    CLSI Validation of Exchangeable Copper Determination in Serum by ICP-MS: A Focus on Alzheimer's Disease and Wilson Disease

    Background: Copper dyshomeostasis has been implicated in a subset of Alzheimer's disease (AD) patients, characterized by elevated non-ceruloplasmin-bound copper (non-Cp Cu). However, traditional methods for estimating non-Cp Cu are indirect and analytically imprecise. This study introduces and validates a direct assay for exchangeable copper (ExcCu) by inductively coupled plasma-mass spectrometry (ICP-MS), compliant with Clinical and Laboratory Standards Institute (CLSI) guidelines. Methods: We performed analytical validation of the ExcCu assay following CLSI protocols (EP5, EP6, EP7, EP9, EP15, and EP28). ExcCu and other copper-related biomarkers were quantified in serum samples from 154 healthy controls, 82 AD patients, and 10 patients with Wilson disease (WD). Diagnostic performance was evaluated via receiver operating characteristic (ROC) curve analysis, and inter-method agreement was assessed using Bland-Altman plots. Results: The ExcCu assay demonstrated excellent linearity, precision (CV < 6%), and inter-laboratory reproducibility. Among AD patients, ExcCu levels were significantly elevated compared to controls (p < 0.001). ExcCu distinguished AD from controls with an AUC of 0.80 and a specificity of 95%. Compared to non-Cp Cu, ExcCu yielded no negative values and showed reduced bias. The relative exchangeable copper (REC) index was more effective in differentiating AD from WD (AUC = 0.88). Conclusions: The validated ExcCu assay overcomes the limitations of the traditional non-Cp Cu calculation, offering a reliable biomarker for copper-related AD subtypes. Its high specificity supports its use in patient stratification, potentially contributing to personalized approaches in AD diagnosis and therapy

    Status of Sepsis Care in European Hospitals Results from an International Cross-Sectional Survey

    : Rationale: Early detection, standardized therapy, adequate infrastructure, and strategies for quality improvement should constitute essential components of every hospital's sepsis plan. Objectives: To investigate the extent to which recommendations from the sepsis guidelines are implemented and the availability of infrastructure for the care of patients with sepsis in acute-care hospitals. Methods: A multidisciplinary cross-sectional questionnaire was used to investigate sepsis care in hospitals. This included the use of sepsis definitions, the implementation of sepsis guideline recommendations, diagnostic and therapeutic infrastructure, antibiotic stewardship, and quality improvement initiatives (QIIs) in hospitals. Measurements and Main Results: A total of 1,023 hospitals in 69 countries were included. Most of them, 835 (81.6%), were in Europe. Sepsis screening was used in 54.2% of emergency departments (EDs), 47.9% of wards, and 61.7% of ICUs. Sepsis management was standardized in 57.3% of EDs, 45.2% of wards, and 70.7% of ICUs. The implementation of comprehensive QIIs was associated with increased screening (EDs, +33.3%; wards, +44.4%; ICUs, +23.8% absolute difference) and increased standardized sepsis management (EDs, +33.6%; wards, +40.0%; ICUs, +17.7% absolute difference) compared with hospitals without QIIs. A total of 9.8% of hospitals had implemented ongoing QIIs, and 4.6% had invested in sepsis programs. Conclusions: The findings indicate that there is considerable room for improvement in a large number of mainly European hospitals, particularly with regard to early identification and standardized management of sepsis, the availability of guidelines, diagnostic and therapeutic infrastructure, and the implementation of QIIs. Further efforts are required to implement a more comprehensive and appropriate quality of care

    La qualità della didattica universitaria può essere sostenuta con una attenzione continua all’innovazione didattica e con una formazione dedicata, attraverso iniziative di Faculty Development. L’Università di Torino promuove iniziative di ricerca e formazione su questi temi con particolare attenzione al coinvolgimento di tutte le componenti della comunità accademica e con un focus specifico alla sfida digitale. Nell’autunno 2023 il Teaching and Learning Center UniTo ha proposto una call sull’innovazione didattica (“Abbraccia il futuro: libera l’innovazione in aula”) che ha previsto la presentazione di progetti, da sviluppare in una giornata dedicata e strutturata in sessioni di lavoro in modalità collaborativa. La Call ha generato coinvolgimento, soddisfazione e condivisione di idee ed esperienze. Sono stati presentati 72 progetti che hanno coinvolto la quasi totalità dei Dipartimenti e delle Direzioni, con circa 500 partecipanti (personale docente, tecnico-amministrativo, non strutturato, comunità studentesca). Le proposte presentate hanno riguardato diverse le aree disciplinari e un numero ampio di corsi di studio, è possibile individuare tematiche comuni e trasversali. Il contributo presenta i risultati della Call e sono inoltre delineate alcune strategie di promozione dell’innovazione didattica all’interno del contesto accademico, a partire da questa esperienza

This aspect urges organizations of all types and sizes to equip individuals with the competences and ethical values needed to continually adapt work processes with a focus on sustainability, understood as a virtuous balance between environmental protection, economic productivity, and human well-being. In particular, the development of new professions in a rapidly changing job market, characterized by high levels of uncertainty, calls for a proper understanding of the relationship between young people and work. It also highlights the importance of the personal, intersubjective, and context-specific dimensions of learning, within a circular dynamic between theory and practice
