    FARA: A Fast Artifact Recovery Algorithm with Optimum Stimulation Waveform for Single-Cell Resolution Massively Parallel Neural Interfaces

    This paper introduces a fast artifact recovery algorithm (FARA) that uses electrochemical impedance spectroscopy to model the electrode-tissue interface and design an optimum stimulation waveform to minimize the residual artifact duration in single-cell resolution neural interfaces. Results in saline solution with a custom PCB and a 30 μm\mu \mathrm{m} diameter microelectrode array show a worst case artifact recovery time of 160 μs\mu \mathrm{s} when measured from the end of the working phase (anodic 500 nA,250μs)\mathrm{n}\mathrm{A}, 250\mu \mathrm{s}). It is caused by intronic GAA repeat expansions that hinder the expression of the FXN gene, resulting in defective levels of the mitochondrial protein frataxin. Sensory neurons in dorsal root ganglia (DRG) are particularly damaged by frataxin deficiency. There is no specific therapy for FRDA. Here, we show that frataxin levels can be upregulated by interferon gamma (IFNγ) in a variety of cell types, including primary cells derived from FRDA patients. IFNγ appears to act largely through a transcriptional mechanism on the FXN gene. Importantly, in vivo treatment with IFNγ increases frataxin expression in DRG neurons, prevents their pathological changes and ameliorates the sensorimotor performance in FRDA mice. Na základě požadavků zákazníka je představen návrh systému. Následně je popsána implementace, testování a nasazení navrženého systému. Z práce vyplynul pro autora i postup, jak na základě získaných zkušeností a reference začít s vývojem informačních systémů pro další koncové zákazníky.This diploma thesis deals with the design and implementation of the Information System for the real customer. The main emphasis is put on the analysis of the business plan of the whole project. On the basis of the customer requirements, the design of the System is introduced. Afterwards, the implementation, testing, and deployment of the System are described. From the thesis, the method emerged that the author can apply to begin with the development of the Information Systems for the next end-users with the help of the gained experience and reference

    Diplomová práce se zaměřuje na interní a externí strategickou analýzu podniku Autoseating, s.r.o. V teoretické části byly vysvětleny základní pojmy o strategii, vizi a misi. Dále se autor zaměřil na analýzu Pest a Porterův model 5 sil, interní zdroje z teoretického hlediska pokryl analýzou zdrojů podniku a finanční analýzou. Závěrem teoretické části byla vysvětlena SWOT analýza a teoretický výběr generických strategií dle Portera. V praktické části byly veškeré zmíněné analýzy provedeny. Pest analýza spíše konstatovala jednotlivé aspekty. V Porterově modelu 5 sil byly zjištěny rizikové síly v podobě dodavatele velkoobchodu sedaček a v roli odběratele ve výrobní sekci firmy Autoseating, pro prodejnu Honzík bylo zjištěno značné ohrožení ze strany e-shopů. Hlavní přínos z interní analýzy měl finanční rozbor podniku, který poukazuje na potenciál lepšího využití kapitálu a současně doporučuje nalézt způsob vyšší obrátky zásob. Swot analýza pro každou sekci firmy Autoseating odhalila stejné silné a slabé stránky, včetně hrozeb, které byly naznačeny již v případě PEST analýzy. Firma se zaměřuje na kvalitní sortiment, proto autor navrhl aplikovat porterovu generickou strategii diferenciace. Společně s výběrem je navrženo i několik návrhů na zlepšení stávajícího systému.The Diploma thesis is focused on internal and external analysis of the company Autoseating. In the first, teoretical, part of the thesis are explained the basic therms as a strategy,a mission and a vision follower by the external Pest and Porter 5 forces analyses. Author continued with an internal analysis represented by resource analysis and financial analysis. At the end of teoretical part were explained swot analysis together with choice of generic strategies made by porter. Practical part fullfiled all the analyses mentioned. Pest analysis was made as a first one, with no breaking news. But porter analysis of 5 forces disclosed endangering by suppliers in a case of import-branch in Autoseating, an endangering by customers in the case of car-parts production, an finally big endangering by e-shops for shop "Prodejna Honzik". The majority contribution brought an internal analysis, particulary the financial one. There were suggested better usage of capital and to take measures to improve stock-turn. Final swot analysis confirmed facts found in Porters model of 5 forces (competition analysis). The company Autoseating deals with the high quality products, so the author decided to recommend diferenciation strategy and added some measures to improve contemporary system

