    A Polyherbal extract treats Streptozotocin induced Diabetes mellitus: Pharmacognostic study, HPLC analysis and in vivo antidiabetic activity

    The plants used in study have been used traditionally for various purposes skin problems, diabetes, astringent, anti-inflammatory, antioxidant, hepatoprotective activities, anthelmintic, antipyretic, antidysentric, anti-hyperlipidemic, anticancer properties. The present study involves the evaluation of pharmacognostic parameters of the four herbal drugs (Azadirachta indica leaves, Pterocarpus marsupium heartwood, Picrorhiza kurroa rhizomes, and Withania coagulans berries and fruit coat), High performance Liquid chromatography (HPLC) as well as antidiabetic activity of polyherbal extract. The qualitative as well as quantitative analysis was done. The drug was given to Streptozotocin induced diabetic rats in doses 250 mg/kg and 500 mg/kg body weight for 21 days. In HPLC analysis it was found that major component in all the extracts was beta carotene along with hesperidin, tannic acid, catechin, ferulic acid, caffeic acid along with other polyphenolic components responsible for the antioxidant activity as well as antidiabetic activity of polyherbal extract. Rats were found to show an increase in body weight, improved blood sugar levels and hemoglobin. The liver glycogen, total proteins, serum lipid profile were restored back to normal range after using the polyherbal extract in diabetic rats. The histopathological study also shows an improvement in diabetic rats at cellular level. The results were found to be dose dependent

    Comparative antioxidant activity of Sesame root and leaf extract - An In-vitro study

    Universe has millions and millions of plants, but not all the plants have medicinal values.  One such gift from nature is Sesame and is crowned as the “Queen of oilseed crops” due to its various medical properties like anti-inflammatory, antioxidant, anti- microbial etc. This research paper aims to compare the antioxidant activity of sesame leaf extract and sesame root extract with the standard ascorbic acid using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydrogen peroxide (H2O2) assay. Different concentrations of the extracts and ascorbic acid were prepared (10 µg/mL, 20 µg/mL, 30 µg/mL, 40 µg/mL, and 50 µg/mL), and the antioxidant activity was measured. The results indicate that both sesame leaf extract and sesame root extract exhibit antioxidant activity, with sesame leaf extract showing slightly higher activity than sesame root extract. The findings suggest that sesame leaf extract has potential as a natural antioxidant source and could be used as an alternative to synthetic antioxidants. Overall, this study provides valuable information on the potential health benefits of sesame extracts and highlights the importance of natural antioxidants in preventing free radical damage and associated diseases

    Protective potential of Quercus leucotrichophora A. Camus seeds in chemically induced urolithiasis in rats

    Background: Quercus Leucotrichophora A. Camus (QL) seeds are used in traditional medicine to treat various urinary and renal diseases. It is used as a diuretic, antioxidant, and nephroprotective agent. In the present study, we investigated the in vitro and in vivo antiurolithiatic activity of methanolic extract of Quercus Leucotrichophora A. Camus (MEQL) Seeds. Methods: An in vitro assay was used to determine whether MEQL prevents the formation of calcium oxalate (CaOx) or promotes CaOx dissolution. In in vivo studies, the effects of oral administration of Methanolic extract of test drug were studied on calcium oxalate urolithiasis in male Wistar rats. Male Wistar rats were divided into six groups of 6 animals each. Animals of Normal control received regular drinking water ad libitum throughout the study. whereas in other groups nephrolithiasis was induced by providing drinking water containing 0.75% ethylene glycol and 1% ammonium chloride for 7 days. Test groups were treated with 250 mg/kg, 375 mg/kg, and 500 mg/kg of the test drug and standard control with Cystone (750 mg/kg) for 21 days. Urine was examined for the presence of crystals on the 8th and 22nd days. On the 22nd day, 24 h urine, serum was collected and various biochemical parameters were estimated in urine, serum, and kidney homogenate along with histology of the kidney. Results: In the in vitro experiments, MEQL exhibited a concentration-dependent inhibitory activity on nucleation and aggregation. The urine volume increased significantly in all test groups. Test groups showed a significant reduction in the number of CaOx crystals in urine. Levels of urinary calcium, phosphorus, and sodium; Serum levels of calcium, phosphorus, creatinine, and urea decreased significantly in standard and test groups. Histopathology shows significant changes. From the above results, it can be concluded that the methanolic extract of Quercus leucotrichophora seeds has a significant inhibitory effect on calcium oxalate urolithiasis due to their diuretic and antioxidant activity. Conclusion: These results indicate that MEQL showed significant activity in urolithiasis which might be due to diuretic and antioxidant action

    Cardioprotective activity of Dashanga Agada in Doxorubicin induced Cardiotoxicity in Mice Cardiomyoblasts (H9c2)

