Spatial and temporal genetic diversity and population structure of hemileia vastatrix from peruvian coffee plantations
This research was supported by project N? 177-2015 FONDECYT (The National Fund for Scientific and Technological Development) and Universidad Nacional Agraria La Molina, Peru.Population genetic studies of Hemileia vastatrix have been conducted in order to describe the evolutionary dynamics of the pathogen and the disease epidemiology as consequence of changes in disease management and host distribution occurred in Peru after the 2013 epidemic. These analyses were performed by sequencing the internal transcribed spacers of the nuclear ribosomal DNA (rDNA-ITS) of H. vastatrix collected from two coffee growing areas in 2014 and 2018. H. vastatrix population showed high haplotype diversity (Hd = 0.9373 ± 0.0115) with a low nucleotide diversity (π = 0.00322 ± 0.00018). Likewise, AMOVA indicated that fungus population has behaved as a large population without structuring by geographical origin and sampling years (FST = 0.00180, P = 0.20053 and FST = 0.00241, P = 0.19693, respectively). Additionally, the haplotype network based on intraspecific phylogenetic analysis of H. vastatrix using Peruvian and NCBI sequences revealed that Peruvian ancestral haplotypes, which were maintained in time and space, would correspond to the reported sequences of the races II and XXII. This re-sult suggests that no substantial changes have occurred through time in Peruvian Hemileia vastatrix population. © 2021, Korean Society of Plant Pathology. Annual to decadal temperature adaptation of the soil bacterial community after translocation across an elevation gradient in the Andes
The response of soil microbial activity to climate warming has been predicted to have a large destabilising effect on the carbon cycle. However, the nature of this feedback remains poorly understood, especially in tropical ecosystems and across annual to decadal timescales. We studied the response of bacterial community growth to 2 and 11 years of altered temperature regimes, by translocating soil across an elevation gradient in the tropical Andes. Soil cores were reciprocally translocated among five sites across 3 km in elevation, where mean annual temperature (MAT) ranged from 26.4 to 6.5°C. The bacterial community growth response to temperature was estimated using a temperature Sensitivity Index (SI): the log-ratio of growth determined by leucine incorporation at 35°C: 4°C. Bacterial communities from soil translocated to their original site (controls) had a growth response assumed to be ‘adapted’ to the original MAT. Translocating soil downslope (warming) resulted in an increased SI relative to their original growth response, and vice versa under cooling, indicating community-level adaptation over the incubation period to the altered MAT. The average level of adaptation (i.e., the extent to which SI converged on the control values) was 77% after 2 years, and was complete after 11 years. The adaptive response was faster when soil was warmed rather than cooled: instances of complete adaptation of SI occurred in soils after 2 years when warmed, but only after 11 years when they were cooled. Taken together, our results show that the majority of the growth adaptation to warming by the bacterial community occurs rapidly, within 2 years, whilst growth adaptation to cooling occurs within a decade. Our analysis demonstrates rapid warm-adaptation of bacterial community growth, with potential consequences for the temperature sensitivity of soil carbon cycling in response to future climate warming. © 2021 Elsevier LtdConsejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
Atovaquone/proguanil resistance in an imported malaria case in Chile
In November 2018, we diagnosed a cluster of falciparum malaria cases in three Chilean travelers returning from Nigeria. Two patients were treated with sequential intravenous artesunate plus oral atovaquone/proguanil (AP) and one with oral AP. The third patient, a 23-year-old man, presented with fever on day 29 after oral AP treatment and was diagnosed with recrudescent falciparum malaria. The patient was then treated with oral mefloquine, followed by clinical recovery and resolution of parasitemia. Analysis of day 0 and follow-up blood samples, collected on days 9, 29, 34, 64, and 83, revealed that parasitemia had initially decreased but then increased on day 29. Sequencing confirmed Tyr268Cys mutation in the cytochrome b gene, associated with atovaquone resistance, in isolates collected on days 29 and 34 and P. falciparum dihydrofolate reductase mutation Asn51Ile, associated with proguanil resistance in all successfully sequenced samples. Molecular characterization of imported malaria contributes to clinical management in non-endemic countries, helps ascertain the appropriateness of antimalarial treatment policies, and contributes to the reporting of drug resistance patterns from endemic regions. PISCO_HyM_GR2M: A model of monthly water balance in Peru (1981–2020)
