    Antibacterial action of prodigiosin on Bacillus genus

    Prodigiozin je rdeči pigment, ki sodi v skupino prodigininov. Zanimiv je, ker deluje protimikrobno, protiglivno, algicidno in proti praživalim. Odkrili so tudi, da ima prodigiozin velik potencial kot protirakasta in imunosupresivna molekula. Protimikrobni mehanizem delovanja prodigiozina je slabo poznan, zato smo v magistrskem delu želeli določiti tarčne celične komponente bakterije B. subtilis, na katere deluje prodigiozin in pri tem povzroči celično smrt. Z dilucijsko metodo v mikrotitrskih ploščah smo različnim sevom Bacillus sp. določili MIK. MIK prodigiozina smo določili tudi bakterijam pri spremembi temperature inkubacije in slanosti v gojišču. Protibakterijsko aktivnost smo določali z merjenjem OD650 bakterijskih kultur v tekočem gojišču LB, kjer smo ob različnih fazah rasti kulturam dodali ustrezno koncentracijo prodigiozina. Hkrati smo celice spremljali tudi z uporabo mikroskopije z diferencialnim interferenčnim kontrastom in merjenjem aktivnosti β – galaktozidaze. V kontrolnih poskusih smo B. subtilis PS-216 tretirali z različnimi bakterolitičnimi in bakteriostatskimi sredstvi. Ugotovili smo, da so bili vsi testirani sevi Bacillus občutljivi na dodatek prodigiozina. Vrednosti MIK so bile različne in so znašale od 2,82 ± 1,81 mg/L pri Bacillus sp. 5A do 7,83 ± 2,86 mg/L pri B. subtilis NCIB 3610. V stresnih pogojih se je občutljivost bakterij na prodigiozin še povečala. Z opazovanjem celic pod mikroskopom in spremljanjem OD650 smo ugotovili, da prodigiozin povzroči avtolizo celic. Največji vpliv ima na hitro deleče se celice v eksponentni fazi rasti. Pokazali smo, da delovanje prodigiozina povzroči večjo prepustnost celične membrane. Iz dobljenih rezultatov sklepamo, da prodigiozin najverjetneje inducira avtolizine, ki povzročijo hitro lizo bakterijske populacije.Prodigiosin is a natural red pigment from a family of prodiginins. It is interesting because of its antibacterial, antifungal, algicidal and antiprotozoal activity. In addition, the anticancer and immunosuppressive properties of prodigiosin were described. The mechanism of prodigiosin\u27s antibacterial action is still unknown. The aim of the study was to determine cellular target components which prodigiosin affects and causes bacterial cell lysis. Minimal inhibitory concentration (MIC) of Bacillus sp. was evaluated by using broth dilution method in microtiter plates. MIC was also evaluated at different incubation temperatures and different medium salinity. The antibacterial activity was evaluated with measuring optical density of bacterial strains in the LB medium at 650 nm, where prodigiosin was added at different cell growth phases. For cell viability evaluation differential interference contrast microscopy was used and cytoplasmic membrane leakage was determined by measuring β – galactosidase activity. As a control, B. subtilis PS-216 was treated with different bacteriolytic and bacteriostatic agents. We have demonstrated, that all tested Bacillus strains are susceptible to prodigiosin. MIC value was between 2,82 ± 1,81 mg/L against Bacillus sp. 5A and 7,83 ± 2,86 mg/L against B. subtilis NCIB 3610. Exposure of bacterial strains to stress caused by temperature fluctuation and NaCl concentraton increased susceptibility to prodigiosin. Microscopy and optical density data indicate that prodigiosin causes celly lysis. The highest impact of prodigiosin was observed on actively dividing cells in the exponental growth phase. We have observed that prodigiosin increases permeability of cytoplasmic membrane. Based on our results, we have proposed that prodigiosin induces autolysins, leading to fast autolysis of bacterial culture

    V diplomskem delu želimo prikazati možnosti za izgradnjo intraneta na Osnovni šoli Tabor Logatec. V prvem delu bomo teoretično obdelali intranet, kaj potrebujemo za izgradnjo in kakšne so njegove prednosti. Prikazali bomo obstoječe stanje v organizaciji. Kritični bomo predvsem do sistema obveščanja in upravljanja z dokumenti. Opisali bomo tudi stanje računalniške opreme v organizaciji. V nadaljevanju bomo predstavili projektni način reševanja problema. Opisali bomo aktivnosti in postopke, ki jih mora organizacija pri tem izvesti. Pri tem bomo upoštevali možnosti, ki jih organizacija ima. Predstavili bomo načine in postopke upravljanja z dokumenti in drugimi vsebinami. Za predlagan sistem smo izbrali sistem za upravljanje z vsebinami Joomla! V prototipni rešitvi bomo prikazali možnost uporabe tega sistema in opisali prednosti in slabosti, ki jih ta prinaša v Osnovno šolo Tabor Logatec.In our graduation thesis we wish to demonstrate the possibilities of constructing an intranet at the primary school Osnovna šola Tabor Logatec. In the first part we will describe the theoretical part of the intranetwhat is needed for its construction and what its advantages are. We will also display the current state in the organization. We will take on a critical view towards the system of information and document managing. We will also describe the state of the computer equipment in the organization. Next we will present the project method of solving problems. We will also describe the activities and procedures that the organization has to carry out. We will take into account all the possibilities the organization has. We will also describe the methods and procedures of managing documents and other contents. For the proposed system, we have chosen the content management system Joomla! In a prototypical solution we will demonstrate the possibility of using this system and describe all the advantages and disadvantages that the above mentioned system brings to the primary school Osnovna šola Tabor Logatec

