Photothermally-Induced Molecular Self-Assembly of Macroscopic Peptide-Inorganic Hybrid Films

    No full text
We present a new strategy for a one-step preparation of macroscopic peptide-inorganic hybrid supramolecular films with functionality. It has been found that photothermal induction is of key importance to mediate the disassembly of supramolecular structures in solution phase, enrichment and reorganization at the interfaces associated with water evaporation. Incorporation of functional nanoscale objects into the film is achieved, providing the potential of integration of multiple functions. Texture and thickness of the hybrid film assembled at interfaces can be tuned and controlled by alteration of experimental parameters such as concentration and irradiation time. The photothermal treatment approach developed in this work is simple, flexible and controllable for design of macroscale functional supramolecular films, directly interfacing with biological systems. The photothermally-induced transformation of supramolecular structures is attractive for preparation of macroscale materials or devices towards a broad range of applications in biology and in nanotechnology.</p

Therefore, new antileukemia drugs with different action mechanisms are urgently needed to cope with this situation. Materials and methods: Arminin 1a-C is an antimicrobial peptide (AMP) developed from the ancient metazoan marine Hydra. In this study, we first explored its antileukemia activity. Results: Our results showed that Arminin 1a-C formed an α-helical structure and efficaciously suppressed the viability of leukemia cell lines whether or not they were multidrug resistant or sensitive, and there were no obvious differences between these cell lines. Arminin 1a-C exhibited distinct selectivity between noncancerous and cancerous cell lines. Arminin 1a-C interfered with K562/adriamycin (ADM) cell (a kind of multidrug-resistant leukemia cell line) proliferation in a very rapid manner and formed pores in its cell membrane, making it difficult to develop resistance against Arminin 1a-C. Conclusion: Our data show that Arminin 1a-C possesses great potential as a therapeutic candidate for the treatment of multidrug-resistant leukemia. Keywords: antimicrobial peptide, Arminin 1a-C, antileukemia, multidrug resistanc

Construction and self-assembly of beta-cyclodextrin derivative composite Langmuir films: Host-guest reaction and nanostructures

    No full text
The design and preparation of functional beta-cyclodextrin-based composite films are of key importance for wide application fields. This study demonstrates a new route to construct composite films by Langmuir-Blodgett (LB) method via cholesterol-modified beta-cyclodextrin derivative (CD-CHOL). The inclusion complex formation of CD-CHOL and azobenzene derivative (N-Azo) at the air/water interface was investigated. It has been found that the interfacial host-guest recognition could easily occur in the in situ formed CD-CHOL/N-Azo composite films and relatively difficult to occur in the ex situ immersed composite films, because there exist host-guest interaction as well as intermolecular hydrogen bonding interaction. The inclusion complex formation was confirmed by changes in the surface pressure-area isotherms, circular dichroism (CD) and UV-vis spectra of the transferred LB films. And the solvent effect of the composite CD-CHOL/N-Azo films was also investigated, indicating that host-guest interaction between N-Azo and CD-CHOL could occur rather in the nonpolar solvent than the polar solvent. The present research work about beta-cyclodextrin-based composite films by LB method may provide new exploration to prepare organized composite films and demonstrate potential applications in many fields.</p

Using a multicenter, randomized controlled trial, (N = 123, age 23 [+ or -] 4 years) we sought to determine whether administration of a novel, topical creatine supplement could improve muscular performance after acute and repeated (7-day) exposure. To study the acute performance enhancing effects of the supplement, subjects completed 5 sets of 15 maximal concentric single-leg knee extensions with and without the application of a low- (low dose [LD]-3.5 ml) or high-dose (high dose [HD]-7 ml) topical creatine cream. After a wash-out period, subjects had one leg randomized to receive either the creatine or placebo cream, with further randomization into an oral creatine or placebo supplement group. Subjects completed 5 sets of 15 maximal concentric single leg knee extensions before and after the supplementation protocol. After acute application, no significant differences in peak power (LD: 252 [+ or -] 93 W, HD: 261 [+ or -] 100 W, p = 0.21), average power (LD: 172 [+ or -] 65 W, HD: 177 [+ or -] 69 W, p = 0.78), or fatigue index (LD: 13.4 [+ or -] 10.6%, HD: 14 [+ or -] 11.9%, p = 0.79) were observed between experimental and placebo creams (peak power: LD: 244 [+ or -] 76 W, HD: 267 [+ or -] 109 W; average power: LD: 168 [+ or -] 57 W, HD: 177 [+ or -] 67 W; fatigue index: LD: 12.4 [+ or -] 9.6%, HD: 12.8 [+ or -] 10.6%) or when controlling for sex. After the 7-day supplementation protocol, a significant increase in average power (creatine: 203 [+ or -] 61-220 [+ or -] 65 W, placebo: 224 [+ or -] 61-214 [+ or -] 61 W) and peak power (creatine: 264 [+ or -] 73-281 [+ or -] 80 W, placebo: 286 [+ or -] 79-271 [+ or -] 73 W) in the leg receiving creatine cream was observed in male subjects. No differences were observed in female subjects. The topical creatine cream did not enhance measures of muscle performance after acute application, but was able to improve peak and average power in male subjects after 7 consecutive days of application

