Polibio e Callistene: una polemica non personale?
The paper has been committed by the organizers of the Volume "The shadow of Polybius. Intertextuality as research Tool in Greek Historiography", in order to complete a full analysis of the impact of this historian over his predecessors; the paper studies carefully all the quotations of Callisthenes made by Polybius – with a particular care for the manuscript tradition of the fragmentary Book XII - and highlights Polybius’ mental attitude towards Callisthenes. The author defines also the place of Polybius among the various ancient traditions about Callisthenes both as a person and as an historian

Poor sleep quality is associated with decreased social activity satisfaction and increased stress in rural-living, late-middle-aged Latinos. Study results help characterize this understudied minority group at high risk for poor occupational health and outcomes.

Suzuki-Miyaura diversification of amino acids and dipeptides in aqueous media

This work is supported by the Scientific Research Network (WOG) “Sustainable chemistry for the synthesis of fine chemicals” of the Research Foundation - Flanders (FWO).The Suzuki-Miyaura derivatisation of free amino acids, peptides and proteins is an attractive area with much potential utility for medicinal chemistry and chemical biology. Here we report the modification of unprotected and Boc-protected aromatic amino acids and dipeptides in aqueous media, enabling heteroarylation and vinylation. We systematically investigate the impact of the peptide backbone and adjacent amino acid residues upon the reaction. Our studies reveal that whilst asparagine and histidine hinder the reaction, by utilising dppf, a ferrocene-based bidentate phosphine ligand, cross-coupling of halophenylalanine or halotryptophan adjacent to such a residue could be enabled. Our studies reveal dppf to have good compatibility with all unprotected, proteinogenic amino acid side chains.Peer reviewe
Studia na temat św. Jana Bosko – między zachwytem a pytaniami krytycznymi
This contribution deals with the theme of the studies made on Don Bosco. In the article the author equally takes into consideration both the studies carried out in the Salesian world and the contributions which do not belong to the Salesian world in the strict sense. The main objective was to analyze the various periods in which different authors made a valid contribution in deepening the figure of Don Bosco throughout his history. Thanks to the edition of the sources and the publications of numerous studies, the traditional image of Don Bosco is no longer the only one. Alongside it gradually grows a richer image, more differentiated and more in conformity with the historical reality of its time. The renewed image of Don Bosco is more suggestive. The use of more reliable historical data has made it possible to have a clearer vision of both the ecclesial religious situations and the social and educational ones, in which it was necessary to relocate Don Bosco. Thanks to the considerations made, Don Bosco appears as one who in his care and action knew how to relate to the concrete situations of his own time. In that climate he developed a pedagogical and spiritual orientation which also for the future could be a light and strength both inside and outside the Salesian world.Ten wkład dotyczy tematu studiów nad Księdzem Bosko. W artykule autorka w równym stopniu uwzględnia zarówno badania prowadzone w świecie salezjańskim, jak i wkłady, które nie należą do świata salezjańskiego sensu stricto. Głównym celem była analiza różnych okresów, w których różni autorzy wnieśli ważny wkład w pogłębienie postaci Księdza Bosko w całej jego historii. Dzięki edycji źródeł i publikacji licznych opracowań tradycyjny wizerunek Księdza Bosko nie jest już jedyny. Wraz z nim stopniowo wyrasta obraz bogatszy, bardziej zróżnicowany i bardziej zgodny z historyczną rzeczywistością tamtych czasów. Odnowiony obraz Księdza Bosko jest bardziej sugestywny. Wykorzystanie bardziej wiarygodnych danych historycznych umożliwiło uzyskanie jaśniejszego obrazu zarówno kościelnych sytuacji religijnych, jak i społecznych i wychowawczych, w których konieczne było przeniesienie Ks. Bosko. Dzięki poczynionym rozważaniom Ksiądz Bosko jawi się jako ten, który swoją troską i działaniem potrafił odnieść się do konkretnych sytuacji swoich czasów. W tym klimacie rozwinął orientację pedagogiczną i duchową, która także na przyszłość może być światłem i siłą zarówno w świecie salezjańskim, jak i poza nim

How are falls and fear of falling associated with objectively measured physical activity in a cohort of community-dwelling older men?
Falls affect approximately one third of community-dwelling older adults each year and have serious health and social consequences. Fear of falling (FOF) (lack of confidence in maintaining balance during normal activities) affects many older adults, irrespective of whether they have actually experienced falls. Both falls and fear of falls may result in restrictions of physical activity, which in turn have health consequences. To date the relation between (i) falls and (ii) fear of falling with physical activity have not been investigated using objectively measured activity data which permits examination of different intensities of activity and sedentary behaviour
Abstract DDT02-04: A novel PRMT5 inhibitor with potent in vitro and in vivo activity in preclinical lung cancer models
Abstract
PRMT5 is a type II methyltransferase that specifically adds methyl groups to arginine as a long-lasting post-translational modification. The PRMT5/MEP50 complex regulates a plethora of cellular processes, such as epigenetics and splicing, which are notable events involved in tumorigenesis. Although not frequently mutated or amplified in tumors, elevated PRMT5 protein levels in lung and hematologic cancers are correlated with poorer survival. The PRMT5 inhibitor JNJ-64619178 has been selected as a clinical candidate based on its high selectivity and potency (subnanomolar range) under different in vitro and cellular conditions, paired with favorable pharmacokinetics and safety properties. JNJ-64619178 binds into the SAM binding pocket and reaches the substrate binding pocket to inhibit PRMT5/MEP50 function in a time-dependent manner. Broad cell line panel profiling of JNJ-64619178 revealed a wide range of sensitivity, which is indicative of a genomic dependency instead of a general cytotoxic on-target consequence of PRMT5 inhibition. Further investigations indicate a synthetic lethal correlation between PRMT5 inhibition and key cancer driver pathways. JNJ-64619178, dosed orally (10 mg/kg, every day), showed selective and efficient blockage of the methylation of SMD1/3 proteins, which are crucial components of the spliceosome and substrates of PRMT5/MEP50. JNJ-64619178 also demonstrated tumor regression in a biomarker-driven human small cell lung cancer xenograft model (NCI-H1048) and prolonged tumor growth inhibition after dosing cessation. In rodent and nonrodent toxicology studies, a tolerated dose of JNJ-64619178 has been identified, with the observed toxicity consistent with on-target activity. In summary, JNJ-64619178 has a favorable preclinical package that supports clinical testing in patients diagnosed with lung cancer and hematologic malignancies.