Quantification of the surface water offer is crucial for its management. In Peru, the low spatial density of hydrometric stations makes this task challenging. This work aims to evaluate the hydrological performance of a monthly water balance model in Peru using precipitation and evapotranspiration data from the high-resolution meteorological PISCO dataset, which has been developed by the National Service of Meteorology and Hydrology of Peru (SENAMHI). A regional-ization approach based on Fourier Amplitude Sensitivity Testing (FAST) of the rainfall-runoff (RR) and runoff variability (RV) indices defined 14 calibration regions nationwide. Next, the GR2M model was used at a semi-distributed scale in 3594 sub-basins and river streams to simulate monthly discharges from January 1981 to March 2020. Model performance was evaluated using the Kling–Gupta efficiency (KGE), square root transferred Nash–Sutcliffe efficiency (NSEsqrt ), and water balance error (WBE) metrics. The results show a very well representation of monthly discharges for a large portion of Peruvian sub-basins (KGE ? 0.75, NSEsqrt ? 0.65, and ?0.29 < WBE < 0.23). Finally, this study introduces a product of continuous monthly discharge rates in Peru, named PISCO_HyM_GR2M, to understand surface water balance in data-scarce sub-basins. © 2021 by the authors. Metabolites, volatile compounds and in vitro functional properties during growth and commercial harvest of Peruvian lucuma (Pouteria lucuma)
The objective of this work was to identify primary and secondary metabolites, volatile compounds, bioactive properties and physicochemical characteristics of lucuma fruit at two ripeness stages, before physiological maturity (bPM) and at physiological maturity (PM) or commercial harvest. The content of reducing sugars decreased, while starch content increased. Myo-inositol and vitamin C content decreased and increased, respectively as maturity stage advanced. The content of stearic acid decreased possibly related to the formation of unsaturated fatty acids and volatile compounds, especially aldehydes, such as 2-hexenal. In fact, key identified volatile compounds included 2-hexenal and hexanoic acid. Regarding secondary metabolites and functional properties, these significantly decreased as maturity stage advanced, only the total carotenoid content significantly increased (0.083–0.148 mg ?-carotene equivalent); and the total phenolic content (83.3 69.3 mg GAE/g dw), sterols (4.4–6.5 ?g ?-sitosterol/g dw, 0.74–0.86 ?g cycloartenol/g dw) and ?-amylase inhibitory activity were not influenced by the maturity stage. The presence of ?-amyrin (25.4 ?g/g dw), myo-inositol (3.17 mg/g dw) and ?-tocopherol (5.1 mg/100 g dw) stood out, so lucuma fruit could have potential as food with antioxidant, hypolipidemic, anti-hyperglycemic, and anti-inflammatory properties. © 2021 Elsevier LtdConsejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
Characterization of starches obtained from several native potato varieties grown in Cusco (Peru)
Abstract: Ten native potato varieties grown in Cusco (at 3,672 m above sea level) were used for starches extraction (at a pilot scale), and their physicochemical, functional, morphological, and structural characteristics were assessed. The content of protein, apparent amylose and phosphorus ranged from 0.1% to 0.44%, 23.42% to 35.5%, and 0.07 to 0.10%, respectively. Starch granules revealed smooth surface, with ellipsoidal and spherical shapes, particle size analysis exhibited bimodal or multimodal distribution, while the averaged crystallinity was 27.7% assessed by XRD. Gelatinization temperatures of the starches ranged from 57.4 to 60.1 °C, 60.8 to 64.3 °C, and 68.4 to 71.1°C for To, Tp, and Tc, respectively; and the gelatinization enthalpies ranged from 15.4 to 17.1 J/g. Respect to pasting properties, the peak viscosity and setback viscosity ranged from 12,970 to 16,970 mPa?s and from 968 to 2498 mPa?s, respectively. Thermogravimetric analysis (TGA) revealed no significant relationship between apparent amylose content and thermal stability. Therefore, the results reveal subtle differences in the functional characteristics of the starches from the native varieties of potato studied, which can be recommended for food industry applications. Practical Application: This study contributes to show several varieties of native potatoes from Cusco and their valorization as nonconventional starch source. Analysis of extreme meteorological events in the central andes of peru using a set of specialized instruments
