Development of a simple and highly sensitive virion concentration method to detect SARS-CoV-2 in saliva
Background
Controlling novel coronavirus pandemic infection (COVID-19) is a global challenge, and highly sensitive testing is essential for effective control. The saliva is a promising sample for high-sensitivity testing because it is easier to collect than nasopharyngeal swab samples and allows large-volume testing.
Results
We developed a simple SARS-CoV-2 concentration method from saliva samples that can be completed in less than 60 min. We performed a spike test using 12 ml of saliva samples obtained from healthy volunteer people, and the developed method performance was evaluated by comparison using a combination of automatic nucleic acid extraction followed by RT-qPCR detection. In saliva spike tests using a 10-fold dilution series of SARS-CoV-2, the developed method was consistently 100-fold more sensitive than the conventional method.
Conclusions
The developed method can improve the analytical sensitivity of the SARS-CoV-2 test using saliva and speed up and save labor in screening tests by pooling many samples. X-Ray computed tomography meets robust chemometric latent space modeling for lean meat percentage prediction in pig carcasses
This study presents a case of processing X-ray computed tomography (CT) data for pork scans using chemometric latent space modeling. The distribution of voxel intensities is shown to exemplify a multivariate, multi-collinear signal mixture. While this concept is not novel, it is revisited here from a chemometric perspective. To extract meaningful information from such multivariate signals, latent space modeling based on partial least squares (PLS) is an ideal solution. Furthermore, a robust PLS approach is even more effective for latent space modeling, as it can extract latent spaces unaffected by outliers, thereby enhancing predictive modeling. As an example, lean meat percentage is predicted using X-ray CT data and robust PLS regression. This method is applicable to X-ray CT quantification analysis, particularly in cases where unclear, erroneous, and outlying observations are suspected in the data.info:eu-repo/semantics/publishedVersio
Toxicity Equivalency Factors for Tetrodotoxin Analogues Determined with Automated Patch Clamp on Voltage-Gated Sodium Channels in Neuro-2a Cells
Tetrodotoxin (TTX) is a potent marine neurotoxin, responsible for numerous poisoning incidents and some human fatalities. To date, more than 30 TTX analogues have been identified, but their individual toxicities and roles in poisoning remain largely unknown. In this work, the toxicity equivalency factors (TEFs) of five TTX analogues were determined by assessing the blockade of voltage-gated sodium channels in Neuro-2a cells using automated patch clamp (APC). All TTX analogues were less toxic than TTX. The derived TEFs were applied to the individual TTX analogues concentrations measured in pufferfish samples, using liquid chromatography coupled to tandem mass spectrometry (LC–MS/MS). A comparison of these results with those obtained from APC analysis demonstrated that TEFs can be effectively used to translate LC–MS/MS analytical data into meaningful toxicological information. Is calcium deficiency the real cause of bitter pit? A review
Bitter pit is a disorder affecting the appearance of apples. Susceptibility is genetically controlled by both the cultivar and rootstock, with both environmental and horticultural factors affecting its severity and proportional incidence. Symptoms appear more frequently at the calyx end of the fruit and consist of circular necrotic spots, which take on a “corky” appearance visible through the peel. Bitter pit may develop before harvest, or after harvest, reducing the proportions of marketable fruit. In this review, current knowledge of the factors associated with the occurrence of bitter pit in apples is summarized and discussed along with their interactions with Ca uptake and distribution to fruit. This disorder has been previously linked with localized Ca deficiencies in fruit during its development. However, these relationships are not always clear. Even with over a century of research, the precise mechanisms involved in its development are still not fully understood. Additional factors also contribute to bitter pit development, like imbalances of mineral nutrients, low concentration of auxins, high concentration of gibberellins, changes in xylem functionality, or physiological responses to abiotic stress. Bitter pit remains a complex disorder with multiple factors contributing to its development including changes at whole plant and cellular scales. Apple growers must carefully navigate these complex interactions between genetics, environment, and management decisions to minimize bitter pit in susceptible cultivars. Accordingly, management of plant nutrition, fruit crop load, and tree vigor still stands as the most important contribution to reducing bitter pit development. Welfare of sheep and goats during killing for purposes other than slaughter
Sheep and goats of different ages may have to be killed on-farm for purposes other than slaughter (where slaughter is defined as killing for human consumption) either individually (i.e. on-farm killing of unproductive, injured or terminally ill animals) or on a large scale (i.e. depopulation for disease control purposes and for other situations, such as environmental contamination and disaster management) outside the slaughterhouses. The purpose of this opinion was to assess the hazards and welfare consequences associated with the on-farm killing of sheep and goats. The whole killing procedure was divided into Phase 1 (pre-killing) – that included the processes (i) handling and moving the animals to the killing place and (ii) restraint of the animals before application of the killing methods and Phase 2 – that included stunning and killing of the animals. The killing methods for sheep and goats were grouped into three categories: (1) mechanical, (2) electrical and (3) lethal injection. Welfare consequences that sheep and goats may experience during each process were identified (e.g. handling stress, restriction of movements and tissue lesions during restraint) and animal-based measures (ABMs) to assess them were proposed. During application of the killing method, sheep and goats will experience pain and fear if they are ineffectively stunned or if they recover consciousness. ABMs related to the state of consciousness can be used to indirectly assess pain and fear. Flowcharts including ABMs for consciousness specific to each killing method were included in the opinion. Possible welfare hazards were identified for each process, together with their origin and related preventive and corrective measures. Outcome tables linking hazards, welfare consequences, ABMs, origins, preventive and corrective measures were developed for each process.
