WRN modulates translation by influencing nuclear mRNA export in HeLa cancer cells
Background The Werner syndrome protein (WRN) belongs to the RecQ family of helicases and its loss of function results in the premature aging disease Werner syndrome (WS). We previously demonstrated that an early cellular change induced by WRN depletion is a posttranscriptional decrease in the levels of enzymes involved in metabolic pathways that control macromolecular synthesis and protect from oxidative stress. This metabolic shift is tolerated by normal cells but causes mitochondria dysfunction and acute oxidative stress in rapidly growing cancer cells, thereby suppressing their proliferation. Results To identify the mechanism underlying this metabolic shift, we examined global protein synthesis and mRNA nucleocytoplasmic distribution after WRN knockdown. We determined that WRN depletion in HeLa cells attenuates global protein synthesis without affecting the level of key components of the mRNA export machinery. We further observed that WRN depletion affects the nuclear export of mRNAs and demonstrated that WRN interacts with mRNA and the Nuclear RNA Export Factor 1 (NXF1). Conclusions Our findings suggest that WRN influences the export of mRNAs from the nucleus through its interaction with the NXF1 export receptor thereby affecting cellular proteostasis. In summary, we identified a new partner and a novel function of WRN, which is especially important for the proliferation of cancer cells.Consejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
Antioxidant effect and chemical composition of Ananas comosus [L.] Merr. peels from Peruvian Northern
Pineapple peels has several beneficial properties including antioxidant activity. We investigated the antioxidant effect of five different peels of pineapple lyophilized extracts, not adsorbed and adsorbed onto Amberlite. They were examined using total phenolic contents (TPC), antioxidant effect by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging and ferric reducing antioxidant power (FRAP). In addition, we analyzed the chemical composition by HPLC-ESI-QTOF-MS/MS. The main constituents of pineapple peels were tentatively identified as quercetin glycosides and N,N’-diferuloylspermidine. We conclude that the antioxidant activity in pineapple peels from District of Poroto, Province of Trujillo, Region of La Libertad, can be associated with the presence of flavonoid and spermidines. © 2019, Universidad de Santiago de Chile. Ethanol pre-treatment to ultrasound-assisted convective drying of apple
For the first time, the ethanol as pre-treatment to the ultrasound-assisted convective drying of food was evaluated. Pre-treatments were performed by immersion of apple slices in ethanol (0-30 min). Pre-treated samples were convectively dried (50 degrees C, 1 m s(-1)), without/with ultrasound (21.77 kHz, 20.5 kW/m(3)). As results, if both technologies were considered, conventional drying time reduction reached 70 +/- 2%. From drying kinetics modelling, it was identified that ethanol pre-treatments mainly reduced the external resistance to mass transfer, while ultrasound had a greater influence on the internal one. In dried samples, as the ethanol pre-treatment time increased, the shrinkage decreased, and their rehydration capacity was greater. After rehydration, samples showed a decrease of> 85% in viscoelastic characteristics. The antioxidant capacity and total phenolic content were better retained with ultrasound application. The obtained results corroborate that the proposed technologies are complementary significantly accelerating the drying without negative effects on physical properties.Fondo Nacional de Desarrollo Científico y Tecnológico - Fondecy
Metagenomic data on the composition of bacterial communities in lake environment sediments for fish farming by next generation Illumina sequencing
This article contains data on the bacterial communities of lagoon sediments with fish potential in the Central Andes of Peru. The surface sediment samples were collected from four lagoons destined for continental water fish farming. DNA extraction was performed from 0.5 g of sample through the Presto(TM) Soil DNA Extraction Kit. Bacterial sequencing of the 16S rRNA amplicon was performed on the DNA extracted from the sediment. At least 36 Phyla bacteria were detected, the bacterial communities being dominated by Proteobacteria, Cyanobacteria, Actinobacteria, Firmicutes, Chloroflexi. These data can be used for predictive analysis to gain a better understanding of the dynamics of bacterial communities in environments under pressure from fish farming. (C) 2020 The Author(s). A distance between bounded linear operators
We extend the classical Banach-Mazur distance [3] from Banach spaces to linear operators between these spaces. We prove in the finite dimensional case that the corresponding topology is metrizable, complete, separable and locally compact. Furthermore, we prove that the Banach-Mazur compactum embeds isometrically into the resulting topological space. (C) 2020 Elsevier B.V. Scientific and technical studies on eco-efficient binary cements produced with thermally activated ichu grass: Behaviour and properties
