Morfološke karakteristike izolata Colletotrichum spp. - prouzrokovača antraknoze
In the morphological studies were included isolates of Colletotrichum spp. originating from pear, apple and sour cherry fruits, as well as reference strains of C. acutatum (CBS 294.67) and C. gloeosporioides (CBS 516.97). The colony characteristics, appearance, shape and size of conidioma, appressoria, and conidia of isolates of Colletotrichum spp. were studied. The four morphological groups of colonies on PDA were observed, confirming the great phenotypic variability of isolates of Colletotrichum spp. Most isolates from apple and pear fruits (KC-6, KC-9, KC-12, JC-5, JC-7, JC-9), formed the first morphological group. The isolates from sour cherry (VC-3, VC-5, VC-7, VC-9) are separated in the fourth morphological group. The size and shape of conidia from these isolates fit the description of C. gloeosporioides. The chromogenic isolates from pear (KC-21, KC-23, KC-82) formed the second morphological group, and in the third morphological group is isolate (JC-4) from apple fruit. The morphology of conidia from these isolates fit the description of C. acutatum. During this research were evident that all isolates formed conidioma - acervuli. The tested cultures of Colletotrichum spp. did not form teleomorpf. Because of outstanding variability, most of the studied characteristics cannot be independently applied as taxonomic criteria. Obtained results showed that, for Colletotrichum species classification, at least two taxonomic criteria should be used: morphological features combined with molecular identification.U morfološka ispitivanja su uključeni izolati Colletotrichum spp. poreklom sa plodova kruške, jabuke i višnje, kao i referentni sojevi C. acutatum (CBS 294.67) i C. gloeosporioides (CBS 516.97). Proučavanjem njihovih morfoloških karakteristika, dobijeni su podaci o izgledu ispitivanih kolonija, odlikama plodonosnog tela, apresorija, veličini i obliku konidija. Na podlozi KDA moguće je razlikovati četiri morfološke grupe kolonija, što potvrđuje fenotipsku varijabilnost proučavanih izolata Colletotrichum spp. Većina izolata sa ploda kruške i jabuke (KC-6, KC-9, KC-12, JC-5, JC-7, JC-9) obrazuje prvu morfološku grupu. Izolati sa ploda višnje (VC-3, VC-5, VC-7, VC-9), su izdvojeni u četvrtu morfološku grupu. Veličina i oblik konidija ovih izolata odgovara opisu vrste C. gloeosporioides. Hromogeni izolati sa kruške (KC-21, KC-23, KC-82), obrazuju drugu morfološku grupu, a u trećoj morfološkoj grupi je izolat sa ploda jabuke (JC-4). Morfologija konidija ovih izolata odgovara opisu vrste C. acutatum. Tokom ovih istraživanja je utvrđeno da svi izolati obrazuju plodonosna tela - acervule. Testirane kulture Colletotrichum spp. ne formiraju teleomorfni stadijum. Zbog izražene varijabilnosti, većina ispitanih osobina ne mogu se primeni kao samostalni taksonomski kriterijum. Na osnovu dobijenih rezultata, za klasifikaciju vrsta roda Colletotrichum, potrebno je koristiti najmanje dva taksonomska kriterijuma: morfološke karakteristike uporedo sa molekularnom identifikacijom
Allele frequency of local maize inbred lines (zea mays L.)
The objective of this study was to screen the genetic divergence of 32 inbred lines in the F4 generation of inbreeding, originating from domestic populations of Eastern Serbia. In order to speed up the selection process, it is important to select genotypes with desirable properties at an early stage. Isoenzymes, as a direct product of genes, can be used as genetic markers in the selection process. Different numbers and distribution of certain alleles in inbred lines of maize indicate their mutual distance and genetic divergence. Differences in the investigated inbred lines can be shown by the presence or absence of specific alleles in loci and their frequency. The highest number of alleles (14) was found in line 5, 14 inbred lines had 13 alleles each, while line 9 had 12 alleles. Three lines each had 10 and 11 alleles, while one line had nine alleles (line 30). Only eight lines were homozygous for all loci (3, 7, 10, 21, 24, 26, 28 and 30). In accordance with allele frequencies, all lines were allocated to three groups with a different number of lines. The first and second groups joined at a distance of 4.2 and formed a new group connected to the third group at the furthest distance of 7. The third group of lines was clearly separated from the first two groups. Lines 26, 28 and 32 can be viewed as independent genotypes, joined to the third group at a distance of 2.25. These results permit a clearer evaluation of the lines during the process of selection and, together with other desirable traits, faster choice of material for further selection
Effect of Cobalt Application on Seed Production in Red Clover (Trifolium pratense L.)
