Germination requirements of the endangered cliff-dwelling Primula palinuri Petagna and implications for conservation
Primula palinuri, an endangered plant species strictly confined to the coastal limestone cliffs of southern Italy, represents a key Mediterranean endemic threatened by habitat loss and environmental stressors. This study investigated the effects of temperature, salinity, water stress, and burial depth on its seed germination. Germination was favoured by cool temperatures, with an optimum at 15 °C. Rates significantly declined at both 5 °C and 25 °C. Increasing salinity markedly inhibited germination, confirming the species’ sensitivity to salt stress. Water stress experiments demonstrated that P. palinuri requires high soil moisture for successful germination, with total inhibition under moderate (- 0.4 MPa) to severe drought conditions (- 1.5 MPa). Burial depth negatively affected seedling emergence, likely due to the small seed size and light-dependency. These results underline the species’ narrow ecological requirements, emphasising the need for careful habitat selection in conservation actions, favouring moist, shady, and salt-protected microsites with shallow seed placement
Revised “iRR6” model in intermediate-1 risk myelofibrosis patients treated with ruxolitinib
Background: The response to ruxolitinib after 6 months (RR6) model allows early identification of ruxolitinib-treated myelofibrosis (MF) patients with poorer overall survival (OS); however, it is less applicable to lower-risk patients. Methods: To further explore this, the authors performed a subanalysis of the “RUX-MF” study (NCT06516406) with an aim to validate the RR6 and to develop a score specific for intermediate-1 DIPSS/MYSEC-PM risk patients. Results: Among the 776 evaluable patients, 34.4%, 47.8%, and 17.8% were at low, intermediate, and high RR6 risk, with 5-year OS of 64.1%, 51.8%, and 44.5%, respectively (p 2, 33.9%), with 5-year OS of 84.8%, 76.4%, and 56.6%, respectively (p <.0001). The iRR6 model was validated in a cohort of 95 intermediate-1 risk patients from the Moffitt Cancer Center, yielding stratification into the same three risk categories, with 5-year OS of 83.3% (low-risk), 71.7% (intermediate-risk), and 54.5% (high-risk) (p =.01). Conclusions: The iRR6 model provides a more refined tool for the identification of intermediate-1 MF patients who may benefit from early therapy shift

Commuting by bicycle (vs. by car) is associated with improved aerobic power, microvascular function and diminished CO2 output in the atmosphere
The study investigated whether bicycle compared with car commuting, over relatively small distances, has positive effects on physiological variables, cardiometabolic fitness and CO2 output in the atmosphere. Bike Commuters (11 M, 15 F; age [median value (interquartile range)] 51.5 (38.3–56.8) years; body mass index [BMI] 22.8 (21.0–24.1) kg m−2) were compared with Car Commuters (12 M, 19 F; age 47.0 (36.0–56.5) years; BMI 23.5 (21.4–24.9) kg m−2). In a longitudinal arm of the study, 20 Car Commuters were re-evaluated after they switched for 24 weeks to bicycle commuting (Car→Bike Commuters). Measurements included peak aerobic power ((Formula presented.)) and ventilatory thresholds on a cycle ergometer, blood flow increase in the common femoral artery during a passive leg movement (PLM) test, energy expenditure and (Formula presented.) exhaled during commuting. Bike Commuters had higher (Formula presented.) (33.7 (31.3–38.1) versus 25.3 (23.5–28.9) mL kg−1 min−1, P < 0.001) and ventilatory thresholds than Car Commuters, higher Δpeak blood flow (+25%, P = 0.04) and area under the blood flow versus time curve (+46%, P = 0.03) during PLM, and an enhanced skeletal muscle oxidative metabolism. (Formula presented.) and PLM variables increased in Car→Bike Commuters. Metabolic CO2 output during bicycle commuting was ∼12 times less than that for a petrol car. In moderately active individuals, short-distance bicycle commuting at moderate intensity was associated, compared with car commuting, with positive effects on several physiological functions and environmental factors