The present research study analyses, for the first time, the viability of using thermally activated ichu grass as a supplementary cementitious material for the design of future eco-cements. To this end, the ichu ash resulting from the activation was characterised and its pozzolanic activity, the evolution of its mineralogical phases during the pozzolanic reaction and the physical-mechanical behaviour of the blended cementitious matrices were analysed. The results show that ichu ash exhibited high pozzolanic activity in a pure pozzolan/lime system as a consequence of its high reactive silica content. Conversely, its use in a pozzolan/cement system gave rise to negative effects due to the presence of important alkali contents in the ash (9.6% Na2O eq). The blended cements presented greater water demand, reduced capillary absorption, increased resistivity and lower mechanical properties (10-14%) as well as reduced average pore size than the reference mortar. Severe pulmonary radiological manifestations are associated with a distinct biochemical profile in blood of tuberculosis patients with dysglycemia
Background Diabetes mellitus (DM) is thought to affect tuberculosis (TB) clinical presentation and treatment response. Whether DM impacts radiological manifestations of pulmonary TB is still not clear. This study investigated the impact of glycemic status on radiological manifestations of pulmonary TB cases and its relationship with concentration of biochemical parameters in peripheral blood. Methods A retrospective cross-sectional study used data from 132 microbiologically confirmed pulmonary TB patients from Lima, Peru, evaluated in a previous investigation performed between February and December 2017. Chest radiographs were analyzed by a radiologist and a pulmonologist. Radiographic lesions were identified as cavities, alveolar infiltrates and fibrous tracts. Hyperglycemia in TB patients was identified by use of fasting plasma glucose, HbA1c and oral glucose tolerance test. Clinical, biochemical and hematological parameters were also analyzed. Results TB patients with hyperglycemia presented more frequently with cavities, alveolar infiltrates and fibrous tracts than those with normoglycemia. Hierarchical clustering analysis indicated that patients with more diverse and higher number of lung lesions exhibited a distinct laboratorial profile characterized by heightened white blood cell counts and circulating levels of total cholesterol, triglycerides and transaminases and simultaneously low levels of albumin and hemoglobin. Multivariable regression analyses adjusted for age, sex, prior TB, hemoglobin levels and acid-fast bacilli >= 2+ in sputum smears, demonstrated that presence of prediabetes or diabetes in TB patients was associated with increased odds of having 3 pulmonary lesion types (p = 0.003 and p = 4 lesions (p = 0.001 and p = 0.01 respectively). Conclusion Hyperglycemia (both DM and prediabetes) significantly affected the presentation of radiographic manifestations and the number of lesions in pulmonary TB patients as well as the biochemical profile in peripheral blood.Consejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
Ce alpha Mn1-alpha O2 CATALYSTS SUPPORTED OVER gamma-Al2O3 PREPARED BY MODIFIED REDOX-COPRECIPITATION METHODS FOR n-HEXANE COMBUSTION
A series of Ce alpha Mn1-alpha O2 catalysts supported on gamma-alumina with various molar concentrations of Ce (alpha, from 0 to 0.90) was synthesized by coprecipitation, applying two different precipitating agents, namely, sodium hydroxide (method 1) and sodium carbonate (method 2), with the use of sodium permanganate as a redox agent for precipitation. XRD profiles of the supported samples revealed the predominant abundance of a typical fluorite crystalline structure. TPR thermograms of supported samples were displaced towards lower temperatures with increasing Mn concentration, in contrast with the bulk samples. The supported Ce-Mn samples exhibited a greater performance in n-hexane elimination than did the corresponding simple oxides. The sample Ce0.33M O-0.67(2) obtained by method 2 presented the best activity, probably due to the enrichment of Ce4+, Mn3+ and Mn4+ surface species, an excess of superficial oxygen species and an easy reducibility as well as the lowest apparent activation energy.Consejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
Characterization of Plukenetia volubilis L. fatty acid-based alkyd resins
Fatty acid-based alkyd resins prepared with different amounts of glycerol and pentaerythritol were characterized. Sacha inchi oil and linseed oil (comparative purposes) were used as fatty acids' sources. FT-IR and H-1 NMR spectroscopy were done for alkyd structural verification. Alkyd resins were evaluated through physico-chemical (colour, density, viscosity) and thermal characterization. Film coating performance (drying, hardness, chemical resistance) was also studied. The oxidative crosslinking time tendency was corroborated by the quartz crystal microbalance (QCM) technique. Alkyd resins obtained with fatty acids from sacha inchi and linseed oils had similar properties. Results indicated that lighter resins can be obtained from sacha inchi oil, whereas pentaerythritol increases viscosity and thermal stability, and retards drying time of fatty-acid based alkyd resins.Consejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