    Background: Cancer is still a complicated and difficult condition with serious consequences for the general population. Currently, it poses a major risk to the global health. There are numerous risk factors associated with cancer. They include tobacco, obesity, excessive consumption of packed foods, lack of physical activity, alcohol consumption, etc. Continued prevention, research & early detection are required to improve outcomes for cancer patients. Anticancer medications have been associated with cardiotoxicity, compromising individuals\u27 heart function. Early detection of cardiotoxicity is essential for avoiding irreparable heart damage. Cardio-oncology has arisen as a distinct area of study and therapeutic practice in recent years. Several aspects of drug-induced cardiotoxicity remain unknown. Such toxicities are considered as garavisha in Ayurveda. They can be treated by using Agada principles. Insect bites can lead to cardiac symptoms, according to both ancient and modern science. Acharya Vagbhata has mentioned hrudpeeda as one of the lakshana of Vataja Vrischika damsha. Dashanga Agada (DA) is the formulation mentioned in context of Keeta visha. As it is the formulation of choice in insect bites exhibiting cardiac symptoms, it is studied in present research to reduce the cardiotoxicity induced by doxorubicin (Dox). The study was carried out on H9C2 mice cardiomyoblasts. Materials and Methods: Hydroalcoholic extract of Ayurvedic formulation Dashanga agada was prepared. H9C2 mice cardiomyoblasts were used to study trial drug Dashanga Agada on cardiotoxicity induced by a chemotherapy drug named Doxorubicin. MTT assay, DAPI nuclear staining, DCFH-DA assay for intracellular ROS generation & SOD assay were the four parameters studied on H9C2 cell line. 4 groups were taken for the study. Dashanga agada was studied at different concentrations & different time intervals on the above mentioned parameters. Results: IC50 values of DA & Dox for 24hrs were calculated to be 357.15 & 5.48 µg/ml respectively. Dashanga agada was effective in increasing cell viability, improving the nuclear morphology & antioxidant activity when compared to Doxorubicin treated group. Conclusion: Dashanga Agada has cardioprotective effect. Within parallel & pre-treatment groups, parallel treatment gave better results

    Experimental study of Pruthvisara taila in excised wound model in Wistar albino rats

     Pruthvisara taila is a formulation indicated in the chikitsa of vrana as topical medicine in classical text Chakradatta. It contains Shuddha chitrakamoola, Shuddha vatsanabha, Shuddha karaveera, Nirgundi moola, Nadibeeja, Kanji and Karanja taila as the base. Aims and Objectives: Evaluation of wound healing property of Pruthvisara taila through experimental study. Methodology: Wound healing property in albino rats by excision wound healing model and its histopathology study. Results: Results of the study on the parameters assessed like percentage of wound contraction and histopathology study; percentage of wound closure was observed in Control group was 94.92%, in standard it was 93.73% and in test drug the percentage of wound contraction was 95.73%.The test drug, Pruthvisara taila showed more angiogenesis and formation of new blood vessels than standard group and in control group there was absence of formation of new blood vessels and proliferation of fibroblast cells. The control group did not show any collagen formation and scab formation the test drug Pruthvisara taila showed moderate formation and deposition compared to standard drug. Compared to control and standard group, the test drug, Pruthvisara taila selected in this study showed remarkable wound healing property in excised wound

    Physiochemical and phytochemical analysis of Karanthai legium - Siddha herbomineral formulation

    Siddha system of medicine is one of the holistic systems. The consumption of traditional medicine is also in upward trend. To manufacture any herbal drug the quality and safety of the drug is to be estimated. Karanthai legium (KL) is one of the herbomineral formulation indicated for kiranthi (Skin disease), karappan (Eczema), soolai (Pain), yoni putru (Cervical cancer), kunmam (Ulcer). This study aims to develop standardization of Karanthai legium as per PLIM (Pharmacopeia Laboratory of Indian Medicine) guidelines. The test drug KL focused on Physico-chemical analysis, Phytochemical Analysis, Oraganoleptic character, Instrumental analysis such as heavy metal analysis, HPTLC Analysis, Aflatoxin, Pesticide residue, Specific pathogen and Microbial contamination. Physiochemical analysis report showed loss of drying KL is 14.96%, total ash is 0.92%, acid insoluble ash is 0.17%, water soluble ash is 0.50%,pH of KL is 5.45.The drug KL had HPTLC fingerprint analysis revealed the presence of nine phytocomponents at 254nm, eight phytocomponents at 366nm and six phytocomponents at 520nm.The heavy metal analysis showed no trace of heavy metal residue in KL. This study revealed the data regarding the physicochemical characterisation of Karanthai legium, which will be helpful in standardising the drugs and for further comparison studies

    The GCMS, antioxidant, and anti-inflammatory activity of the Ayurveda oil, Arukaladi tailam

