Immobilization of flavan-3-ols onto sensor chips to study their interactions with proteins and pectins by SPR
Interactions between plant polyphenols and biomacromolecules such as proteins and pectins have been studied by several methods in solution (e.g. isothermal titration calorimetry, dynamic light scattering,nuclear magnetic resonance and spectrophotometry). Herein, these interactions were investigated inreal time by Surface Plasmon Resonance (SPR) analysis after immobilization of flavan-3-ols onto a sensor chip surface. (−)-epicatechin, (+)-catechin and flavan-3-ol oligomers with an average degree of polymerization of 2 and 8 were chemically modified using N-(2-(tritylthio)ethyl)propiolamide in order to introduce a spacer unit onto the catecholic B ring. Modified flavan-3-ols were then immobilized onto a carboxymethylated dextran surface (CM5). Immobilization was validated and further verified by evaluating flavan-3-ol interaction with bovine serum albumin (BSA), poly-l-proline or commercial pectins. BSA was found to have a stronger association with monomeric flavan-3-ols than oligomers. SPR analysis of selected flavan-3-ols immobilized onto CM5 sensor chips showed a stronger association for citrus pectins than apple pectins, regardless of flavan-3-ol degree of polymerization
Comparison of Wine Grape Qualities between Organic and Conventional Systems and Yearly Weather Patterns
Wine grapes may be susceptible to quality changes based on organic and conventional growing practices, as well as differences in yearly weather, which can impact wine quality. Four varieties of certified organic grapes at the Neely Kinyon Farm in Greenfield, Iowa in 2023 were tested for pH, titratable acidity, Brix, phenolics, anthocyanins, and yeast assimilable nitrogen and compared to organic grapes grown in 2021 under a different weather profile. Chemical analyses of organic Frontenac grapes grown at the Neely Kinyon Farm in 2023 and conventional Frontenac grapes grown at The Cellar Winery in Madrid, Iowa in 2023 were loosely compared for identifying potential differences in grape quality based on growing practices. Organic grapes harvested with higher rainfall in 2023 showed to have a more dilute level of acidity. Organic Frontenac grapes showed a greater phenolic level than conventional grapes, which is consistent with the literature. In conclusion, there appeared to be a correlation between grape quality and rainfall amount and organic production may lead to greater levels of phenolics in grapes

Thermodynamical parameters and kinetics of interactions between polyphenols (procyanidins and anthocyanins) and pectic fractions were defined by physico-chemical methods in solution and with a solid support. Experiments confronted major anthocyanins presents in blackcurrant or B-type procyanidins from apple with various degrees of polymerization to pectic fractions from apple pectins presenting different degree of methylation and different neutral sugar side chains.Interactions between anthocyanins and pectins are influenced by the glycosyl substituent and the number of hydroxyl groups in the B-ring of anthocyanins, and by the composition of pectins. The affinity constant of procyanidins – pectins interaction in solution are the highest when both the procyanidins degree of polymerization and the pectins degree of methylation are the highest. Moreover, the ramification state of pectins limits their association with procyanidins. Those interactions are due to hydrogen bonds and hydrophobic interactions.After chemical modifications of procyanidins and immobilization on a solid support, the resonance units obtained by surface plasmon resonance between (-)-epicatechin or dimer B2 and apple pectins are similar to those obtained with the prolin-rich protein IB5 and lower than with bovine serum albumin.Les paramètres thermodynamiques et les cinétiques d’interactions entre des polyphénols (des procyanidines et des anthocyanes) et des fractions pectiques ont été déterminés par des méthodes physico-chimiques en solution et sur support solide. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