A field experiment involving red clover varieties K-39, K-17, Una and Viola was established on an acidic (pH(H2O) 4.8) soil to evaluate the effects of foliar application of cobalt (a beneficial nutrient for efficient nitrogen fixation) on seed yield, and on seed yield components. The foliar spray of the crop was carried out using cobalt nitrate [Co(NO3)(2)] with the treatments: one application at the intensive growth phase during the first growth, while two others during the second growth within the second year of cultivation. Seed yield and yield components were recorded from the second growth within the second year of the study. Regardless of foliar cobalt application, the varieties produced a significantly higher seed yield in 2011, when the rainfall received from the onset of flowering until seed maturation was recorded as lower than that in 2010. The foliar treatment with cobalt was in general accompanied by a positive effect on seed yield and seed yield components in all the varieties. As compared with control, a significant increase in seed yield, in the cobalt applied treatment, was obtained only in Viola, mostly due to the significant increase in flower number i.e. seed number per inflorescence. The more favorable response of Viola to foliar cobalt application may have been attributed to a greater percentage of foliage cover during the treatment, as compared with the other varieties. This suggests that foliar cobalt treatment in future studies should be performed at the early stages of development i.e. during intensive growth throughout the first and second cuts, in order to stimulate nodulation and have greater nitrogen fixation in a needed timely manner
First Report of Alder Yellows Phytoplasma Associated with Common Alder (Alnus glutinosa) in the Republic of Macedonia.
Biological control of Rhamnus cathartica: is it feasible? A review of work done in 2002-2012
Rhamnus cathartica (common buckthorn) is a shrub (or small tree) of Eurasian origin, which has become invasive in North America. Internal feeders and sap suckers were prioritized for biological control from over 30 specialized insects identified from the target plant in its native European range. Five leaf-feeding moths were also considered for further investigations. Field observations and preliminary host range tests with the stem-boring beetle Oberea pedemontana, the root-boring moth Synanthedon stomoxiformis, the shoot-tip-boring moth Sorhagenia janiszewskae and the leaf-feeding moths Ancylis apicella, A.unculana, Triphosa dubitata, Philereme transversata and P.vetulata confirmed that all of these species were lacking host specificity in no-choice conditions. Choice oviposition tests carried out with most of the prioritized species to assess their ecological host range yielded unreliable results. Three psyllids, Trichochermes walkeri, Cacopsylla rhamnicolla and Trioza rhamni are promising in terms of host specificity, but are infected with the plant disease Candidatus Phytoplasma rhamni'. Fruit- or seed-feeding insects may present the best potential for biological control of buckthorn in directly reducing seed set and thus seedling establishment. However, it was not possible to obtain adult fruiting trees of native North American Rhamnus species for testing. It is concluded that there are no promising arthropod agents based on what is known to date. Pathogens could offer new opportunities for biological control of R.cathartica in North America
The "potato road" and biogeographic history of potato cyst nematode populations from different continents
The general opinion about the introduction of potato in Europe is the one regarding the direction from South America to Spain and subsequent distribution to other continents. Some historical data point out an alternative road. The potato spread from its place of origin to other continents in the light of parasite-host relationship, relying on nematode molecular data, is discussed in the present work. Biogeographic history of potato cyst nematode populations from different continents is in congruence with historical records
Fusarium spp. - patogeni semena nevena (Calendula officinalis L.) u Srbiji