    Ayurvedic tailams have a long history of use and efficacy. They have been used for centuries, with many individuals reporting positive results in managing and improving their health. Ayurvedic tailams typically have fewer side effects compared to synthetic medications, making them a preferred choice for individuals seeking natural remedies and  can be applied both externally (for massage) and, in some cases, internally (for specific therapies). This versatility allows for a wide range of applications and health benefits.The current study focuses on the GC MS analysis of Arukaladi tailam, an oil composition used in Ayurveda to treat jaundice when administered topically. Before being subjected to the GC MS analysis, the product was appropriately prepared after being purchased from a reputable Ayurvedic dealer in Chennai, India. The IC50 value of the DPPH assay was found to be 744.7391μl/ml and FRAP reveals that the tailam had good reducing power. Further the anti-inflammatory of the tailam exhibited 14.2% of haemolysis at 1000μl/ml and inhibited protein denaturation by 63.74% with an IC50 value of 789.4013 µl/ml. The presence of biomolecules as shown in the GC MS profile are 2-Fluoro-6-trifluoromethylbenzoic acid, 2- tetradecyl ester, Tridecanol, 2-ethyl-2-methyl-, etc. These chemicals could help the medication work better to treat jaundice

    Management of COVID-19 patients of CT Severity 15/25 and above with Ayurveda drugs as add-on therapy – A Case Series

    Ayurveda, which is the oldest life science in the world, has yet to be fully accepted as the only or even an additional therapy for severe diseases. It is a common perception that Ayurveda has a significant role in the preventive aspect of diseases, and the drugs of Ayurveda are more effective in chronic ailments. Most publications are also limited to these areas only, as use of Ayurveda in acute or severe cases is not yet justified scientifically because of the lack of significant scientific clinical data as very few case reports are published about the use of Ayurveda in acute cases. Here we present a case series of treatment of five COVID-19 patients with CT Severity 15/25 and above with Ayurveda drugs as add-on therapy admitted to tertiary care, a Level-3 COVID-19 treatment hospital. These five patients sought Ayurveda opinion and consented to Ayurveda treatment along with the ongoing treatment regime. All the patients showed improvement with Ayurveda treatment as an add-on therapy, and none of the patients needed ICU, and all patients were discharged when they started maintaining oxygen saturation at room air. This report addresses the clinical effectiveness of Ayurveda drugs as add-on therapy in severe cases of COVID -19. It also attempts to invite the attention of researchers towards the use of Ayurveda concepts and drugs in severe or acute cases of diseases.

    Inefficiency of medical therapies used in order to cure patients with microbial infestations requires safe, effective new antimicrobial agents based on variety of parameters. Ayurveda being a holistic science uses many such safe and effective herbal and herbo-mineral interventions in the management of microbial infestations. Nimba (Azadirachta indica A. Juss) is one commonly available and well known medicinal plant used as krimihara dravya (antimicrobial drug). It has tikta, rasa (bitter taste), Laghu guna (light in property) sheeta veerya (cold in potency), katu vipaka (pungent of metabolism) act as krimighna (Antimicrobial), kusthaghna (alleviates Skin disease), vranaropana (wound healing), raktashodhaka (blood purifier). Also it acts as antioxidant, inhibits bacterial growth. Aim and objectives: To evaluate the physico- phyto-constituents and to assess anti-microbial effect of aqueous extract of Nimba patra. Materials and methods:  The official part of the trial drug was subjected for exhaustive extraction by using distilled water and analyzed for physico-chemical, phyto-chemical analysis and antimicrobial evaluation by using  aqueous extract on Staphylococcus aureus (SA), Pseudomonas aeruginosa (PA), Proteus mirabilis (PM), E. coli (EC) and Candida albicans (CA) with Agar well diffusion method to asses Minimum Inhibitory concentration, Minimal Bactericidal Concentration with a dose of Stock 10g/10ml. Observation and results: Physico-chemical study shows 4.63%w/w moisture content,  9.05%w/w Ash content, 5.16 pH value, phyto-chemical analysis indicates the presence of tannins, flavonoids, alkaloids, and anti- microbial study of aqueous extract of nimbi patra shows significant effect on bacteria and fungus in comparison with known standard drug

    Mechanism of Arkeshwara rasa to combat hepatocellular carcinoma

    Background: Immunomodulatory treatments along with systemic and loco-regional methodology of treatment approach have reduced the incidence of Hepatocellular carcinoma (HCC) in recent observations. The phytomolecular applications have been seen to reverse the hepatic fibrosis aiding in regeneration of hepatocytes. The cytotoxicity of Arkeshwara rasa (AR) was investigated on HCC cell lines to observe the general signs of apoptosis. Methodology: The MTT (3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide) assay and Ethidium Bromide/Acridine orange Assay (EtBr/AO) were subjected on AR to know the cytotoxicity and the apoptotic results. Results: The IC50 of 37.14 μg/ml was exhibited by AR against Huh 7 cell lines and the (EtBr/ AO) Assay demonstrated the early and late apoptotic features in comparison with Untreated and Doxorubicin standard drug. Conclusion: AR on the basis of present evaluation against HCC cell lines and previous results on various other cell lines might be able to check the pathogenesis progression of HCC cells initiating the apoptotic features or reviving the vulnerable cell from further damage because of the phyto-constituents or mineral inclusions. These preliminary observations have to be again revalidated with reverse transcription, cDNA synthesis, western blotting, cell cycle analysis, etc
