Unfreezing of molecular motions in protein-polymer conjugates: a calorimetric study
Get PDFProtein-polymer conjugates are a promising class of biohybrids. In this work, the dynamics of a set of biodegradable conjugates myoglobin-poly(ethyl ethylene phosphate) (My-PEEP) with variations in the number of attached polymers and their molar mass in the dry-state, have been investigated to understand the role of polymer on protein dynamics. We performed Differential Scanning Calorimetry measurements between 190 and 300 K, observing the large-scale dynamics arising from reorganization of conformational states, i.e. within the 100 s timescale. The application of an annealing time during the cooling scans was used to investigate the non-equilibrium glassy-state of the samples, observing the relaxation enthalpy at different annealing temperatures. This procedure permitted to extensively describe the transition broadness and the system relaxation kinetics in the glassy state. The samples show an experimental behaviour different from the theoretical predictions, suggesting the establishment of interactions among the protein and the polymer chains. The different behaviour of the conjugates and the physical mixture (composed of the protein and the polymer physically mixed) highlighted the importance of the covalent bond in defining the system dynamics

No full textThis is a critical introduction to the educational thought of F. R. Leavis (1895-1978), the greatest English literary critic of the twentieth century, providing the first in-depth examination of Leavis's ideas in relation to contemporary mass higher education. During the course of a long, prolific and controversial academic career, which saw him take issue with figures such as Wittgenstein, T. S. Eliot and C. P. Snow, Leavis became one of the most articulate advocates for the idea of the university as 'a centre of consciousness and human responsibility' in the face of what he saw as the relentless technological drive of civilisation. With the journal Scrutiny which he co-founded, as well as his critical writings, Leavis became a decisive influence on generations of teachers in Britain and overseas. Widely misrepresented as narrowly elitist, his ideas about 'the creative university', with their radical, student-centred approach to teaching, constitute a powerful resource for a higher education system grappling with the contradictory demands of continuity and change. Based on original research, the study provides an overview of Leavis's life, work and heritage and his educational world view, and a comprehensive exploration of Leavis's pedagogy from theoretical and practical perspectives. Thermodynamic stability of myoglobin-poly(ethylene glycol) bioconjugates: A calorimetric study
Get PDFPEGylated proteins are widely used for therapeutic applications, therefore a fundamental understanding of the
conjugates’ structure and their behaviour in solution is essential to promote new developments in this field. In
the present work, myoglobin-poly(ethylene glycol) conjugates were synthesized and studied by differential
scanning calorimetry and UV–vis spectroscopy to obtain information on the bioconjugates’ thermodynamic
stability, also focusing on PEG’s role on the solvent-protein surface interaction. The overall results of this study
indicated a thermal destabilization of the protein that follows the extent of the bioconjugation without, however,
compromising the native structure which remains functional. Moreover, the myoglobin PEGylation prevented
the post-denaturation aggregation phenomena and enhanced the protein thermal reversibility. The thermodynamic
interpretation of the data indicated that the bioconjugation influences the solvent-exposed protein
surface difference between native and denatured state, contributing to the interpretation of the overall protein
modification and functionality

No full textDer Terminus "switch-reference" ist eine Schöpfung von W. Jacobson (1967). Er bezog sich auf ein Phänomen, das zunächst in den putativen Hokan-Sprachen Tonkawa, Washo und Kashaya (Southern Pomo) entdeckt worden war. Es wurde bald als areales Merkmal von Sprachen des Südwestens Nordamerikas erkannt. Es findet sich in allen Great-Basin-Sprachen sowie der westlichen Hälfte des Südwest-Phylums und in Teilen der Phyla Kalifornien, Plateau, Plains und Südost (Jacobson 1983:172). [...] Seit einigen Jahren bringt man diese Erscheinung funktional und terminologisch in Verbindung mit den sog. Medialverben der Papua-Sprachen […]. "Switch-reference" ist die oppositionelle explizite Signalisierung der Identitätsrelation zwischen dem Subjekt des Satzes, an dem die Markierung vorgenommen wird (oder dem sie unmittelbar folgt) und dem Subjekt eines kommenden Satzes (vorzugsweise des nächsten). Sie hat also kriterial eine antizipatorische Komponente. Sie tritt in verbfinalen Sprachen auf und wird deshalb meist durch Enklitika oder Suffixe verkörpert (quasi als Brücke zum folgenden Satz), die den markierten Satz subordinieren. Mit der Subjektidentität (die zwangsläufig auf die eine oder andere Weise mit den Kategorien Person und Numerus interagiert) gehen meist andere Bedeutungen einher, vorzugsweise interpropositionale. Die "switch-reference" hat logischerweise zwei Optionen: Disjunktheit, "different subject" (DS) , und Identität "same subject" (SS). "Oppositionelle Signalisierung" impliziert, daß eine Option gegenüber der anderen keinen grundlegenden Umbau der Satzstruktur erfordert. Davon unberührt bleibt die Tatsache, daß DS-Markierung merkmalhaft er ist. [...] Die Arbeit sollte sich ursprünglich auf Papua- und amerindische Sprachen erstrecken. Obwohl auch letztere ausgiebig untersucht wurden, kam die Darstellung nicht über die Verhältnisse in den Papua-Sprachen hinaus. Angesichts der Sprachenvielfalt scheint mir der Begriff Typologie im Titel noch gerechtfertigt. Ebenfalls keine Berücksichtigung fand die SV der südamerikanischen Sprachen, wofür die Untersuchungsgrundlage aber ohnehin dürftig gewesen wäre.
