Ambient Awareness : Experiencing Always-On Displays in the Life of PV Households
The adoption of photovoltaic (PV) panels, electric vehicles (EVs), and dynamic electricity pricing is transforming households into active "prosumers" who generate, consume, and sell electricity. This shift, driven by rising costs and environmental concerns, requires new technologies to help households manage their production and consumption. Electricity’s invisibility adds complexity, necessitating interfaces that make energy use and generation comprehensible. This paper presents the Always-On In-Home Display (AOIHD), a technology probe designed for prosumer households to navigate the dynamics of this production and consumption - balancing periods of solar abundance and grid reliance, by making energy data persistently and collectively accessible within the household. Adopting a practice theory lens, we explore how the AOIHD was experienced in daily life over a four-year autobiographical study and through deployments in other Swedish households. Our findings highlight four experiential qualities—Learning, Triggering, Including, and Troubling—that illustrate how the display supports the domestication of energy feedback technologies in prosumer contexts. Flow Dynamics and Thrombus Formation in Extracorporeal Membrane Oxygenation : A Combined Computational and Experimental Study
In cases of severe cardiac and/or respiratory failure, extracorporeal membrane oxygenation (ECMO) can be used as a bridge to organ recovery or transplantation. In ECMO, blood is drained through a venous drainage cannula by a pump, pushed through a membrane lung for gas exchange, and returned to the patient through a return cannula. Along this path blood passes through tubing and several connectors. These components expose blood components to elevated shear rates, highly unsteady flow fields, prolonged residence times, and artificial surfaces, increasing the risk of hemolysis, bleeding, and thrombosis. This work investigates the flow structures and stresses that arise in ECMO circuit components and connects them to mechanisms for blood damage. It also provides a methodology where the thrombus morphology and composition can be assessed with scanning electron microscopy (SEM) and ultra small angle X-ray scattering (USAXS) to the local flow field. By using large eddy simulations (LES) complemented by Reynolds averaged Navier Stokes (RANS) modeling, flow phenomena were evaluated under clinically relevant operating conditions and compared with experimental observations. Results revealed that the size and intensity of recirculation zones and other flow structures were highly dependent on operating conditions, with inlet recirculation and larger rotating flow cells blocking the main flow at low inlet flow rates in the DP3 pump (Xenios AG,Heilbronn, Germany). The low-weight DP3 impeller exhibited wobbling, that could potentially increase both hemolysis and plastic spallation. Cavities and protrusions were found to promote stagnation, shear layers, with recirculation volumes growing with increased flow rates. Shear rate analysis further identified elongational shear in several locations, including the pump inlet, blade wakes and pump outlet. These regions pose a risk for von Willebrand factor (vWf) unfolding followed by platelet activation and consequently thrombus growth. Overall, the findings emphasized that both operational settings and circuit design strongly influenced the formation of damaging flow conditions in the ECMO circuit. Linking flow features to thrombus morphology and known mechanisms for blood damage provides a foundation for improved models for thrombosis, refined clinical guidelines, and helps with future device design.Vid kraftigt nedsatt hjärt- och/eller lungfunktion kan extracorporal membranoxygenering (ECMO) användas i väntan på att organen återhämtar sig eller tills dess att transplantation kan utföras. I ECMO dras blodet ut genom en venös dränagekanyl med hjälp av en pump som sedan trycker blodet genom en membranlunga för gasutbyte. Därefter återförs det syresatta blodet till patienten via en returkanyl. På vägen genom systemet passerar blodet också slangar och ett flertal konnektorer. Dessa komponenter utsätter blodet för höga skjuvkrafter, kraftigt instabila flöden, artificiella ytskikt och risk för att blodkomponenter fastnar i flödesstrukturerna under en längre tid vilket kan leda till både hemolys, blödning och trombos. Det här arbetet utvärderar flödesstrukturer och skjuvkrafter som uppstår i ECMO systemets komponenter och kopplar dem till mekanismer för skador på blodets beståndsdelar. Arbetet ger också förslag på en metod för att utvärdera trombers morfologi och komposition med svepelektronmikroskopi och småvinkelspridning av röntgenstrålar (small angle