    Quantifying the major sinks and sources of phosphorus in Tabor Lake: implications for management and remediation

    A study of Tabor Lake and its watershed during the open water season of 1995 was conducted in order to quantify the major sinks and sources of phosphorus, and the timing of phosphorus transfer between these sources and sinks. The results of this thesis are useful to help determine appropriate management strategies aimed at reducing phosphorus availability in Tabor Lake's water column and understanding some of the processes that control phosphorus availability in the lake. An exploratory modeling exercise was undertaken to evaluate morphometric and watershed variables in predicting spring phosphorus for 39 lakes in the region, including Tabor Lake. Two multiple linear regression models were constructed. The first model uses a stepwise technique to select statistically significant variables, and the second model uses variables found in the literature which have been shown to influence phosphorus loading. These two models account for 70% and 59% of the variation in spring phosphorus, respectively, and may provide managers with an expected phosphorus loading based on easily measured morphometric and watershed variables. The models predict that Tabor Lake should have a spring phosphorus concentration of 20 and 18 ug/L, respectively. The models are useful to establish the limits of water quality improvement for Tabor Lake. Another model was developed to estimate the quantity and timing of external sinks and sources of phosphorus in Tabor Lake for 1995. Phosphorus loading into Tabor Lake was estimated from runoff (136 kg) and atmospheric deposition (60 kg). Phosphorus output from Tabor Lake through the outlet creek was estimated at 188 kg for the entire year. Based on this input-output model, there was a net gain of 8 kg phosphorus into Tabor Lake during 1995. These results indicate that external phosphorus inputs and output is approximately balanced. Results from a prior monitoring program indicate that Tabor Lake experienced internal loading of phosphorus during periods of hypolimnetic anoxia. However, phosphorus loading also occurred when oxygen was present in the hypolimnion, suggesting another phosphorus loading mechanism was active in Tabor Lake. A specific effort was made to evaluate the role of senescing macrophytes (primarily Elodea canadensis) as a source of this phosphorus during the 1995 sampling season. Two experiments were conducted, one invitro and the other in-situ, to estimate phosphorus leaching from senescing macrophytes. The first in-vitro experiment used weeds collected August 8, 1995 and show that 87% (S.E.= 6%) of the total plant phosphorus is leached from these macrophytes, whereas the second in-vitro experiment, using samples collected September 22, 1995, show that only 9% (S.E.= 1%) of the phosphorus is leached from macrophytes. The difference between the two leaching estimates is believed to be the result of overwintering development and expected starch accumulation in the September samples. The in-situ study provided the best prediction of phosphorus leaching from senescing macrophytes, estimating 1958 kg of phosphorus (S.E.= 867 kg) was released from senescing macrophytes in 1995. The results from both the input-output model and the internal loading estimates show that Tabor Lake is dominated by the internal cycling of nutrients beginning in June and continuing until September. Furthermore, it quantifies two important phosphorus sources and the timing of phosphorus delivery from these sources. The quantification and timing of phosphorus delivery in Tabor Lake is useful to managers establishing remediation strategies which try to reduce the total phosphorus present in the water column. This research also shows that mechanical harvesting of macrophytes removes more phosphorus from the lake than exits through the outlet and can be useful in reducing the total phosphorus available to internal loading cycles in Tabor Lake.The original print copy of this thesis may be available here: http://wizard.unbc.ca/record=b118088

    Historians know almost nothing about the two decades following the crucifixion of Jesus when his followers regrouped and began to spread his message. During this time, the apostle Paul joined the movement and began to preach to the Gentiles. Using the oldest Christian documents that we have -- the letters of Paul -- as well as other early Christian sources, historian and scholar James Tabor reconstructs the origins of Christianity. Tabor reveals that the familiar figures of James, Peter, and Paul sometimes disagreed fiercely over everything from the meaning of Jesus\u27 message to the question of whether converts must first become Jews. Zamieszczone w artykule opisy budowy i działania zespołów charakterystycznych dla elektropneumatycznych i mikroprocesorowych układów sterowania ilustrują rozwój tych układów w ostatnich dwudziestu latach. Efektem tego rozwoju jest przedstawiona w tym artykule propozycja układu sterowania z uniwersalną tablicą pneumatyczną o budowie modułowej.In the paper the braking control systems developed by Rail Vehicles Institute “TABOR” are presented. These systems are used in the modernized diesel and electric locomotives in Poland. Design and operation of electropneumatic and microprocessor based control systems are described. The development of these systems over the last twenty years is shown. The proposal of control system with the new universal modular pneumatic board for modernized and new-built locomotives, being a direct result of the research of Rail Vehicles Institute “TABOR”, is described
