    No full text

A CRISPR-associated transposase presents null cargo integration efficiency when targeting a transcriptionally highly active region

    No full text
CRISPR-Cas effectors (e.g. Cas9) have been widely used to perform genetic knock-outs. Performing knock-ins however, remains challenging due to the inefficiency of the endogenous pathway cells use to integrate a donor genetic cargo into its genome (homology directed repair) when compared to other repair pathways like non-homologous end joining. CRISPR-associated transposases are complexes formed by a catalysis-deficient effector and a transposase. These complexes are able to sequester a transposon, localize a genomic target specified by a CRISPR RNA (crRNA), and integrate the transposon near the targeted site; thereby bypassing homology directed repair. In this study we aimed at developing a screening method using a CRISPR- associated transposase known as CAST, to detect integration events based on the disruption of lacZ. During the development, we found that CAST is unable to integrate a cargo in this highly active gene, most likely due to RNA polymerase-mediated dislodgment of the complex, and physical impediment for transposition proteins to reach the target DNA.Applied Sciences | Nanobiolog

We also provide unpublished information on its construction by master engraver Bartolomé Gonzalez de Espinosa, who finished it in the early 1660s, and on his golden work, accomplished between 1676 and 1678. In addition, unknown news about painters José Rodríguez and Juan Rodríguez de José are included. The former may be the author of the workroom’s splendid polychromatic cut. Some data on master chipper Domingo Ibáñez from Ciudad Rodrigo (Salamanca) are also provided, who was the designer of the stone pedestal in 1678.Junta de Extremadura (Consejería de Educación, Ciencia y Tecnología), «II Programa Regional de Investigación, Desarrollo Tecnológico e Innovación de Extremadura»: proyecto de investigación “El retablo en la Diócesis de Coria (Siglos XVI-XVIII). This essay is more or less a very boring analog game in paper, which is trying to find the similitude between ART and CONSCIOUSNESS, as well as the boundaries of legality of taking over or stealing someone else's work. The dynamic of this game of mine starts by choosing a lecture of any research and change some keywords to create a new research. The format of this essay suggests an interaction and will need an interaction of the reader with the essay itself in order to add more concepts and could make it more personal and transform the main subject of this essay. It does not necessarily have to be an art topic, so it is very open to any personal interest. There are no rules of this game, you can change the subject, the actors, the author or try to change it back to the original lecture. Everything is possible; at the end the reader could acquire by signing this essay or theory.[I examensarbetet ingår utställningen "YSO":] YSO explore metaphysical information in order to expand the idea of Art and consciousness, which experience and perception are both essential as a part of the information processing. Condensing this information to be able to exchange it on different states the Meta, Virtual and physical state. The formal aesthetic display of YSO is an interpretation of the different inputs objects or data that will or could construct an assumption with deliberate inputs through the senses to be able to interpret the correlation between Art and Consciousness. Material: Freezer, pick-up car, silicon form, water, wood, plastic sheet, tv, ice machine, media player and paint. Teknik: Installation/video/variable. Should I shrink or should I grow: cell size changes in tissue morphogenesis

    No full text
Cells change shape, move, divide and die to sculpt tissues. Common to all these cell behaviours are cell size changes, which have recently emerged as key contributors to tissue morphogenesis. Cells can change their mass—the number of macromolecules they contain—or their volume—the space they encompass. Changes in cell mass and volume occur through different molecular mechanisms and at different timescales, slow for changes in mass and rapid for changes in volume. Therefore, changes in cell mass and cell volume, which are often linked, contribute to the development and shaping of tissues in different ways. Here, we review the molecular mechanisms by which cells can control and alter their size, and we discuss how changes in cell mass and volume contribute to tissue morphogenesis. The role that cell size control plays in developing embryos is only starting to be elucidated. Research on the signals that control cell size will illuminate our understanding of the cellular and molecular mechanisms that drive tissue morphogenesis. The presentation of the authors' names and (or) special characters in the title of the pdf file of the accepted manuscript may differ slightly from what is displayed on the item page. The fiction Purple Hibiscos, by Nigerian author Chimamanda Adichie, portrays the effects of British colonization in Nigeria. From the narrator-character, Kambili, the reader has access to the marks of violence and injustice that fall on the characters, especially the female ones. In this context, the main objective of this research was to carry out a critical reading of the work, from the perspective of decolonial feminism, expression created by María Lugones based on the foundations of the coloniality of power established by Aníbal Quijano (2005) and developed by other contemporary authors, such as Ochy Curiel (2020), Suzana de Castro (2020), Lélia Gonzalez (2020), Sueli Carneiro (2011), Oyèrónkẹ́ Oyěwùmí (2021), bell hooks (2020) and Angela Davis (2017). Thus, we seek to understand how the book engenders and questions the memory shaped by coloniality, based on the experiences of the women portrayed in the literary work. Next, we seek to analyze the formation of the main female characters represented in the Adichian novel, namely Kambili, Beatrice, Amaka and Ifeoma, based on the proposal of decolonial feminism