X-ray scattering) och kopplar det till det lokala flödesfältet. Genom att använda large eddy simulations (LES) och Reynolds averaged Navier Stokes (RANS) modellering kunde olika flödesfenomen utvärderas med kliniskt relevanta testvillkor och jämföras med experimentella observationer. Resultaten visade att storlek och intensitet hos recirkulationszoner och andra flödesstrukturer var beroende av testvillkoren, där kraftigare inloppsrecirkulation och större roterande flödesceller blockerade flödet. Detta var särskilt tydligt vid lägre inloppsflöden i DP3-pumpen. Den lätta impellern hos DP3-pumpen konstaterades wobbla, något som kan ge upphov till både hemolys och utfällning av mikroplaster. Hålrum och utbuktande delar påvisades ge upphov till flödesstagnation, skjuvlager och recirkulationsvolymer vilka växte med ökad flödeshastighet. Genom att undersöka skjuvhastigheter kunde regioner med elongerande skjuvegenskaper konstateras i pumpinloppet, bladvakerna och pumputloppet. I dessa regioner är risken större för att von Willebrandfaktorn (vWf) ska rullas ut och aktivera blodplättarna runt sig vilket följaktligen kan leda till trombos. Sammanfattningsvis visar resultaten att testvillkor och komponentdesign hos ECMO-komponenter kraftigt påverkar skapandet av flödesvillkor, vilka kan skada och aktivera blodkomponenter. Genom att koppla flödesstrukturer till trombmorfologin och mekanismer som är kända för att skada blodkomponenter skapas i detta arbetet en grund för bättre trombosmodeller, tydligare kliniska riktlinjer och hjälp för framtida komponentdesign.QC 251015</p
Parallelized robust distributed model predictive control in the presence of coupled state constraints
In this paper, we present a robust distributed model predictive control (DMPC) scheme for dynamically decoupled nonlinear systems which are subject to state constraints, coupled state constraints and input constraints. In the proposed control scheme, all subsystems solve their local optimization problem in parallel and neighbor-to-neighbor communication suffices. The approach relies on consistency constraints which define a neighborhood around each subsystem's reference trajectory where the state of the subsystem is guaranteed to stay in. Contrary to related approaches, the reference trajectories are improved consecutively. In order to ensure the controller's robustness against bounded uncertainties, we employ tubes. The presented approach can be considered as a time-efficient alternative to the well-established sequential DMPC. In the end, we briefly comment on an iterative extension. The effectiveness of the proposed DMPC scheme is demonstrated with simulations, and its performance is compared to other DMPC schemes.QC 20241010</p
Harnessing cell aggregates for enhanced adeno‐associated virus manufacturing : Cultivation strategies and scale‐up considerations
The possibility to produce recombinant adeno-associated virus (rAAV) by adherent HEK293T cells was studied in a stirred tank bioreactor (STR) culture of cell aggregates. A proof-of-concept of rAAV production was successfully demonstrated in a process where single cells were first expanded, then cell aggregates were formed by dilution into a different medium 1 day before triple plasmid transfection was conducted. An alternative approach for the STR inoculation using a seed taken from a high cell density perfusion (HCDP) culture was also investigated. It was, however, found that the spent medium of the HCDP inhibited the transfection of HEK293T cell aggregates, which was confirmed when testing with single-cell suspension culture. The formation of aggregates in shaken multi-well plates was also investigated to develop a screening system using the average power input as a scale-down criterion, which revealed that cell aggregates could be generated in 12-well plates, however with a larger size than in a STR. Taking into account the reported higher rAAV production of adherent cells in comparison with single cells for triple-plasmid transfection, HEK293T cell aggregates can possibly surpass single-cell suspension in space–time rAAV yield. The formation of HEK293T cell aggregates in a STR system offers a promising approach for scaling up and intensifying rAAV production by triple-plasmid transfection, in comparison with traditional 2D scale-up methods
Laboratory Soft X-Ray Microscopy for Biological Imaging
