    TACC3-ch-TOG track the growing tips of microtubules independently of clathrin and Aurora-A phosphorylation

    The interaction between TACC3 (transforming acidic coiled coil protein 3) and the microtubule polymerase ch-TOG (colonic, hepatic tumor overexpressed gene) is evolutionarily conserved. Loading of TACC3–ch-TOG onto spindle microtubules requires the phosphorylation of TACC3 by Aurora-A kinase and the subsequent interaction of TACC3 with clathrin to form a microtubule binding surface. Whether there is a pool of TACC3–ch-TOG that is independent of clathrin in human cells, and what is the function of this pool, are open questions. Here, we report that TACC3 is recruited to the plus-ends of microtubules by its association with ch-TOG and that this pool is independent of phosphorylation and binding to clathrin. The plus-end binding of TACC3–ch-TOG persists in interphase and we propose that one cellular function of TACC3–ch-TOG is to modulate cell migration. We also describe the distinct subcellular pools of TACC3, ch-TOG and clathrin. TACC3 is often described as a centrosomal protein, but we show that there is no significant population of TACC3 at centrosomes.     Adaptive refinement of all-hexahedral elements for three-dimensional metal forming analysis

    In many 3-D forging simulations, the master-grid method or uniform-grid method is adopted as an effect tool for hexahedral element mesh generation, because the method is robust and reliable to construct the intermediate mesh during forging simulation. However, almost equalsized element mesh is employed regardless of the local characteristics of the deforming region. In order to overcome this drawback, a new refinement technique for hexahedral element mesh is proposed by selecting the element regions to be refined and iteratively inserting the zero-thickness element layers into the necessary interfaces of elements. To generate refined mesh adaptive to the gradient of physical property or to the complexity of geometry, the desired mesh density is obtained from the Z-Z posteriori error analysis. In the course of expanding and smoothing the refined mesh to ensure the desired mesh density, the weighting factor based on mesh density of neighbor entities is introduced to the conventional Laplacian smoothing method. Comparative simulations of the selected forging processes have shown that the proposed refinement technique is effective from the viewpoint of computational economy of computations and accuracy.     Determination of temperatures using CH radical emission spectroscopy

    An improved Boltzmann plot method where the intensity is taken as the integral of the experimental spectrum within a special band for a cluster of a rotational line of R and Q branches is proposed. This method aims at deducing rotational and vibrational temperatures using CH radical A(2) Delta -> X-2 Pi band emission spectroscopy accurately. In addition, the data relative to the rotation lines of CH(A(2) -> X-2 Pi) for both temperatures are assembled. The emission spectrum of CH(A(2) Delta -> X-2 Pi) at the inner cone of an acetylene-oxygen flame in a rich oxygen state is recorded and both of the temperatures are determined by the above-mentioned method. The values are recorded as 3141 K and 3097 K, for the rotational and vibrational temperatures, respectively. This result reveals that the equilibrium between the rotation and vibration states is achieved. A simple discussion for this method is also provided

    The R. anatipestifer strains (clockwise from top left) RA-CH-1pLMF03, RA-CH-1ΔB739_1343pLMF03, and RA-CH-1ΔB739_1343pLMF03::B739-1343 were grown on TSA plates containing cefoxitin (1 μg/mL) and 0 μM Dip (A) or 50 μM Dip (B). Growth was assessed by the appearance of bacterial colonies on plates. Pictures were taken after 48 h of growth at 37°C. All the experiments were repeated three times. Representative plates are presented.</p
