Linear psoriasis or ILVEN? A pediatric case unraveling the mosaic puzzle
: Linear psoriasiform eruptions in children are diagnostically challenging, particularly when they follow Blaschko's lines. The main differential diagnosis includes inflammatory linear verrucous epidermal nevus (ILVEN) and linear psoriasis (LP), a rare mosaic variant. We describe the case of a 5-year-old boy with extensive unilateral lesions clinically suggestive of both entities. Histology revealed Munro microabscesses and psoriasiform hyperplasia. The patient showed rapid improvement with topical calcipotriol/betamethasone, followed by long-term remission. Notably, after 8 years of follow-up, the lesions remained strictly confined to the original Blaschkoid distribution, with no extracutaneous involvement and only mild recurrences. This clinical course strongly supports the diagnosis of isolated linear psoriasis (type 1 mosaicism). Few pediatric cases in the literature have documented such a long and stable evolution, making this report a valuable contribution to the distinction between LP and ILVEN. The case highlights the importance of integrating clinical, histological, and longitudinal data when evaluating linear dermatoses in children
Interaction of Erdosteine with TrkA Signaling Pathways: Implications for Analgesia
Thiol-containing drugs may interact with a region of tropomyosin receptor kinase A (TrkA), potentially inhibiting its activation by nerve growth factor (NGF). This action has been linked to potential analgesic activities. Here, we describe the ability of erdosteine, a thiolic compound classified as a mucolytic agent, to bind to the TrkA receptor sequence in silico and its in vitro effects on TrkA activation induced by NGF in cultured human neuroblastoma cells. Our results show that erdosteine and its metabolite, Met-1, bind to the TrkA receptor pocket, involving the primary TrkA residues Glu331, Arg347, His298, and His297. Furthermore, Met-1 has the ability to reduce the disulfide bridge between Cys300 and Cys345 of TrkA. In vitro measurement of TrkA autophosphorylation following NGF activation confirmed that erdosteine and Met-1 interfere with NGF-induced TrkA activation, leading to a consequent loss of the molecular recognition and spatial reorganization necessary for the induction of the autophosphorylation process. This effect was inhibited by low millimolar concentrations of the two compounds, reaching a maximal inhibition (around 40%) after 24 h of exposure to 1 mM erdosteine, and then plateauing. These findings suggest that erdosteine can act as a TrkA antagonist, thus indicating that this drug may have potential as an analgesic via a novel non-opioid mechanism of action operating through NGF signaling inhibition at the level of TrkA

issemination and publishing. This paper aims to outline a panorama of current research issues, challenges and practices between Digital Humanities and Digital Cultural Heritage, and to present some case studies addressed in the DAda and PLAY Lab at the University of Pavia
Non-invasive PD-L1 stratification in non-small cell lung cancer using dynamic contrast-enhanced MRI
Objectives: This study aimed to assess whether pharmacokinetic parameters derived from DCE-MRI can stratify Programmed Death-Ligand 1 (PD-L1) expression in NSCLC. The secondary aim was to identify a suitable pharmacokinetic model configuration for anisotropic temporally-spaced DCE-MRI sequences, considering Tofts variants, population-averaged arterial input functions (AIF), and bolus arrival time (BAT) estimation methods. Materials and methods: From April 2021 to May 2023, patients with locally advanced non-small cell lung cancer (NSCLC) were prospectively enrolled. Tumors were categorized based on: PD-L1 absence/presence (threshold 1%) and hyperexpression/hypoexpression (threshold 50%). Pharmacokinetic parameters were extracted using several candidate configurations; fit quality was evaluated using coefficient of determination (R2). Mann–Whitney U-test and ROC-AUC were used to assess correlation with PD-L1 for the best-fit configuration. Results: Thirty-eight patients (mean age 68 ± 9 years, 28 men) were included. PD-L1 expression was present in 25 patients (66%) and absent in 13 (34%). PD-L1 was hyperexpressed in 13 (34%) patients and hypoexpressed in 25 (66%). Voxel-wise pharmacokinetic parameters were extracted using the best-fit configuration—extended Tofts model (ETM) with Georgiou AIF and Peak-Gradient (PG) BAT estimation (R2 = 0.79). Ktrans median (0.25 vs. 0.12 min−1, p = 0.02), Ktrans standard deviation (0.32 vs. 0.23 min−1, p = 0.01) and Kep median (1.09 vs. 0.59 min−1, p = 0.02) were significantly higher in PD-L1 < 50% group (ROC-AUC 0.71–0.76). Conclusion: DCE-MRI pharmacokinetic parameters could stratify PD-L1 hypo/hyperexpression in NSCLC. The ETM with PG BAT estimation method and Georgiou AIF was the best-performing pharmacokinetic configuration. Findings Quantitative DCE-MRI parameters Ktrans (the volume transfer rate) and Kep (the efflux rate constant) show potential for distinguishing PD-L1 hyperexpression from hypoexpression. Clinical relevance Preliminary results suggest that DCE-MRI could be a safe method to stratify PD-L1 hypo/hyperexpression in non-small cell lung cancer, potentially optimizing treatment decisions, given the high cost of immunotherapy
