    Comparison of the limulus amebocyte lysate (LAL) and recombinant factor C (rFC) endotoxin bioassays: characterization of aerosols and settled dust on a sheep farm

    2011 Spring.Includes bibliographical references.The goals of this study are: 1) To optimize an extraction protocol for bulk sheep dust analysis by rFC; 2) To compare the traditional chromogenic LAL to the newer fluorometric rFC assay; 3) To compare the effects of the two most widely used extraction media, pyrogen-free water (PFW) and pyrogen-free water with 0.05% Tween 20 (PFW-Tween 20), on the assay outcomes; and 4) To characterize endotoxin exposure in a sheep farm environment. Settled and airborne dust samples were collected from a sheep farm for analysis. Settled dust was analyzed with endpoint fluorogenic rFC and kinetic chromogenic LAL in two different laboratories. A total of 34 stationary airborne dust samples (32 matched) were collected with Institute of Occupational Medicine (IOM) inhalable dust samplers, using polyvinyl chloride (PVC) filters. Airborne samples were analyzed with endpoint fluorogenic rFC and endpoint chromogenic LAL in the same laboratory. The results of the bulk dust analysis showed a significant difference in polystyrene (PS) tubes and polypropylene (PP) tubes used for extraction indicating that PS tubes yielded higher endotoxin levels (t = 3.82, p = 0.000). No difference in endotoxin levels was found for centrifugation and spike recovery was closest to 100% for extraction with PFW. The results of the airborne dust study showed a strong positive correlation between the rFC and LAL assays with PFW-Tween 20 extraction. All samples extracted in PFW-Tween 20 had a higher endotoxin recovery compared with those extracted in PFW for both the rFC and LAL assays

    Piezoelectric Potential Gated Field-Effect Transistor Based on a Free-Standing ZnO Wire

    We report an external force triggered field-effect based on a free-standing piezoelectric fine wire (PFW). The device consists of an Ag source electrode and an Au drain electrode at two ends of a ZnO PFW, which were separated by an insulating polydimethylsiloxane (PDMS) thin layer. The working principle of the sensor Is proposed based on the piezoelectric potential gating effect. Once subjected to a mechanical impact, the bent ZnO PFW cantilever creates a piezoelectric potential distribution across It width at Its root and simultaneously produces a local reverse depletion layer with much higher donor concentration than normal, which can dramatically change the current flowing from the source electrode to drain electrode when the device is under a fixed voltage bias, Due to the free-standing structure of the sensor device, it has a prompt response time less than 20 ms and quite high and stable sensitivity of 2%/mu N.     In an opinion drafted by Justice Pickering, the Nevada Supreme Court considered the scope of NRS 534.110(8), which allows the State Engineer to restrict the drilling of wells in specific basins if the State Engineer determines that the drilling of additional wells would result in undue interference with existing wells. At issue in this case was the State Engineer’s Order No. 1293A, which prohibits the drilling of additional domestic wells in the Pahrump Artesian Basin unless the drilling applicant relinquishes 2.0 acre-feet annually from an alternate source. Respondents, Pahrump Fair Water, LLC, along with its individual members (“PFW”) challenged the order, arguing that (1) the State Engineer lacked authority to issue the Order because substantial evidence did not support the conclusion that the drilling of wells would result in undue interference with existing wells and (2) the State Engineer violated due process by issuing the order without providing notice and a public hearing. The Court reversed the district court’s decision invalidating the Order and held that NRS 534.110(8) authorized the State Engineer’s Order based on substantial existing evidence, and that because water is a public resource—as opposed to a private resource—and because Nevada’s allocation system for water does not guarantee there will be enough to meet every request for it, that notice and a hearing prior to the State Engineer’s restriction was not necessary

    Quality based strategy: modelling for lean manufacturing

    The research develops and applies an integrated methodology for creating a Lean Manufacturing Environment in a traditional industry: the Portuguese Textile and Clothing Industry. This is achieved by developing a modelling tool using quality as a basis of performance assessment. In the context of the textile industry specific research objectives were: to evaluate current and potential application of Lean Manufacturing; to determine current business performance assessment criteria; to determine current practice in formulation of quality policy and metrics, and their impact on the effectiveness of new production technologies and techniques; to develop an integrated methodology to link the variables identified as important for the creation of a Lean Manufacturing environment; to apply the methodology in selected industrial test sites; and to derive quality system specifications which allow the realisation of Lean Manufacturing. The idea proposed in this thesis uses a quality approach to facilitate the application of Lean Manufacturing to the Textile and Clothing Industry. The author proposes a model for this evolution. Ile model developed includes objective variables (quality, productivity, delivery, cost, innovation, and time related elements), as well as subjective variables (flexibility, technological, and anthropocentric elements). It assesses the company performance from a Lean perspective, and not only from traditional Financial or Quality Assurance perspectives. The model development derived from applied research in 324 companies. A technique based on Data Envelopment Analysis was developed to analyse data from those companies. It assisted quantification of Lean Manufacturing assessment. The research revealed that a particular set of companies, which have implemented quality assurance systems, are closer to achieving Lean Manufacturing objectives. This research project concludes with a proposal for new quality system requirements specifically designed to facilitate the adoption of Lean Manufacturing
