    This study aims to provide a contextualised analysis of a well-known document that has never before been examined in detail from palaeographical and diplomatic perspectives: the agreement signed in 1105 by the inhabitants of Gaeta and Tolomeo, count of Tusculum, to settle a dispute concerning the shared ownership of a boat. This dispute had previously involved Tolomeo’s father, Gregorio. The document is remarkable for its graphic elements, especially in the section about Tolomeo, which seems to be the result of a deliberate effort by the count and his scriniarius to achieve a sense of ‘exoticism’. Relating these elements to what we know about Tolomeo – starting with the Latin form of his name, Ptolomeus – and the counts of Tusculum between the 11th and 12th centuries reveals that they form part of a broader political and cultural programme implemented by the family to distance itself from its Roman and papal past and establish a new “seigneurial” identity

    A rectilinear-upward planar drawing of a digraph G is a crossing-free drawing of G where each edge is either a horizontal or a vertical segment, and such that no directed edge points downward. Rectilinear-Upward Planarity Testing is the problem of deciding whether a digraph G admits a rectilinear-upward planar drawing. We study the complexity of Rectilinear-Upward Planarity Testing and provide several algorithmic results. Precisely, we prove that: (i) the problem is NP-complete, even if G is biconnected; (ii) it can be solved in linear time when an upward planar embedding of G is fixed; the problem is polynomial-time solvable for biconnected digraphs of treewidth at most two, i.e., for digraphs whose underlying undirected graph is a series-parallel graph; (iii) the problem is fixed-parameter tractable (namely, fixed-parameter linear) for all biconnected graphs, when parameterized by the number of sources and sinks in the digraph

    The natural L370F ERα variant confers endocrine resistance and sensitivity to ATRA in metastatic breast cancer cells

    Background: Metastatic breast cancer (MBC) remains a major clinical challenge, particularly in estrogen receptor α (ERα)-positive patients who develop resistance to endocrine therapy (ET). While hotspot mutations such as Y537S in the ligand-binding domain (LBD) are well-characterized drivers of resistance, other ERα variants remain poorly studied. Understanding the molecular mechanisms underlying resistance in these variants is crucial for identifying novel therapeutic strategies. Here, we investigated the functional role of the L370F and E471D ERα variants, which are spatially close in the ERα structure. Methods: Stable overexpressing HEK293 cells and CRISPR/CAS9 engineered MCF-7 cells were generated and treated with 17β-estradiol (E2), fulvestrant (Ful) and all-trans retinoic acid (ATRA) to measure ERα stability, transcriptional activity and gene expression analyses using different cellular assays and RNASeq techniques. Direct in vitro measurement of ligand binding affinity to ERα were performed using the purified full-length wild type (wt) as well as L370F and Y537S ERα. In silico structural simulations were also performed to predict the structure of the mutated L370F ERα. Senescence analyses of MCF-7 and Y537S MCF-7 cells were performed using direct measurement β-galactosidase activity in vitro and in cell lines. Results: The L370F variant conferred resistance to Ful in terms of in vitro ERα binding, ERα transcriptional activity, receptor degradation and cell proliferation by modifying the folding of the receptor structure. Furthermore, L370F-expressing cells exhibited a hyperactive response to low doses of E2 and basally upregulated late estrogen responsive genes. Additionally, we found that both L370F and Y537S ERα variants displayed increased RARα expression, rendering them highly sensitive to ATRA. Notably, ATRA killed L370F-expressing cells and induced senescence in Y537S-expressing cells, highlighting mutation-specific responses. Conclusions: Our findings expand the understanding of ERα mutations beyond known hotspots, identifying L370F as a novel mutation contributing to ET resistance and further indicate the necessity to characterize all the less-studied ERα variants found in MBC. Furthermore, we demonstrate that ATRA selectively targets MBC cells harboring L370F and Y537S mutations, suggesting its potential as a mutation-specific therapeutic agent. These results support further investigation of ATRA in clinical settings to improve treatment strategies for ERα-mutant MBC

    Le stampe della selva oscura in due edizioni cinquecentesche della 'Comedia' di Dante Alighieri nella Biblioteca Paolo Baffi della Banca d'Italia

    Costituiscono oggetto di studio di questo contributo le rappresentazioni della selva oscura nelle illustrazioni a stampa dei primi due volumi di pregio da cui prende inizio il Tour virtuale Dante nella Biblioteca Paolo Baffi. I due volumi, entrambi stampati a Venezia, sono le edizioni della Comedia con l'esposizione di Cristoforo Landino (1529, stampata per Iacob del Burgofranco su istanza di Lucantonio Giunta) e con l'esposizione di Alessandro Vellutello (1544, stampata per Francesco Marcolini su istanza dello stesso Vellutello). Analizzando queste stampe, per la prima volta ne è stata ricostruita la genesi e sono stati individuati elementi significativi sul diverso contesto storico, culturale e politico in cui sono state pubblicate, nella stessa città, a distanza di 15 anni l'una dall'altra. Riguardo l'illustrazione presente nell'edizione del 1529, è stato individuato, per la prima volta, l'adattamento di una matrice già utilizzata in una edizione del 1491, impiegando la stessa rappresentazione dell'episodio dantesco, inserito in una cornice figurata diversa. Grazie all'identificazione di tutti i soggetti raffigurati nella stampa dell'edizione del 1529 (comprensiva di episodio e cornice) e al raffronto con altre stampe, interne ed esterne al volume, sono emersi contenuti iconografici legati all'ambiente antimediceo, con il motto stoico «SVSTINE ET ABSTINE», e di natura economico-editoriale. Quanto alla stampa presente nell'edizione del 1544, nonostante le componenti innovative del libro, su cui si è concentrata la bibliografia rintracciata, per la prima volta sono stati individuati e dimostrati anche elementi di continuità con la tradizione della specifica illustrazione, fatti propri e resi più moderni, in linea con un mutato atteggiamento culturale strettamente correlato con l'epoca del pontificato di Paolo III Farnese, a cui Vellutello ha dedicato il volume, il papa umanista che avrebbe dato inizio al Concilio di Trento nel 1545