During 2013, 14 samples of infected seed grown calendula were collected from two commercially grown crops in the localities in Pančevo and Plandište and analyzed for the presence of plant pathogenic fungi. In all samples of seeds, weaker germination and a significant seed infection ranging from 2-8%, with phytopathogenic fungi of the genus Fusarium, was found. From the infected calendula seed, monosporial cultures, based on morphology, were identified as Fusarium verticillioides and Fusarium cf. incarnatum, and their pathogenicity proved on artificially inoculated calendula seedlings. Molecular detection was performed by PCR and amplification of the TEF-1α protein gene. Further studies will include molecular identification of this isolate to the species level, a definitive taxonomic determination of the Serbian isolate and a comparison with other isolates and species of Fusarium genus infecting calendula worldwide.Tokom 2013. godine, na dva lokaliteta komercijalne proizvodnje nevena u Pančevu i Plandištu, prikupljeno je 14 uzoraka zaraženog semena gajenog nevena i analizirano na prisustvo fitopatogenih gljiva. U svim uzorcima semena, ustanovljena je slabija klijavost i značajan stepen zaraze fitopatogenim gljivama iz roda Fusarium, 2-8%. Iz zaraženog semena izolovane su monosporijalne kulture, čija je patogenost potvrđena pojavom simptoma na veštački inokulisanim klijancima nevena, a na osnovu morfoloških svojstava identifikovane su kao Fusarium verticillioides i Fusarium cf. incarnatum. Molekularna detekcija obavljena je primenom PCR i amplifikacije proteinskog gena TEF-1α. Dalja istraživanja obuhvatiće primenu molekularne identifikacije, do nivoa vrste uz određivanje tačnog taksonomskog mesta izolata iz Srbije patogenih za neven poređenjem sa drugim izolatima i vrstama roda Fusarium spp. u svetu
Uticaj agrotehnike na osobine semena italijanskog ljulja pri različitim dužinama skladištenja
Italian ryegrass seed crop was established in 2007 with two sowing densities (D1 = 60 cm row spacing and 5 kg ha-1 seeding rate; D2 = 20 cm row spacing and 20 kg ha-1 seeding rate) and using two spring nitrogen rates (0 and 150 kg N ha-1). Seed germination and thousand seed weight (TSW) of Italian ryegrass was observed in first production year. After harvest in June, seeds were stored under standard storage conditions and sampled 90 days after harvest (DAH), and then 2000 DAH. At 90 DAH, seeds were tested for TSW, as well as germination energy and total germination percentage at incubation temperatures of 10, 15, 20 and 25°C. Ryegrass seeds had the best germination energy 90 DAH at 20°C and maximum total germination at 15°C, which implies that early autumn (September-October) is proper sowing period for freshly harvested seeds of Italian ryegrass. Italian ryegrass seeds could maintain satisfactory germination energy (59.3%) and total germination (77.3%) up to 2000 DAH. High seed quality was obtained and applied treatments did not change seed quality significantly unlike storage period which had considerable influence on seed quality. The data can serve for the determination of a proper storage duration management between harvest and sowing of the tested species under ambient conditions of Serbia.Setva semenskog useva italijanskog ljulja obavljena je u 2007 godini u dve gustine setve (D1=60 cm međuredno i 5 kg ha-1 setvena norma; D2=20 cm međuredno i 20 kg ha-1 setvena norma) i primenom dve količine azota u prihrani (0 and 150 kg ha-1). Posmatran je klijavost i masa 1000 semena italijanskog ljulja u prvoj proizvodnoj godini. Posle junske žetve, seme je skladišteno u standardnim skladišnim uslovima i uzorkovano 90 dana posle žetve (DPŽ), a potom 2000 DPŽ. Posle 90 DPŽ seme je ispitivano na masu 1000 semena, energiju klijanja i ukupnu klijavost na temperaturama klijanja od 10, 15, 20 i 25°C. Seme ljulja je imalo najbolju životnu sposobnost 90 DPŽ na 20°C i maksimalnu ukupnu klijavost na 15°C, što sugeriše da je rana jesen (septembar-oktobar) odgovarajući period za setvu sveže požnjevenog semena italijanskog ljulja. Seme italijanskog ljulja može zadržati zadovoljavajuću energiju klijanja (59,3%) i klijavost (77,3%) i 2000 DPŽ. Dobijeno je kvalitetno seme i primenjeni tretmani pri gajenju semenskog useva nisu menjali značajno kvalitet semena, za razliku od dužine perioda skladištenja, koje je imalo značajan uticaj na kvalitet semena. Podaci mogu poslužiti za određivanje dužine perioda skladištenja semena između žetve i setve italijanskog ljulja u uslovima Srbije
