Lexical vs. compositional Agency: The role of the context in licencing Agency
In this paper, we propose a taxonomy of verbs based on their compositional behaviour regarding Agency. The taxonomy comprises three primary classes: agent-selecting verbs, non-agent-selecting verbs, and verbs that are lexically underspecified regarding Agency. We describe the semantic and syntactic characteristics of each class using evidence from corpora and considering the potential influence of coercive phenomena in semantic composition, which may be prompted by contextual cues such as agent-oriented or patient-oriented adverbials (intentionally, accidentally). Our analysis focuses on Italian, but the conclusions generally apply to other languages as well

La mobilità sociale a Bergamo tra tardo Medioevo ed Età Moderna: fonti e metodi d’indagine
This article discusses sources and methods to analyze the dynamics of social mobility in the ancient régime, focusing on the case of Bergamo between the fifteenth and eighteenth centuries. After a brief methodological reflection, the evolution of Bergamo’s economic and demographic context over the centuries examined is outlined. Thanks to the analysis of fiscal sources (tax-land registers) and the registers of municipal deliberations, two indicators are then proposed to measure social mobility in relation to the distribution of wealth and to the access to local political institutions. Finally, by selecting some exemplary cases among the local kinships, the possible relationship between the two mobility dynamics examined is considered
Role of gelator concentration in shaping the molecular and macroscopic properties of thymol:camphor supramolecular eutectogels
Monoterpenoid-based hydrophobic deep eutectic solvents (HDES) are attracting attention across various industrial sectors. A further advancement is their application as gel materials, known as eutectogels, which enable a broader range of task-specific applications. Here we report the straightforward preparation of hydrophobic eutectogels (HEGs) through the self-assembly of the low molecular weight gelator 1,3:2,4-dibenzylidene-D-sorbitol (DBS) within the thymol:camphor (Thy:Cam) HDES at 1:1 molar ratio. Recognizing that gelator concentration is crucial for tailoring the properties of the HEGs, five eutectogels were prepared with DBS concentrations ranging from 1 wt% to 10 wt%, forming sustainable gels in minutes. Comprehensive characterization at both molecular and macroscopic scales revealed a complex relationship between gelator content and the resulting material properties. All systems exhibit significant thermal stability, with degradation temperatures as high as 200 °C. Intermolecular interactions between HDES species and the DBS matrix, as well as possible microheterogeneities, were found to strongly influence the rheological and thermal behavior of the eutectogels, as well as the molecular mobility. Altogether, our results imply that HDES species experience multiple dynamic processes, underscoring the intricate interplay between the HDES components and the DBS matrix. This research highlights the critical role of gelator content in tuning the bulk and molecular physicochemical properties of Thy:Cam HEG and provides critical insights into the design of tailored gel materials. The findings emphasize the need for multiscale investigations to fully understand and optimize these materials, contributing to further advancements in innovative applications in material science, green chemistry, and biotechnology
Altering substrate specificity of a thermostable bacterial monoamine oxidase by structure-based mutagenesis
Bacterial monoamine oxidases (MAOs) are FAD-dependent proteins catalyzing a relevant reaction for many industrial biocatalytic applications, ranging from production of enantiomerically pure building blocks for pharmaceutical synthesis to biosensors for monitoring food and beverage quality. The thermostable MAO enzyme from Thermoanaerobacterales bacterium (MAOTb) is about 36 % identical to both putrescine oxidase and human MAOs and can be efficiently produced in Escherichia coli. MAOTb preferentially acts on n-alkyl monoamines but shows detectable activity also on polyamines and aromatic monoamines. The crystal structures of MAOTb in complex with putrescine, benzylamine, spermidine and n-heptylamine at resolution ranging from 1.6 to 2.3 Å resolution revealed the binding mode of substrates to the enzyme. The MAOTb active site is highly conserved in the inner part of the cavity in front of the flavin ring (re face), where the presence of two tyrosine residues creates the substrate amine binding site that is found also in human MAOs. Instead, more distantly from the flavin, the entrance of the catalytic site is much more open in MAOTb and features a different arrangement of amino acids. Site-directed mutagenesis targeting residues Ala168, Thr199 and Val324 allowed the identification of key residues in ligand binding to alter substrate specificity. The A168D variant showed a higher activity on putrescine than wild-type, whereas by replacing either Thr199 or Val324 to Trp a marked enhancement in kcat/KM values was found on n-alkyl-monoamines and on aromatic amines