Class IIa HDACs forced degradation allows resensitization of oxaliplatin-resistant FBXW7-mutated colorectal cancer
: Epigenetic plasticity and large-scale chromatin remodeling characterize tumor evolution and the emergence of subclones resistant to conventional therapies. Catalytically inactive class IIa HDACs (HDAC4, HDAC5, HDAC7, HDAC9) control the targeted recruitment of chromatin remodeling complexes, making them attractive therapeutic targets in oncology. In this study, we found that HDAC4 is degraded by the proteasome in cancer cells with impaired DNA repair by homologous recombination and after oxaliplatin (OXPT) treatment. Genetic screening identified FBXW7 as the E3 ligase responsible for HDAC4 degradation. FBXW7 loss-of-function mutations are frequently found in patients with colorectal cancer (CRC) and were found associated with the development of resistance to OXPT. Forced degradation of Class IIa HDACs using a PROTAC-based compound restored OXPT sensitivity in FBXW7-mutated CRC cells, patient-derived organoids (PDOs), and mice. Mechanistically, removal of HDAC4 in FBXW7-mutated CRC treated with OXPT recreated an epigenetic state comparable to OXPT-sensitive cells. Furthermore, patient profiling based on the epigenetic state of the super-enhancers controlled by HDAC4 successfully identified a priori CRC patients resistant to platinum. This study supports HDAC4 as a key mediator of oxaliplatin resistance in FBXW7-mutated CRC and highlights the remodeling of a well-defined super-enhancer repertoire as part of the process of OXPT resensitization
The importance of patient fitness in expert and multidisciplinary multiparametric management of HCC: a narrative review
Hepatocellular carcinoma (HCC) management is a challenging task. Despite continuous efforts to develop more effective treatments, patient prognosis often remains poor. Recently, a new management concept-the “multiparametric therapeutic Hierarchy”-has been proposed, with the hypothetical advantage of avoiding the risk of undertreatment inherent in “classical” HCC management modalities while also minimizing the risk of overtreatment that may occur when inexperienced clinicians adopt overly aggressive treatment hierarchies. The “multiparametric” concept emphasizes a more comprehensive evaluation of each patient, focusing on their individual clinical presentation. Among the various factors considered, patient fitness plays a critical role. Here, fitness is understood as the set of patient-specific characteristics that can influence the outcomes of therapies within the hierarchical framework (or even preclude certain therapies altogether). This multifaceted concept extends beyond traditional “Performance Status” measurements, incorporating considerations of frailty and comorbidity. In this review, we explore the available evidence regarding the role of patient fitness within the multiparametric therapeutic hierarchy for HCC. Finally, we discuss fitness considerations across different levels of the therapeutic hierarchy, highlighting current evidence, existing challenges, and limitations, with the aim of providing new insights for physicians involved in HCC management

This leads to a dilemma for the critic of anthropocentrism: either they accept immanentism (i.e., the rejection of the natural-unnatural and natural-supernatural distinctions, resulting in the denial of ethical indifferentism and thereby trumping any criticism of anthropocentrism) or they reject it (and must then acknowledge the legitimacy of non-immanentist forms of anthropocentrism that view nature as sacred within a broader supernatural framework)

Dans Lire Houellebecq, Raphaël Baroni analyse les mécanismes qui influencent les jugements des lecteurs sur les textes narratifs, en se concentrant sur la réception des romans de Michel Houellebecq. S’appuyant sur les travaux de Liesbeth Korthals Altes et Jérôme Meizoz, il montre comment la posture publique de l’auteur influe sur l’interprétation de son œuvre. L’ambiguïté de l’énonciation et les thématiques controversées de Houellebecq brouillent les frontières entre fiction et réalité, suscitant des vastes débats. L’étude explore également la présence transmédiale de l’auteur et met en lumière une mutation dans la réception littéraire, où contexte et contenu thématique jouent un rôle clé dans la lisibilité d’un texte. Ageing and Gender in European Cinema (AGE-C)
The AGE-C dataset results from a data-driven investigation into ageing and gender in contemporary European cinema (2014-2023) – an intersectional approach to film representation and film labour so far missing – intervening in the mostly qualitative field of ageing and film with quali-quantitative data from 9 European territories. The dataset collects data on fiction, documentary and animation feature films in relation to filmmakers and ageing actors’ biographical details. It also includes data on roles, marketing, nationality, genres, film awards, documentary subjects and film characters. An innovative tag system relates characters and films to topics in gerontology, describing their narrative and thematic content, as well. The dataset also factors in awards and film festivals, illuminating the gender gap affecting 50+ actors. The data set was last updated on October 2, 2025
