Preparation and Characterization of Large Unilamellar Vesicles Mixed with Trimethylchitosan (TMC): The Effect of Polyelectrolyte Concentration
Background: The effect of different concentrations of the absorption enhancer trimethylchitosan (TMC) to the physicochemical properties of large unilamellar vesicles (LUV) comprised of L-a-Phospahtidylcholine (PC) were investigated in the current study. Methods: The degree of quartenization (DQ) of trimethylchitosan was assessed with nuclear magnetic resonance (H NMR). The vesicles were characterized by means of dynamic light scattering (DLS), ζ-potential, differential scanning calorimetry (DSC) and contact angle goniometry (CAG) measurements. Results: The data showed that the surface charge of the PC liposomes was significantly altered as a function of the TMC concentration, giving evidence of presence of the polyelectrolyte to the liposome’s membrane. Varying the concentration of TMC affected the phase transition temperature (T) of the lipid, verifying the miscibility of the polyelectrolyte with the lipid bilayer. The association of the polymer with the liposomes was related to the amount of the polyelectrolyte present, reflecting changes to the wettability of the dispersion as measured by CAG. Conclusions: The results demonstrated that presence of TMC significantly modified the physical properties of liposomes. Such systems might have a potential use for mucosal delivery (e.g. nasal route of administration)
Chitosan derived formulations and EmzaloidTM technology for mucosal vaccination against diphtheria : nasal efficacy in mice

(Pharmaceutics))--North-West University, Potchefstroom Campus, 2005.Previous studies have demonstrated that chitosan and its derivative, N-trimethyl chitosan chloride (TMC) are effective and safe absorption enhancers to improve mucosal delivery of macromolecular drugs including vaccines. Furthermore, chitosan and TMC can easily form microparticles and nanoparticles, which have the ability to encapsulate large amounts of antigens. Emzaloid™ technology has proven in the past to be an effective delivery system for numerous drugs. Emzaloids can entrap, transport and deliver large amounts of drugs including vaccines. In this study, the ability of chitosan microparticles and nanoparticles, TMC microparticles as well as micrometer and nanometer range Emzaloids to enhance both the systemic and mucosal (local) immune response against diphtheria toxoid (DT) after nasal administration in mice was investigated. The above mentioned formulations were prepared and characterised according to size and morphology. DT was then associated to the chitosan microparticles and nanoparticles as
well as TMC microparticles to determine the antigen loading and release. It was found
that the loading efficacy of the formulations was 88.9 %, 27.74 % and 63.1 % respectively, and the loading capacity of the formulations was 25.7 %, 8.03 % and 18.3 %. DT loaded and unloaded (empty) chitosan microparticles and nanoparticles, TMC
microparticles, micrometer and nanometer range Emzaloids as well as DT in phosphate
buffered saline (PBS) were administered nasally to mice. Mice were also vaccinated
subcutaneous with DT associated to alum as a positive control. All mice were vaccinated
on three consecutive days in week 1 and boosted in week 3. Sera was analysed for anti-DT IgG and nasal lavages were analysed for anti-DT IgA using an enzyme linked
imrnunosorbent assay (ELISA). In the study conducted to determine the systemic (IgG) and local (IgA) immune responses it was seen that DT associated to all the experimental formulations produced a
systemic immune response. The said formulations produced a significantly higher
systemic immune response when compared to the formulation of DT in PBS. Furthermore, the mice vaccinated with DT associated to the TMC formulations showed a much higher systemic immune response than the mice that were vaccinated
subcutaneously with DT associated to alum, whereas the other formulations produced
systemic immune responses that were comparable to that of DT associated to alum. It was also found that DT associated to the experimental formulations produced a local immune response, however only DT associated to TMC microparticles produced a consistent local immune response. It can be concluded from the in vivo experiments that the TMC formulations, moreover, the TMC microparticles is the most effective and promising formulation for the nasal delivery of vaccines.Master
The effect of pharmaceutical excipients on rifampicin release from chitosan beads
Thesis (M.Sc. (Pharmaceutics))--North-West University, Potchefstroom Campus, 2005.Controlled release systems aim at achieving a predictable and reproducible drug release over a desired time period. These systems allow reduced dosing frequency, constant drug levels in the blood, increased patient compliance and decreased adverse effects. In a recent study, Chitosan beads, containing N-trimethyl Chitosan chloride, have shown a potential in the delivery of rifampicin. However, because of inadequate amounts of rifampicin released over 24 hours, incorporation of other pharmaceutical excipients to increase the swelling behaviour of the beads to improve drug release, was considered in this study. Chitosan beads were prepared through ionotropic gelation with tripolyphosphate (TPP) as a crosslinking agent. To increase the porosity if the Chitosan beads Explotab®, Ac-Di-Sol® and vitamin C were added individually to Chitosan solutions at concentrations of 0.1, 0.25 and 0.5 % w/v before adding the mixture to the TPP solution. Swelling and morphology studies were used in the evaluation of the different formulations. The swelling and morphology results were then used to select a set of combination and concentrations of two excipients sand then prepare and characterise beads containing two combinations. The combination formulations and formulations containing single excipients were then loaded with rifampicin. Pure chitosan beads exhibited a higher drug loading capacity (67.49 %) compared to the lowest loading capacity of 41.61 % exhibited by chitosan beads containing a combination of Explotab®, Ac-Di-Sol®. For all the other formulations the drug loading capacity ranged within 48 and 63 %. These formulations were used for dissolution studies over a period of 6 hours at pH 5.60 and 7.40. The dissolution results showed that no chitosan has dissolved at both pH values. A significant amount of rifampicin was, however, released from the beads, especially at pH 7.40. chitosan beads containing vitamin C also exhibited high rifampicin release (48.34 ± 1.00) %) at pH 5.60 compared to the other formulations and this makes vitamin C a potential excipient for enhanced drug release over a wide pH range (both acidic and alkalinic). However, further studies are necessary to optimise the preparation method to minimise drug loss during loading and to improve the drug loading capacity of the beads.Master
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
