    Transient outward K+ current (ITO) reduction prolongs action potentials and promotes afterdepolarisations: a dynamic-clamp study in human and rabbit cardiac atrial myocytes

    Background and aim: Human atrial transient outward K+ current (ITO) is decreased in a variety of cardiac pathologies, but how ITO reduction alters action potentials (AP) and arrhythmia mechanisms is poorly understood, owing to non-selectivity of ITO blockers.<p></p> Aim: to investigate effects of selective ITO changes on AP shape and duration (APD), and on afterdepolarisations or abnormal automaticity with beta-adrenergic-stimulation, using the dynamic-clamp technique in atrial cells.<p></p> Methods and Results: Human and rabbit atrial cells were isolated by enzymatic dissociation, and electrical activity recorded by whole-cell-patch clamp (35-37oC). Dynamic-clamp-simulated ITO reduction or block slowed AP phase 1 and elevated the plateau, significantly prolonging APD, in both species. In human atrial cells, ITO block (100% ITO subtraction) increased APD50 by 31%, APD90 by 17%, and APD-61mV (reflecting cellular effective refractory period) by 22% (P<0.05 for each). Interrupting ITO block at various time points during repolarisation revealed that the APD90 increase resulted mainly from plateau-elevation, rather than from phase 1-slowing or any residual ITO. In rabbit atrial cells, partial ITO block (~40% ITO subtraction) reversibly increased the incidence of cellular arrhythmic depolarisations (CADs; afterdepolarisations and/or abnormal automaticity) in the presence of the beta-agonist isoproterenol (0.1 μM; ISO), from 0% to 64% (P<0.05). ISO-induced CADs were significantly suppressed by dynamic-clamp increase in ITO (~40% ITO addition). ISO+ITO decrease-induced CADs were abolished by beta1-antagonism with atenolol at therapeutic concentration (1 μM).<p></p> Conclusion. Atrial cell action potential changes from selective ITO modulation, shown for the first time using dynamic-clamp, have the potential to influence reentrant and non-reentrant arrhythmia mechanisms, with implications for both the development and treatment of atrial fibrillation

    Bonding InGaAsP/ITO/Si Hybrid Laser With ITO as Cathode and Light-Coupling Material

