Tetrahydropyrido[4,3-d]pyrimidines as a new active scaffold for human topoisomerase II inhibitors

    Get PDF
Human topoisomerase II (topoII) is a well-known and validated target for cancer treatment. We previously reported a first set of 6-amino-tetrahydroquinazoline derivatives as novel human topoII inhibitors. Here, we report on the expansion and this molecular scaffold and present 17 additional analogs centered on the tetrahydropyrido[4,3-d]pyrimidine heterocycle. Some of these compounds exhibit promising topoII inhibitory and antiproliferative activities. Compound 24 (ARN21929) shows good in vitro potency, with an IC50 of 4.5 ± 1.0 μM, excellent kinetic and thermodynamic solubility, and good metabolic stability. Our results indicate that this new chemical class of topoII inhibitors deserves further exploration and represents a promising starting point for developing novel and potentially safer topoII-targeted anticancer drugs

The Internationalisation of Veneto schools: Addressing a growing superdiversity. A mixed-method study on the impact of Erasmus+ short-term mobilities and eTwinning virtual projects on the linguistic, intercultural and digital dimensions of internationalisation in secondary schools.

Recent studies on the topic of internationalisation (Deardorff, 2020; Baiutti, 2019) mostly focus on higher education (HE) or long-term mobilities of school staff and students, while short term mobilities (from 5 to 30 days), as well as virtual and blended partnerships, are less considered, even though they represent the majority of international experiences in schools, especially upper secondary ones, particularly after the two years of stop due to the Covid-19 pandemic. As noticed by Yemini (2014), the dimensions of school internationalisation developed in the HE sector cannot fully apply to elementary and secondary schools, since the contexts are quite different. However, a systematic literature review retrieved a limited number of studies on the topic, mostly relevant to the Asian and American areas. Therefore, there seems to be a need for updated research on school internationalisation, especially as far as intercultural, language and digital education is concerned, from a specific local perspective (Humphreys, 2023). The Italian region of Veneto, presently facing the new challenges posed by globalization, such as an increasing superdiversity (Vertovec, 2007) seems to be the perfect location. Therefore, building on European and National school internationalisation programmes, the present study aims to answer the following research questions: • How has the internationalisation of primary and secondary schools changed in the specific Italian region of Veneto in the last five years in the three main dimensions of students’ intercultural, plurilingual and digital competences development? • Which experiences led to a significant development of intercultural, plurilingual and digital competences in high school students and can be considered as best practices? • How are these developments assessed? The research paradigm is pragmatic (Dewey, 1920-2008) in the definition of both the problem and the research design, and the methodology used to investigate the topic and try to find answers to the research questions has been the Explanatory sequential mixed-method design (Younas et al., 2019). Data were collected through three online questionnaires and two in-depth case studies. Outcomes presentation was carried out through a series of joint displays (Creswell & Plano-Clark, 2018). Results from data analysis point towards a substantial potential for further development in the internationalisation of schools, and the development of new approaches in teaching and learning English, especially as far as CPD of teachers in concerned. Conclusions drawn from research show that Erasmus+ short term mobilities can enhance both students’ and teachers’ linguistic, intercultural and digital competences, and that the combination of physical mobilities with eTwinning virtual collaboration and innovative ways of teaching English can help students’ assessment being blended in the curriculum

An integrative conservation and management strategy based on biological and cultural diversity assessment: A case study of Miaoling mountainous region, China

    No full text
The Miaoling mountainous region, a typical karst landscape in Southwest China, is rich in biological resources and possesses an indigenous cultural heritage. However, ecological degradation and the encroachment of external cultures present significant threats to these areas, creating dual challenges to their biological and cultural integrity. To effectively protect the bio-cultural features in the Miaoling region, this study proposes a strategy for establishing a conservation framework based on the evaluation of bio-cultural diversity for protection. First, the four indicators of biodiversity and five indicators of cultural diversity were used to assess the value of bio-cultural diversity. Next, the Zonation model was integrated to identify priority zones. Finally, using these priority zones, a ’source − corridor − network’ strategy was developed. The results have revealed that nearly half of the region exhibits a lack of coordination between bio-cultural diversity. The priority conservation zones for bio-cultural diversity, which cover a total area of 2,286.76 km2, are located within small watershed and agroforestry ecosystems, displaying a fragmented distribution. Also, the conservation network encompasses 29 primary corridors, 76 secondary corridors, and 25 nodes. Based on that, a multi-stakeholder adaptive management framework has been proposed, emphasizing policy and financial support, community participation, and collaboration to integrate ecological protection with sustainable development. This study highlights the potential of bio-cultural diversity assessments in identifying priority zones and guiding strategic conservation planning in the karst mountainous regions

Field Testing of a Smart Geomat Based on Fiber Optic Sensors and Sensor Fusion for Levee Monitoring

    Get PDF
Real-time monitoring of the functionality of river embankments, especially during flood events, is an increasingly important topic in the new scenario outlined by climate change. To address this issue, the concept of the “smart levee” has recently emerged, based on the fusion of information from different types of sensors to obtain, remotely and in real time, an improved knowledge of its structural health status. The work here presented applies this concept by proposing an innovative monitoring solution consisting of an anti-erosion reinforced geomat equipped with Fiber Optic Sensors. This “smart revetment” aims to detect any surface displacement induced by failure mechanisms such as erosion and piping phenomena, as well as any physical change in soil properties (e.g., temperature and water content). These measurements, once integrated with other environmental variables, can then be associated with the variability of the most common environmental and operational factors, such as the ri..
