efficacy of Mamajjaka (Enicostemma littorale Linn.) in the Management of Madhumeha (Diabetes Mellitus)
Diabetes Mellitus is in top ten list of diseases causing mortality. In the year 2011, estimated 366 million people were found to be suffering from diabetes mellitus. By year 2030 this morbidity data will rise to approximately 552 million people worldwide. India is having the largest number of diabetics in the world and gets the name ‘Diabetic capital’. This study was carried out with an aim:-‘To Conduct a Randomised Controlled (Upashayatmaka) Trial on the role of Mamajjaka capsule in the management of Madhumeha (DM type-2)’. In this study total 110 patients of Diabetes were selected from NIA hospital and randomly divided into two groups. Group A (Control group) comprise 54 patients were administered with Tab. Metformin and dose was 500mg tablet 1 BD before meals as advised according to allopathic consultant. Group B (Trial group) comprise 56 patients were administered with Cap. Mamajjaka, dose: 500 mg Capsule 2 BD, before 30 min. of meal with lukewarm water. Total duration trial was 100 days. Results obtained in subjective and objective parameters were analysed for the statistical significance by adapting paired T test and unpaired T test. The study revealed that Group-B was found to be more effective in bringing symptomatic relief and Group-A was found to be more effective in improving biochemical markers in the patients of Madhumeha. Cap. Mamajjaka (Enicostemma littorale Linn.) provided better results, suggests that this can be a good complimentary medicine for the patients who are not getting good relief from modern medicine alone
Development of tools of Prakriti assessment in neonates- A cross sectional study
Prakriti (body constitution) is amalgamation of psychosomatic traits. The inherited features depend upon shukra(sperm) and shonita(ovum), whereas acquired features develop due to environmental elements. Aim of this contemporary study is to assess Prakriti of neonates by psychosomatic and neurological characteristics and objectives are to prepare a comprehensive proforma to assess Prakriti (body constitution) in neonates by using subjective criteria, psychosomatic and neurological characteristics. Material & methods: A total of 100 neonates fulfilling inclusion criteria were enrolled from OPD, IPD of MGACH & RC, DMIHER and NICU, A.V.B.R.H., Sawangi, Wardha, Maharashtra and this prevailing study was conducted as a survey study using a validated questionnaire. Compilation of a comprehensive list of distinctive traits (predictors) for Prakriti evaluation found in Ayurvedic texts (grouping of items) was used to create N-PAS. Observations & Result: 100 neonates were taken and after applying frequency distribution in descriptive statistics and one-way ANOVA in inferential statistics to compare concerned parameters in the N-PAS with anthropometrical measurements, it was observed that Kapha-Vataj Prakriti was found to be more dominant with 27% because in early childhood, kapha dosha is found to be the paramount one. Discussion and Conclusion: This study has a vital role in determining the role of Prakriti in promotion of health, agni status of individual, determination of bala/ strength, diagnosis, management of diseases, prognosis, decision of preventive medicine, determination of genomic relevance with doshik predominance. Effectiveness of Nimbasava Nasya with Sigru Beeja in Acute Exacerbation of Chronic Rhino Sinusitis - A Comparative Clinical Trial
Chronic Rhinosinusitis is defined as a group of disorders characterized by inflammation of the mucosa of the nose and paranasal sinuses lasting for more than 12 weeks. Acute exacerbation of Chronic Rhinosinusitis is defined as a sudden worsening of symptoms in a patient who has chronic sinusitis. According to the science of Ayurveda, Dushta Pratisyaya condition, is in proximity to chronic sinusitis. Asava (fermented formulation) is mentioned for Virechana Nasya (type of Errhine therapy). Nimbasava is a formulation of Asava Kalpana prepared with Guggulu Tikthaka Gritha Yoga explained in Vaatavyadhi Chikitsa. Sigrubeeja (seed of Moringa Oleifera) and Saindhava (rock salt) are explained among Shirovirechana Gana (a group of medicines that can be used for nasal administration). The study was conducted on 30 patients of chronic sinusitis who were randomly divided into two groups and were given treatment for 7 days and follow-up after for 3 months. Out of two groups, 15 patients were given Virechana Nasya with Nimbasava, Sigru Beeja, and Saindhava for 7 days. The other 15 patients were treated as a standard control group and given the modern conventional management with Amoxicillin Clavulanate 500 mg, Fluticasone nasal spray, and Otrivin nasal drops for 7 days. The statistical analysis of data was done with the Wilcoxon Signed Rank Test and Mann-Whitney U test. During this trial, it was observed that Nimbasava Nasya with Sigrubeeja is effective in acute exacerbation of Chronic Rhinosinusitis. Pain relieving action of this Nasya was statistically highly significant. It was also found that the effectiveness of Nasya is more in reducing the recurrence of acute exacerbations of Chronic Rhinosinusitis