Soft x-ray microscopy within the water window is a powerful technique for high-resolution biological imaging due to its capability to image whole, intact cells (approximately 10 μm thick) in their near-native cellular environment. The short wavelength of water-window radiation (λ = 2.3 − 4.4 nm, E = 284−540 eV) used in this imaging technique provides high natural contrast for cellular imaging due to the significant difference in soft x-ray attenuation lengths between organic materials, such as proteins and lipids (i.e., carbon), and water (i.e., oxygen). In addition to the high imaging contrast, the high penetration of soft x-rays eliminates the need for laborious sample preparation, including sectioning, chemical fixation, heavy-metal staining, and fluorescence labeling. The majority of soft x-ray microscopes are operated using synchrotron radiation sources, as they require x-ray sources with high spectral brightness, which limits accessibility. To complement these synchrotron-based instruments, we develop a laboratory-based soft x-ray microscope as alternative system for biological imaging. Motivated by this background, this thesis presents the development of laboratory soft x-ray microscopy focused on improving image resolution and optimizing sample preparation. The resolution has been improved to 25 nm(half-period) through vibration analysis and mitigation. Sample preparation optimization was achieved by controlling the ice thickness during devitrification process, applied to both manual plunge-freezing and automated systems, allowing for the preservation of cellular structures and improved image quality. These developments have enabled the establishment of methodology for investigating nanoparticle interactions in-vitro and in-vivo, relying solely on x-ray imaging. These advancements have enabled the investigation of uptake and dynamics of nanoparticles in organelles. Moreover, the applications extend beyond bio-nano interactions; they have also facilitated quantitative studies in viral infections of giant DNA viruses.Mjukröntgenmikroskopi inom vattenfönstret är en kraftfull teknik för hög-upplöst biologisk avbildning tack vare dess förmåga att avbilda hela, intakta celler (ungefär 10 μm tjocka) i deras nära naturliga cellulära miljö. Den korta våglängden hos strålning i vattenfönstret (λ = 2.3–4.4 nm, E = 284–540 eV) som används i denna avbildningsteknik ger hög naturlig kontrast för cellulär avbildning, tack vare den betydande skillnaden i mjukröntgens absorptionslängd mellan organiska material, såsom proteiner och lipider (dvs. kol), och vatten (dvs. syre). Förutom den höga bildkontrasten eliminerar den höga penetrationen av mjukröntgen behovet av tidskrävande provberedning, inklusive sektionering, kemisk fixering, tungmetallfärgning och fluorescensmärkning. De flesta mjukröntgenmikroskop finns hos synkrotronstrålningskällor eftersom de kräver röntgenkällor med hög spektral ljusstyrka, vilket begränsar tillgängligheten. För att komplettera dessa synkrotronbaserade instrument utvecklar vi ett laboratoriebaserat mjukröntgenmikros-kop som ett alternativt system för biologisk avbildning. Motiverad av denna bakgrund presenterar denna avhandling utvecklingen av laboratoriebaserad mjukröntgenmikroskopi med fokus på att förbät-tra bildupplösningen och optimera provberedningen. Upplösningen har förbättrats till 25 nm (halvperiod) genom vibrationsanalys och dämpning. Optimering av provberedning uppnåddes genom att kontrollera istjockleken under vitrifieringsprocessen, tillämpad både vid manuell snabbfrysning och automatiserade system, vilket möjliggjorde bevarandet av cellstrukturer och förbättrad bildkvalitet. Dessa utvecklingar har möjliggjort etableringen av en metodik för att undersöka nanopartikelinteraktioner in vitro och in vivo, enbart med hjälp av röntgenavbildning. Orientation selectivity properties for the affine Gaussian derivative and the affine Gabor models for visual receptive fields
This paper presents an in-depth theoretical analysis of the orientation selectivity properties of simple cells and complex cells, that can be well modelled by the generalized Gaussian derivative model for visual receptive fields, with the purely spatial component of the receptive fields determined by oriented affine Gaussian derivatives for different orders of spatial differentiation. A detailed mathematical analysis is presented for the three different cases of either: (i) purely spatial receptive fields, (ii) space-time separable spatio-temporal receptive fields and (iii) velocity-adapted spatio-temporal receptive fields. Closed-form theoretical expressions for the orientation selectivity curves for idealized models of simple and complex cells are derived for all these main cases, and it is shown that the orientation selectivity of the receptive fields becomes more narrow, as a scale parameter ratio κ, defined as the ratio between the scale parameters in the directions perpendicular to vs. parallel with the preferred orientation of the receptive field, increases. It is also shown that the orientation selectivity becomes more narrow with increasing order of spatial differentiation in the underlying affine Gaussian derivative operators over the spatial domain. A corresponding theoretical orientation selectivity analysis is also presented for purely spatial receptive fields according to an affine Gabor model, showing that: (i) the orientation selectivity becomes more narrow when making the receptive fields wider in the direction perpendicular to the preferred orientation of the receptive field; while (ii) an additional degree of freedom in the affine Gabor model does, however, also strongly affect the orientation selectivity properties.QC 20250318Covariant and invariant deep network