Primena kukuruzne prekrupe pri siliranju lucerke, ježevice i njihove smeše
One of the solutions for the economical use of alfalfa and cocksfoot is making the silage. However, these plant species are difficult to be conserved without appropriate additives. The aim of this study was to determine the impact addition of cornmeal on the quality of silage of alfalfa (Medicago sativa L.) cv. Kruševačka 22, cocksfoot (Dactilys glomerata L.) cv. Kruševačka 40 and their mixture (50% alfalfa, 50% cocksfoot). Mowing both species was carried out at the stage when 1/3 to 1/5 of the plants were in flowering in alfalfa or in stage appearance of inflorescence with cocksfoot. Corn grain is ground at the mill with a sieve ø 2 mm, then mixed with chopped fodder of alfalfa, cocksfoot and the mixture in the following amount: 6% (treatment A2), 3% (treatment A1), and without cornmeal addition (control A0). After six months was determined silage quality. Cornmeal addition, according to the basic indicators of the quality of silage had a significant impact on changing the quality silage of alfalfa, cocksfoot and their mixtures. It is evident that the addition cornmeal addition a significant impact on increasing the energy value (primarily the content inside BEM) of silage in all studied treatments.Jedno od rešenja za ekonomično korišćenje lucerke i ježevice je spravljanje silaže. Međutim ove biljne vrste se teško mogu konzervisati bez odgovarajućih dodataka. Cilj ovih ispitivanja bio je određivanje uticaja kukuruzne prekrupe na kvalitet silaže od lucerke, ježevice i njihove smeše (50% lucerka, 50% ježevica). Ispitivanje je sprovedeno na krmi: lucerke (Medicago sativa L.) sorte Kruševačka 22, ježevice (Dactilys glomerata L.) sorte Kruševačka 40 i njihove smeše (50% lucerka i 50% ježevica). Košenje obe vrste obavljeno je u fazi kada je 1/3 do 1/5 biljaka bilo u cvetu kod lucerke odnosno u klasanju kod ježevice. Kukuruzno zrno je mleveno na mlinu sa sitima ø 2 mm, zatim je mešano sa seckanom krmom lucerke, ježevice i smeše u količini: 6% (tretman A2), 3% (tretman A1), i bez prekrupe (kontrola A0). Nakon šest meseci utvrđen je kvalitet silaže. Dodatak kukuruzne prekrupe je prema osnovnim pokazateljima kvaliteta silaža imao značajan uticaj na promenu kvaliteta silaže lucerke ježevice i smeše. Evidentno je da dodavanje kukuruzne prekrupe značajno utiče na povećanje energetske vrednosti (pre svega sadržaja BEM) u silaži svih ispitivanih vrsta
Geometric morphometric study of geographic and host-related variability in Aceria spp. (Acari: Eriophyoidea) inhabiting Cirsium spp. (Asteraceae)
The russet mite, Aceria anthocoptes (Nalepa), is the only eriophyoid that has been recorded on Cirsium arvense (L.) Scop. It has been noted in several European countries and recently in the USA. In this study we explored the geographic and host-related variability of Aceria spp. inhabiting different Cirsium spp. We applied landmark-based geometric morphometric methods to study morphological variability of three body regions (ventral, coxigenital and prodorsal) of 13 Aceria spp. populations inhabiting five Cirsium spp. in Serbia (Europe) and four Cirsium spp. in Colorado (North America). Analyses of size and shape variation revealed statistically significant differences between Aceria spp. living on European native and North American native Cirsium spp., as well as between A. anthocoptes s.s. inhabiting European C. arvense and North American C. arvense. The coxigenital region was the most informative when considering inter-population shape differences. European Aceria spp. dwelling on Cirsium spp., including A. anthocoptes s.s. from C. arvense, are characterized by higher inter-population size and shape variability than their North American counterparts. This finding supports a Eurasian origin of A. anthocoptes, presumed to consist of a complex of cryptic taxa probably coevolved with host plants in the native environment. Morphological similarity among Aceria spp. inhabiting North American native Cirsium spp. may indicate that speciation of A. anthocoptes started relatively soon after the host shift to plants different from C. arvense in the invaded region
