    Droplet microfluidics for biomolecule separation and detection

    Droplet microfluidics is a new approach for chemical and biological analysis. Discrete nano-litre droplets ensure chemical reactions occur quickly without cross-talk, allowing samples to be processed without dispersion. Droplet microfluidics is effectively a digital sample processing platform enabling sample droplets to be handled in a continuous serial format.This thesis describes a method for in situ compartmentalization of biological and chemical samples after separation using Slipchip technology. Isoelectric focusing (IEF) was used to separate biomolecules within the Slipchip. The device was used to compartmentalise the IEFseparated samples into micro-droplets. This approach solves the compartmentalization challenge faced by current IEF systems. The digitised sample droplets can be collected serially or in parallel. In serial collection, droplets are collected in tubing that maintains their spatial sequence.For parallel collection, droplets are collected with a multi-pipette. Separated samples were assayed by gel electrophoresis on an Agilent Bioanalyzer. Separated samples were also postprocessed on-chip by mixing with pH indicator droplets for calibration of droplet pH. Suchcontinuous-flow/micro-droplet conversion methods have the potential of hyphenating different separation techniques for performing advanced analysis of complex samples.One disadvantage with droplet microfluidics is the very small dimensions of the sample meaning that classical absorption assays are difficult. Therefore, a high sensitivity absorption-based optical method called Cavity Ring Down Spectroscopy (CRDS) was developed and evaluated. The integration of CRDS with microfluidic devices was explored. Microfabricated cylindrical lenses were used to increase the light coupling efficiency within a chip, but results showed that light losses in the system were too high to enable the effective use of CRDS for analysis

    Power spectral density analysis for nonlinear systems based on Volterra series

    A consequence of nonlinearities is a multi-harmonic response via a mono-harmonic excitation. A similar phenomenon also exists in random vibration. The power spectral density (PSD) analysis of random vibration for nonlinear systems is studied in this paper. The analytical formulation of output PSD subject to the zero-mean Gaussian random load is deduced by using the Volterra series expansion and the conception of generalized frequency response function (GFRF). For a class of nonlinear systems, the growing exponential method is used to determine the first 3rd-order GFRFs. The proposed approach is used to achieve the nonlinear system's output PSD under a narrow-band stationary random input. The relationship between the peak of PSD and the parameters of the nonlinear system is discussed. By using the proposed method, the nonlinear characteristics of multi-band output via single-band input can be well predicted. The results reveal that changing nonlinear system parameters gives a one-of-a-kind change of the system's output PSD. This paper provides a method for the research of random vibration prediction and control in real-world nonlinear systems

    Substrate Protection in Controlled Enzymatic Transformation of Peptides and Proteins

    Proteins are involved in practically every single biological process. The many enzymes involved in their synthesis, cleavage, and posttranslational modification (PTM) carry out highly specific tasks with no usage of protecting groups. Yet, the chemists’ strategy of protection/deprotection potentially can be highly useful, for example, when a specific biochemical reaction catalyzed by a broad-specificity enzyme needs to be inhibited, during infection of cells by enveloped viruses, in the invasion and spread of cancer cells, and upon mechanistic investigation of signal-transduction pathways. Doing so requires highly specific binding of peptide substrates in aqueous solution with biologically competitive affinities. Recent development of peptide-imprinted cross-linked micelles allows such protection and affords previously impossible ways of manipulating peptides and proteins in enzymatic transformations.This article is published as Zhao, Yan.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