Fastighetsvetenskaplig forskning vid KTH : Ett supplement, åren 2021-2024
Under 2021 sammanställdes och publicerades en forskningsöversikt (Ekbäck & Mattsson, 2021), där målsättningen var att inventera, dokumentera och systematisera den fastighetstekniska och fastighetsrättsliga forskning som bedrivits och publicerats vid avdelningen för fastighetsvetenskap (och motsvarande äldre organisatoriska enheter), institutionen för fastigheter och byggande på Kungl. Tekniska Högskolan. Den tidsmässiga omfattningen för översikten spänner från 1932 – då den första professuren inom det fastighetstekniska ämnesområdet inrättades vid KTH – fram till hösten 2021. Syftet med detta supplement är att bygga vidare på den tidigare dokumentationen och de analyser som då genomfördes. Supplementet är emellertid inte avsett att läsas fristående, utan för en helhetsbild förutsätts den tidigare rapporten ha studerats. Som underlag för detta supplement har, liksom i den tidigare publicerade forskningsöversikten, en litteratursökning genomförts. Sökmetoden ansluter i stort till metodiken i den tidigare rapporten, givetvis under betydligt enklare och mer begränsade former med hänsyn till den kortare tidsperioden och tillgängliga digitala publikationsregister. Den fullständiga publikationslistan finns som bilaga sist i rapporten. Av den tematiska analysen framgår att inriktningen mot Nya rättighets- och upplåtelseformer i dagsläget kan sägas dominera de fastighetsvetenskapliga forskningsinriktningarna. I de senare årens forskning utkristalliseras även ett nytt tema: Digitalisering av samhällsbyggnadsprocessen. Exploring Mid-IR FSO Communications With Unipolar Quantum Optoelectronics
Free space optical (FSO) communication is considered a critical part of future ICT infrastructure, particularly in non-terrestrial communication segments. In this context, the ability to achieve fast and reliable FSO propagation through long-distance atmospheric channels is the most important factor in choosing technological solutions. One property of optics directly related to this factor is the choice of wavelength. It has been identified that the mid-infrared (mid-IR) regime, which includes two atmospheric transmission windows—the mid-wave IR (MWIR, 3-5 μm) and the long-wave IR (LWIR, 8-12 μm)—can potentially offer a promising solution for achieving such performance. Additionally, viable semiconductor sources and detectors that support high-speed and efficient signal transmission are also considered critical to generating sufficient critical mass to advance the application of mid-IR FSO. Unipolar quantum optoelectronics, including quantum cascade lasers (QCL), Stark modulators, quantum cascade detectors (QCD), and quantum-well IR photodetectors (QWIP), among other components, emerge as potential candidates to build such FSO subsystems and systems. We present our recent efforts in conducting subsystem and system-level studies with different variants of these unipolar quantum optoelectronics and demonstrate the potential for feasible transmitter and receiver performance in a laboratory environment. We also discuss the key challenges and considerations of such technologies towards practical development. Capturing the Shape and Pose of Horses in 3D
Animals play a significant role in the Earth's ecology and have lived alongside humans throughout history. Studying and understanding their movements and behaviors is important for advancing scientific knowledge and benefiting practical applications. In this thesis, we focus specifically on horses, which are key subjects in both computer vision and biological research due to their strength in speed and unique locomotion systems. Traditional systems for capturing horse motion often rely on attaching sensors or markers to the horse's body. These systems, however, are often limited to constrained environments and difficult to use in natural, unconstrained settings. In contrast, capturing horses using standard video cameras, where horses are observed in their natural environments, presents a more practical solution. However, capturing horses in 3D, specifically the 3D shape and pose, from 2D images is a highly challenging problem due to the ambiguity with only 2D data. To address these challenges, we propose model-based