    This thesis tackles key challenges in fluid-structure interaction (FSI) mod- eling, focusing on the Arbitrary Lagrangian-Eulerian (ALE) method, which couples fluid motion with Lagrangian solid tracking. A major contribution is novel re-meshing strategy addressing ALE's instability under large deforma- tions. The author developed ADmesh software, an iterative mesh optimizer that maintains subdomain structures and enables stable simulations of elastic rebound without artificial contact laws. A validated implementation confirms accuracy through benchmarks. including Turek-Hron. Finally, we reformulate ALE system suitable to support higher-order time integration, as illustrated in numerical tests confirming expected convergence. These developments signifi- cantly improve ALE-based FSI, particularly for large deformations and complex interface dynamics. 1Tato disertační práce se zabývá klíčovými výzvami modelování interakce tekutiny a struktury (FSI), se zaměřením na metodu Arbitrary Lagrangian- Eulerian (ALE), která kombinuje popis pohybu tekutiny na pohybující se síti s Lagrangeovským sledováním pevné látky. Hlavním přínosem je nová strategie přesíťování, která řeší nestability ALE při velkých deformacích. Autor vyv- inul ADmesh, iterativní nástroj pro optimalizaci sítě, jenž zachovává struk- turu poddomén a umožňuje stabilní simulace pružného odskoku bez použití umělých kontaktních zákonů. Práce dále rozvíjí modelování skluzu na rozhraní tekutina-těleso, čímž překonává tradiční předpoklad spojitosti. Implementace byla ověřena pomocí numerických experimentů, včetně náročného benchmarku Turek-Hron. Závěrem je představena nová formulace ALE rovnic vhodná pro vyšší řády časové diskretizace, u níž testy potvrzují správné konvergenční vlast- nosti.     The early Pliensbachian (Early Jurassic) is known as a time of marked provincialism in the marine realm, notably between the Mediterranean Tethys and North-West Europe. In order to test this observation quantitatively, we compiled 104 locality-level species lists from those areas based on a comprehensive revision of early Pliensbachian ammonites. With this dataset, we also explore the relationship between ammonite richness and biogeography at the scale of the sub-chronozone during the early Pliensbachian. Using various multivariate statistics and rarefaction techniques, we show that: (i) there is a sharp contrast between the NW European (NWE) and the Mediterranean (MED) provinces, although there is some mixing in Austroalpine and Pontic ammonite faunas; (ii) species richness in the MED province is about twice that in the NWE province for each chronozone; (iii) ammonite species richness tends to decrease during the early Pliensbachian, especially at the Ibex-Davoei transition; and (iv) the NWE and MED sensu stricto provinces both record the same pattern of variations in richness despite the fact that their taxonomic compositions have virtually nothing in common at the species level. We suggest that the low ammonite richness of the Davoei chronozone may be related to a coeval warming of seawaters, but that this was insufficient to affect the sharp palaeobiogeographic contrast between the two provinces. This persistent compartmentalisation probably reflects a major palaeogeographical structure, such as an emerged or near-emerged barrier running from the Betic range to the Briançonnais ridge.     A Tool to Solve Sentence Segmentation Problem on Preparing Speech Database for Indonesian Text-to-speech System

    AbstractCreating a training data ready to be used for developing a text-to-speech (TTS) system can be a difficult task, since sometimes the recorded audio data is not the same with the prepared texts. To overcome differences between audio and text data, we developed a tool to segment audio data into sentences. As it is known, doing sentence segmentation of audio data manually needs efforts and resources. This paper presents a solution for alleviating problems encountered during segmentation process of audio data for developing an Indonesian TTS system. The tool was developed based on a fact that bahasa Indonesia is a syllable-timed language. We found that our tool reduces resources needed for segmenting Indonesian audio data