Cross-sectional study for profiling health status, health-related demographics, and health-seeking behaviour of the Scheduled Caste Population in selected areas of Chennai and Chengalpet Districts of Tamil Nadu
Background: Scheduled Caste (SC) population forms a large section of the Indian population who are often marginalised and deprived of social, economic and health benefits due to ignorance and socio-cultural milieu. Though various studies have been conducted on the socio-economic and political conditions of the SC population in Tamil Nadu, limited data is available on the status and determinants of their health. Objectives: The study\u27s objectives were to estimate the health status of the SC population, health-related socio-demographic conditions and the population\u27s health-seeking behaviour. Method: The observational, cross-sectional study was conducted in five SC-predominant areas of Chennai and Chengalpet districts of Tamil Nadu. Their health status, socio- demographic conditions, including household conditions, and health-seeking behaviour were estimated. Result: A total of 1452 households and 6072 participants of the SC community were studied, mostly belonging to the Adidravida community. Most households were of Upper-lower socioeconomic class with basic household amenities. A total of 11.54% of the adult population had a history of diagnosed chronic illnesses. Nearly one-fifth of reproductive-age women had menstrual irregularities. Non-communicable and infectious diseases were the most frequent reason for health care centre visits. Conclusion: Extensive and continuous screening of health status and chronic diseases is essential for early diagnosis and intervention. Focused interventions on improving socio- demographic status can further strengthen the health condition of the SC community. Integrating all systems of medicine in public health can help cater to the health requirements of all sections of the community. Concerted efforts can cut down the burden of chronic diseases
Microscopic Characterization and Analysis of Ayurvedic Herbal Products Using Light Microscopy
Objective: Today sophisticated modern research tools for assessment of the plant drugs are achievable but microscopic method is still one of the ingenious and procurable methods to start for substantiate the correct recognition of the source material. Powder microscopy helps to find out the impurities and also helps in quality assessment of the drug. To standardize and evaluate the readymade as well as homemade herbal powder and thus provides means for assessing the authenticity and morality of herbal drugs. Materials and Methods: Microscopic detailed examination of herbal plant parts such as G. glabra, P. emblica, P. nigrum, P. longum was established with different reagents such as acetic acid, iodine, sulphuric acid and hydrochloric acid followed by observation of slide under LEICA Software Capture and Display Software and photographs of each slide was captured for evaluation of the drug. Observation and Results: Microscopic assessment of the homemade and readymade herbal parts of G.glabra and P.emblica shows original cellular structures while unidentified cellular structure were observed in readymade powder of P. longum which perhaps the growth of fungal mycelium and leaf part were observed in P. longum, thus Microscopy method permits more detailed examination of a drug and it can be used to identify the organized drugs by their known histological characters. Conclusion: Powder microscopic evaluation of herbal powder is one of the simplest and authenticated methods for the proper identification of the drug. It helps in purity assessment of the readymade herbal powder. Microscopic study and physiochemical standards can be useful to substantiate and authenticate the drug
Physicochemical and Phytochemicals investigation of Dooshivishari Agada by HPLC
Background: Dooshivishari Agad (DA) is an herbal formulation that has been documented in Indian Traditional Medicine as a remedy for latent poisoning. Objective: this research was conducted to study Physicochemical parameter & Phytochemical analysis of DA by HPLC. Methods: It includes authentication of raw drugs and preparation of DA. Physicochemical parameter was tested by Loss on drying ,Acid insoluble Ash, Water soluble ash, Total ash value, Water & Alcohol soluble Extractive Value. HPLC was used for phytochemical screening. Observation: In analytical study obtained values are Loss on drying 2.54%,Water soluble Ash 1.84%, Acid insoluble Ash 0.5% , Total Ash value 5.20% Water Soluble Extractive Value 16.89%, Acid Soluble Extractive Value 23.94%, pH 5.6.Preliminary data of phytochemical screening revealed that it is rich in phenol compound and terpenoids Conclusion: This study provide the data for standardisation & phytochemical characterisation of DA for its pharmacological action.