methods to capture the 3D shape and pose of horses from monocular images or videos. We start by presenting hSMAL, a horse-specific 3D parameterized model, capable of expressing diverse horse shapes, which is learned from 3D scan data. We also demonstrate the practical utility of this model in lameness detection, a critical veterinary task for assessing the well-being of horses. Additionally, we present a comprehensive horse motion dataset, collecting data from horses of varying shapes and performing diverse movements, using dense motion capture markers. This motion capture data allows us to animate hSMAL with real horse movements, providing details about how horses move and also tackling the common issue of limited data in animal research. Building on the proposed model and the dataset, we develop data-driven regression methods, to capture horses in 3D from monocular images and videos in an end-to-end manner. First, we integrate multimodal data, combining video clips and audio. Our findings show that incorporating audio enhances the robustness of the method, especially in situations of visual ambiguity and occlusion. Second, we integrate vision foundation models and disentanglement learning with an on-the-fly synthetic data generation pipeline. The pipeline allows the creation of paired data during network training, facilitating the learning of disentangled feature spaces. Together, these approaches enhance the generalization and adaptability of the method, improving performance on images from various domains and other four-legged animals. Through experiments on both our own collected datasets and public datasets, we demonstrate the effectiveness of the proposed methods in advancing horse-specific capture from monocular images and videos. This thesis contributes methodologies for capturing horses from standard video cameras, specifically focusing on the 3D shape and pose, opening new possibilities for animal motion capture and analysis.Djur spelar en betydande roll i jordens ekologi och har levt tillsammans med människor genom historien. Att studera och förstå deras rörelser och beteenden är viktigt för att främja vetenskaplig kunskap och gynna praktiska tillämpningar. I detta examensarbete fokuserar vi specifikt på hästar, som är nyckelämnen inom både datorseende och biologisk forskning på grund av deras styrka i hastighet och unika rörelsesystem. Traditionella system för att fånga hästens rörelser förlitar sig ofta på att fästa sensorer eller markörer på hästens kropp. Dessa system är dock ofta begränsade till begränsade miljöer och svåra att använda i naturliga, oinskränkta miljöer. Att fånga hästar med vanliga videokameror, där hästar observeras i sina naturliga miljöer, är däremot en mer praktisk lösning. Men att fånga hästar i 3D, särskilt 3D-formen och posituren, från 2D-bilder är ett mycket utmanande problem på grund av tvetydigheten med endast 2D-data. För att möta dessa utmaningar föreslår vi modellbaserade metoder för att fånga hästars 3D-form och ställning från monokulära bilder eller videor. Vi börjar med att presentera hSMAL, en hästspecifik 3D-parameteriserad modell, kapabel att uttrycka olika hästformer, som lärs från 3D-skanningsdata. Vi visar också den praktiska användbarheten av denna modell för att upptäcka hälta, en viktig veterinäruppgift för att bedöma hästars välbefinnande. Dessutom presenterar vi en omfattande datauppsättning för häströrelser, som samlar in data från hästar med olika former och utför olika rörelser med hjälp av markörer för täta rörelsefångst. Denna motion capture-data tillåter oss att animera hSMAL med riktiga häströrelser, tillhandahålla detaljer om hur hästar rör sig och även ta itu med det vanliga problemet med begränsad data inom djurforskning. Med utgångspunkt i den föreslagna modellen och datamängden utvecklar vi datadrivna regressionsmetoder för att fånga hästar i 3D från monokulära bilder och videor på ett heltäckande sätt. Först integrerar vi multimodal data, kombinerar videoklipp och ljud. Våra resultat visar att inkorporering av ljud ökar robustheten i metoden, särskilt i situationer med visuell tvetydighet och ocklusion. För det andra integrerar vi grundmodeller för vision och inlärning av disentanglement med en pipeline för generering av syntetisk data i farten. Pipelinen gör det möjligt att skapa parad data under nätverksträning, vilket underlättar inlärningen av disentangled funktionsutrymmen. Tillsammans förbättrar dessa tillvägagångssätt generaliseringen och anpassningsförmågan hos metoden, vilket förbättrar prestandan på bilder från olika domäner och andra fyrbenta djur. Genom experiment på både våra egna insamlade datamängder och offentliga dataset visar vi effektiviteten hos de föreslagna metoderna för att främja hästspecifik fångst från monokulära bilder och videor.