Cystic Artery Bleeding: Imaging Insights and Systematic Review of Endovascular Management
Background: Cystic artery bleeding (CAB) is a rare but potentially life-threatening condition. Its etiologies span iatrogenic trauma, inflammatory diseases, and trauma, often presenting variably as hemoperitoneum, upper gastrointestinal bleeding, or hemorrhagic shock. The clinical heterogeneity of CAB complicates its diagnosis, necessitating a high index of suspicion and reliance on imaging modalities, particularly computed tomography (CT), for accurate identification of bleeding sources and differentiation from other causes of abdominal pain. Methods: This pictorial essay highlights key imaging findings in CAB and pseudoaneurysms, emphasizing the role of ultrasound, CT, and digital subtraction angiography (DSA) in diagnosis and management planning. Additionally, a systematic review of transcatheter arterial embolization (TAE) is presented, consolidating data from 64 studies encompassing 90 patients. Results: The review evaluates patient demographics, etiologies, clinical presentations, and procedural outcomes, underscoring TAE’s high efficacy and safety as a first-line treatment. Conclusions: The findings reinforce the importance of early diagnosis and tailored intervention strategies to optimize outcomes in CAB management
Pathogenic mutation impairs functional dynamics of Hsp60 in mono- and oligomeric states
Mitochondrial chaperonin Heat Shock Protein 60 kDa (Hsp60) oversees the correct folding of client proteins in cooperation with Hsp10. Hsp60 monomers M first form 7-meric Single rings (S), which then pair into 14-meric Double rings (D) that accommodate clients in their lumen. Recruitment of 7 Hsp10 molecules per pole yields a sealed 28-meric Football-shaped complex (F). ATP hydrolysis in each Hsp60 unit drives client folding and F disassembly. The V72I mutation in hereditary spastic paraplegia form SPG13 impairs Hsp60 function despite being distant from the active site. We here investigate this impairment with atomistic molecular dynamics (MD) simulations of M, S, D, and F for both WT and mutant Hsp60, considering catalytic aspartates in D and F in different protonation states (even simulating one such state of D post-hydrolysis). Our findings show that—as observed experimentally—V72I rigidifies Hsp60 assemblies, significantly impacting internal dynamics. In monomers, V72I introduces a new allosteric route that bypasses the ATP binding site and affects mechanisms driving reactivity. These insights highlight a multiscale complexity of Hsp60 that could inspire the design of experiments to better understand both its WT and V72I variants

Wikibase LibMovIt is a project developed within the framework of PRIN 2022 LibMovIt – Libraries on the Move: Scholars, Books, Ideas Traveling in Italy in the 18th Century, funded by the European Union – Next Generation EU. The research is based on published accounts by travelers documenting their experiences of the scholarly journey known as the Grand Tour. This instance of Wikibase Cloud was created with the purpose of publishing the data generated over the course of the research, through the development of a specifically designed dataset. Introduction on the Concept of the Aesthetic of Regenerative Design and Development
While the challenges for the socioecological future are becoming harder and harder, the idea of a “sustainable development” is no more enough: is clear the need for new concepts and strategies within contemporary design, able to propose innovative solutions to regenerate our original environments. While the concept of not only limiting socio-environmental damage but actively restoring and regenerating existing ecosystems is not new in the field of design, its focus on the Latin-Mediterranean context, rich in environmental resources and architectural traditions, has not been extensively explored. In this region, where concerns about socioenvironmental decisions meet debates in practice and academia, a formal collection of experiences, good practices, and theoretical considerations is more than ever needed. A seven-year-long collaboration among Tecnológico de Monterrey (Mexico) and University of Pavia (Italy) set the bases for this book, which aims to provide the initial foundation for this necessary “formal collection”. The focuses on ecological design and social design meet in this edited book, with the aim to propose a holistic view on regenerative design in Latin-Mediterranean context