Ph.D.Introduction: HPV vaccination for the prioritized group of adolescent girls is well accepted and implemented in developed countries as an effective measure for cervical cancer prevention and control with increasing population-level effectiveness evidence accumulated. The government has played a key role in achieving universal HPV vaccination of target girls for different levels of organization lead to variations in program implementation outcome. Therefore, HPV vaccination is worthy of being analyzed politically to assess the engagement of the government and other actors to generate evidence for this health initiative globally and nationally.General objective: When comparing Hong Kong and Shenzhen, the difference in political actions of the two governments that Hong Kong government’ commitment on underprivileged girls and allocating resources earlier than Shenzhen was identified with similar level of political priority and available technical evidence according to the criteria in the WHO position paper. Political feasibility has not been assessed in both settings, which may have contributed to relatively low level of political attention. This prospective policy analysis is an entry point to inform policy-making to achieve target girls’ universal coverage either from a partial program or no further actions taken by government.Methodology: Hong Kong and Shenzhen were selected as two cases to explore the feasibility of the policy change on HPV vaccination. Document review and in-depth interviews with identified stakeholders were conducted to obtain data. The framework of Health Policy Triangle was adapted to guide data collection and analysis in terms of context, actors, process and content. Specific approaches of stakeholder analysis examining actors’ position, power, role and interest in force-field mapping and thematic analysis focusing on data coding and theme development were used. Results: In Hong Kong, contextual influences included new government seeking proposals, political and health system and a mix of Chinese and Western values. Most professionals and non-governmental organizations had high level support but medium power. Legislative organization and bureaucracies had high power level but their positions were not high support. Mothers had low power for policy formulation and their position was between neutral and opposition. Themes for the policy process include problem definition of cervical cancer as a priority, perceptions on voluntary immunization, economic evaluation evidence, the political action of the Pilot Scheme and advocacy efforts for promotion in Hong Kong. Mixed results were identified on policy content designs.Shenzhen’s contextual factors were legislative authority under a unitary political system, economic developments and cultural values on immunization and sexuality. Stakeholders’ position and power could be explained by their role and interest in the Top-down health administration. A category of supporters including stakeholders from adult immunization clinic and community health centers and a gynecologist for cervical cancer treatment was identified having lower power. Mothers could be potential bystanders if having little knowledge on HPV vaccination. Additional themes generated were HPV vaccination related knowledge, vaccine approval and use procedures, domestic development and production, HPV vaccine demand and access, the role of media and political attention on evidence-informed policy-making in Shenzhen. These stakeholders also discussed different aspects of program planning prospectively.Discussions: In Hong Kong, different positions of stakeholders could be explained by their ethical stances and political reasons because HPV vaccination is becoming high politics. The degree of legitimacy of policy-making process ending up with a recommendation or statement was debated. Policy design should focus on reframing the issue in terms of arguing facts and values related to HPV vaccination. Although identifying policy entrepreneurs is of challenges, mobilizing relevant groups and individuals to build a coalition is recommended.Shenzhen witnesses a possibility of demonstration projects for local government’s horizontal accountability but no potential advocates were identified at local level for fragmented organization of public health facilities and health professionals’ lacking mobilization skills. A cervical cancer prevention expert could be a policy entrepreneur. Despite of these challenges, the recommendations include multi-participation to engage NGOs, suppliers and target girls and their mothers, power enforcement along governing system, as well as better use of media.Comparison: There are similarities in cultural value, technical issues and HPV vaccination policy-making rooting in internal and political dynamics of the two settings respectively. Identified differences of political and health care system, stakeholder constitution and engagement in policy-making as well as their power level, underlying values and ethical perspectives offer plausible explanations for cross-setting variations of feasibility. However, common policy strategies to enhance the feasibility include mixed approach of marketing and planning, moving the issue to legislative procedure and evidence-informed policy-making to achieve good governance. Chen, Ruirui.Thesis Ph.D. Chinese University of Hong Kong 2018.Includes bibliographical references (leaves 150-167).Abstract also in Chinese; appendix III, IV in Chinese.Title from PDF title page (viewed on …).Chen, Ruirui