    A 1.5-mu m InGaAsP/ITO/Si hybrid laser with indium tin oxide (ITO) as both a cathode and a light-coupling material is presented. The InGaAsP gain structure with a transparent ITO cathode is flip-chip bonded onto a patterned silicon-on-insulator wafer. The light generated in the InGaAsP multiquantum wells is coupled through the ITO cathode into the Si waveguide to form an InGaAsP/ITO/Si hybrid laser. The threshold current density of this hybrid laser is 20 kA/cm(2) at 210 K.     Introduction: hypomelanosis of Ito is a neurocutaneous syndrome and a multisystem disorder, described by Japanese dermatologist Minor Ito in 1951. This disease is characterized by hypopigmented lesions of linear or annular distribution on the skin, associated or not with neurological and ocular manifestations, as well as dental and musculoskeletal malformations.Case report: 12-year-old male patient, son of young non-blood parents with a family history of vagal crises, personality disorders and epileptic seizures in childhood. Personal pathological history included right unilateral appendageal since birth, spasms during sleep at 3 months, joint hyperlaxity and hypotonia at one year, delayed psychomotor development, focal temporal lobe and tonic-clonic epilepsy seizures at 10 years old, diagnosis of right pectoral agenesis at 1 year old and bilateral pyelectasis. He attended the Provincial Clinical Genetics Service referred by the Dermatology Clinic for presenting hypopigmented macules in the upper right limb, accompanied by the neurological, musculoskeletal and genitourinary signs described above. Taking into account the international criteria based on cutaneous and extra cutaneous manifestations, hypomelanosis of Ito was diagnosed.Conclusions: hypomelanosis of Ito is a rare, multisystem disorder, with a fundamental clinical diagnosis. Early diagnosis is essential in order to provide a timely treatment, as well as knowing the forms of presentation and manifestations, achieving a better quality of life for the patient.Introducción: la hipomelanosis de Ito es un síndrome neurocutáneo y una enfermedad multisistémica, descrita por el dermatólogo japonés Minor Ito en 1951. Esta enfermedad se caracteriza por lesiones hipopigmentadas de distribución lineal o anular en la piel asociadas o no a manifestaciones neurológicas y oculares, así como malformaciones dentales y musculoesqueléticas.Presentación del caso: paciente masculino de 12 años de edad, hijo de padres jóvenes no consanguíneos y con antecedentes patológicos familiares de crisis vagales, trastornos de la personalidad y crisis de epilepsia en la infancia. En los antecedentes patológicos personales se recogió apéndice preauricular unilateral derecho desde el nacimiento, espasmos durante el sueño a los 3 meses, hiperlaxitud articular e hipotonía al año, retraso en el desarrollo psicomotor, crisis de epilepsia focal lobal temporal y tónico-clónicas a partir de los 10 años, diagnóstico de agenesia pectoral derecha hace 1 año y pielectasia bilateral. Acudió al servicio de Genética Clínica provincial remitido por la consulta de Dermatología por presentar máculas hipopigmentadas en miembro superior derecho, acompañada de las manifestaciones neurológicas, musculoesqueléticas y genitourinarias antes descritas. Teniendo en cuenta los criterios internacionales que se basan en las manifestaciones cutáneas y extracutáneas, se llegó al diagnóstico de hipomelanosis de Ito,Conclusiones: la hipomelanosis de Ito es una entidad rara y multisistémica, de diagnóstico fundamentalmente clínico. El diagnóstico precoz es esencial para tratar oportunamente las formas de presentación y manifestaciones, logrando propiciar una mejor calidad de vida al paciente.

    UV-sensitive optical sensors based on ITO-gallium phosphide heterojunctions

    Design and characteristics of wide-band UV sensors based on ITO/GaP heterostructures are discussed. Such sensors have perfect electrical parameters and high UV-visible sensitivity in comparison with surface-barrier structures using a semitransparent thin metal film as an electrode. Many applications require UV sensors with an effective rejection of visible radiation and a wide temperature operating interval. For this aim, the theoretical modelling of extreme selective optical sensors with a double Ag/ITO thin film on the GaP surface, in which the thin silver film serves as a narrow bandpass filter at 320 nm, has been conducted. With this modelling the optimal thickness combination for the silver and ITO films was found for the maximum rejection of the sensitivity to visible radiation

    La alternancia –ito/-illo en hablantes sevillanos de nivel educacional bajo: Un estudio en tiempo real

    El objetivo de este estudio es comparar el empleo de -ito/-illo en hablantes sevillanos de un nivel instruccional bajo en dos esta- dios cronológicos diferentes. Para ello se ha tomado una muestra de 48 entrevistas que fueron grabadas en dos etapas (en torno a 1985, la primera, y a 2010, la segunda). La submuestra más an- tigua corresponde a los materiales del grupo de investigación Sociolingüística Andaluza: Estudio Sociolingüístico del Habla de Sevilla (HUM 141), mientras que la más reciente pertenece al corpus PRESEEA-SE. El análisis cuantitativo del fenómeno ha permitido constatar una evolución hacia un mayor uso del di- minutivo, especialmente de –ito en detrimento de –illo. En rela- ción con las variables lingüísticas y discursivas consideradas, se ha comprobado que se mantiene la preferencia por la función centralizadora del diminutivo, así como por su aplicación a sus- tantivos. Asimismo, se observa un descenso en la productividad de –ito; -illo resulta más productivo, pues posee una capacidad más amplia para aplicarse a bases distintas y, por último, se trata de un empleo predominante en los hombres y los hablantes más jóvenes, que, sin embargo, lo abandonan con la edad
