An H₂O₂ Generation Pathway for Riboflavin-sensitized Oxidation of Ascorbic Acid
光励起リボフラビン(Rib)存在下でのアスコルビン酸(Asc)の光酸化におけるH₂O₂生成機構について,Asc,H₂O₂,O₂間の化学量論関係をもちいて検討した。1.25 μM または12.5 μM Rib の存在下,Ascの光酸化によるO₂吸収速度は光強度に比例した。O₂吸収量に対するH₂O₂生成量のモル比は,光強度の増大とともに増加し,高光強度で約0.5の一定値になった。ローズベンガル(RB)を光増感剤とした場合は,0.125 μM Rb 存在下では,H₂O₂/O₂比は光強度に依存せず約0.5であった。1.25 μM RBでは,Rib の場合と同様の傾向を示し,比は最終的に約0.6であった。Asc光酸化におけるO₂⁻が関与するType Iおよび¹O₂ が関与するType II反応の寄与の割合を検討するために,SOD や¹O₂ 消光剤の影響を調べた結果,Type II反応と伴にType I反応も関与していることが示された。空気開放系でAscを完全に酸化させた場合,Ascと生じたH₂O₂ の化学量論関係は1: 2であった。以上の結果から得られたH₂O₂/O₂比とH₂O₂/Asc比を満足させる反応機構を考察した。An H₂O₂ generation pathway for ascorbic acid (Asc) oxidation in the presence of sensitized riboflavin (Rib) was investigated by analyzing the stoichiometric relation between H₂O₂, Asc and O₂. When 1.25 or 12.5 μM Rib was present, the O₂ uptake rate of Asc photooxidation increased linearly with increasing light intensity. The molar ratio of the amount of O₂ uptake to the amount of H₂O₂ produced increased with increasing light intensity, and then became a constant value of approx. 0.5. With 0.125 μM rose bengal (RB) instead of Rib, the ratio of H₂O₂/ O₂ did not depend on light intensity, and was about 0.5. When 1.25 μM RB was used, the ratio showed similar patterns to those of Rib, and a constant value of approx. 0.6 was obtained.
To determine the contribution of Type I and Type II reactions in Asc photooxidation, the effect of SOD and ¹O₂ quencher on the O₂ uptake rate was investigated. The results showed the occurrence of not only Type II but also Type I reactions. When Asc was oxidized completely in open air, the stoichiometry of Asc to H₂O₂ was 1:2. Based on these results, we discussed a pathway of Asc photooxidation to accommodate the obtained values of H₂O₂/O₂ and H₂O₂/Asc.departmental bulletin pape

Practices and Evaluation of Hands-On Disaster Prevention Education in Elementary Schools : Through the Practices of Classes Cooperating with Civil Engineering
自然災害が多い日本では、学校教育において、防災教育に取り組む必要性が高まっている。本稿は、小学校6年生を対象に、体験型防災教育の授業を実践し、その有効性を評価したものである。授業実践では、土木工学の専門家と連携し、「地震発生の仕組み」「液状化実験」「共振実験」「火山噴火の仕組み」を取り上げた。授業後、学習者が授業に取り組んだ態度やワークシートへの記述等から実践を評価した。その結果、本実践が、学習者の防災意識を高めることに有効であることを検証することができた。departmental bulletin pape
Postmodernity of Mobility between the Basque Homeland and the Diaspora
The Basque people has migrated from its homeland to the New World since the 16th century. Especially from the 19th century, they experienced massive migration to the end of the 1970s. At the destinations, they maintained their Basque identity and formed diaspora. They also moved frequently between the diaspora and the homeland. As a result, they continued to keep close relationship beyond the borders of geographically separated spaces, the Basque homeland and the diaspora. The migrants' network within those spaces contributed to unify them as a transnational social space. Despite drastic diminishment of human migration in the end of the 1970s, the mobility network continues functioning, while altering its character. in order to fulfill a requirement both of the Basque society and of the global citizens in the postmodern era.The Basque community, not only in the homeland but also in the diaspora, tries to enhance its open nature and to incorporate newly defined members into their diaspora, as discussed in this article on the cases of mobility of the Basque government subvention and of academic elites.論説Articlesdepartmental bulletin pape
Wasabi increases insulin sensitivity ant attenuates glomerular hyperfiltration and proteinuria in insulin-resistant diabetes of fructose-fed Wistar rats
過剰なフルクトース負荷は2型糖尿病を発症させることが報告されている。本研究では、わさびの水抽出エキスがフルクトース負荷ラットでみられるインスリン抵抗性を軽減するか否かについて検討した。Wistarラットを、1)15%フルクトース溶液群、 2)15%のフルクトース溶液及び10%わさぴ水抽出物群、または 3)水投与群の3群に分け、6週間経過観察した。実験最終に24時間尿を採取し、麻酔下に血液及び臓器を摘出し以後の実験に用いた。3群間で体重及ぴ収縮期血圧に変化はみられなかったが、フルクトース負荷群では大動脈壁の重量が増加し、これはわさび投与群で有意に軽減した。フルクトース負荷では、血糖及び血漿インスリン濃度が有意に増加し、インスリン抵抗性の指標であるHOMA-R値は水投与群に比して有意に高値であった。血糖及びインスリン濃度の上昇はわさび群では有意に軽減し、水投与群との間に差がみられなかった。フルクトース負荷によるインスリン感受性の低下は、糸球体濾過量の増加を伴っており、尿量、尿中ナトリウム排泄量及び尿中タンパク排泄量が有意に高値を示した。わさび抽出物の投与により、尿量、尿中ナトリウム排泄量及びタンパク排泄量は低下し、水投与群との間で差はみられなかった。高速液体クロマトグラフィー分析により、わさび抽出物には強力な酸素ラジカル・スカベン ジャーである6 メチルスルフィニル ヘキシル イソチオ シアン酸 (6MSITC) が含有されることが示されたが、わさび抽出物中の6MSITC量はインスリン感受性の改善に必要とされる量より低いことが明らかとなったことから、わさび抽出物中のインスリン抵抗性の改善には、6MSITC以外の物質の関与も示唆された。Fructose loading results in the development of type-2 diabetes. In the present study we examined whether Wasabi extract attenuates insulin resistance in fructose-loaded Wistar rats.The rats were randomly assigned to one of the following groups: 1) rats given 15% fructose solution, 2) rats given 15% fructose and 10% Wasabi extracts, and 3) rats given water. They were maintained for 6 weeks. There were no differences in body weight or systolic blood pressure among the three experimental groups. Weights of aortic walls increased with fructose loading and this was abolished with Wasabi extracts treatments. Fructose loading significantly increased plasma insulin concentrations in association with increased plasma glucose levels, thereby increasing levels of HOMA-R, an indicator of insulin resistance. These alterations were reversed with Wasabi extract to normal levels as in control rats. Decreased insulin sensitivity was associated with glomerular hyperfiltration and increases in urine volume, and urinary sodium and protein excretion. These altered urinary parameters in fructose loading were significantly attenuated with treatment with Wasabi extracts. Wasabi extracts contain a potent oxygen radical scavenger, 6-methylsulfinyl hexyl isothiocyanate (6-MSITC) as assessed by C₁₈ reversed phase HPLC; however, the amount of 6-MSITC present was much lower than the amount required to restore insulin sensitivity. These results suggest that Wasabi extracts attenuate insulin resistance induced by fructose loading, and this is associated with decreased glomerular hyperfiltration and urinary protein excretion. These benefits may be attributed to substances other than 6-MSITC in Wasabi extracts.departmental bulletin pape