A set of instruments to measure several physical, microphysical, and radiative properties of the atmosphere and clouds are essential to identify, understand and, subsequently, forecast and prevent the effects of extreme meteorological events, such as severe rainfall, hailstorms, frost events and high pollution events, that can occur with some regularity in the central Andes of Peru. However, like many other Latin American countries, Peru lacks an adequate network of meteorological stations to identify and analyze extreme meteorological events. To partially remedy this deficiency, the Geophysical Institute of Peru has installed a set of specialized sensors (LAMAR) on the Huancayo observatory (12.04◦ S, 75.32◦ W, 3350 m ASL), located in the Mantaro river basin, which is a part of the central Andes of Peru, especially in agricultural areas. LAMAR consists of a set of sensors that are used to measure the main atmosphere and soil variables located in a 30-meter-high tower. It also has a set of high-quality radiation sensors (BSRN station) that helps measure the components of short-wave (SW) (global, diffuse, direct and reflected) and long-wave (LW) (emitted and incident) irradiance mounted in a 6-meter-high tower. Moreover, to analyze the microphysics properties of clouds and rainfall, LAMAR includes a set of profiler radars: A Ka-band cloud profiler (MIRA-35c), a UHF wind profiler (CLAIRE), and a VHF wind profiler (BLTR), along with two disdrometers (PARSIVEL2) and two rain gauges pluviometers. The present study performs a detailed dynamic and energetic analysis of two extreme rainfall events, two intense frost events, and three high-pollution events occurring on the Huancayo observatory between 2018 and 2019. The results show that the rainfall events are similar to the 1965–2019 climatological 90th percentile of the daily accumulated rainfall. The results also highlighted the patterns of reflectivity in function of height for both events, which is measured by highlighting the presence of convective and stratiform rainfall types for both events. The first intense rainfall event was associated with strong easterly circulations at high levels of the atmosphere, and the second one was associated with the presence of strong westerly circulations and the absence of BH-NL system around the central Andes. The first frost event was mainly associated with continuous clear sky conditions in the few previous days, corresponding to a radiative frost event. The second one was mainly associated with the intrusion of cold surges from extra-tropical South America. For both events, the energy budget components were strong-lower in comparison to the mean monthly values during early morning hours. Finally, for the high pollution events, the study identified that the main source of aerosols were the forest fires that took place in Peru with certain contributions from the fires in the northern area of Bolivia. © MDPI AG. A Selective Electrochemical Sensor for the Detection of Cd(II) Based on a Carbon Paste Electrode Impregnated with a Novel Ion-imprinted Hybrid Polymer
An electrochemical sensor (CPE-IIHP) was developed for Cd(II) using a carbon paste electrode (CPE) impregnated with an ion-imprinted hybrid polymer (IIHP). A CPE-NIHP was also prepared for comparison. DPASV was used to optimize the sensor response and quantify Cd(II). The sensor presented a wide linear range from low concentrations of Cd(II): 1 to 100 ?g L?1 and high concentrations of Cd(II): 2.75 to 5.0 mg L?1. Ions such as Co(II), Pb(II), Ni(II), Zn(II), Fe(II), Fe(III), Sn(II) and Cu(II), showed no variation in the Cd(II) signal. The CPE-IIHP was successfully applied in river and drinking water analysis, revealing the great potential for its application.Consejo Nacional de Ciencia, Tecnología e Innovación Tecnológica - Concyte
