    The role of the non-canonical NF-κB pathway during steady state and autoimmune inflammation

    The NF-κB inducing kinase (NIK) is a primary stimulator of the non-canonical NF-κB signalling pathway, which regulates many aspects of the immune response. Mutations in the NIK/Map3k14 gene have been associated with various autoimmune diseases, including multiple sclerosis (MS) and mice with germline deletion of NIK are resistant to experimental autoimmune encephalomyelitis (EAE). We found that NIK has a minimal effect on the number and activation state of microglia in the CNS during steady state. However, the NIK∆CX3CR1 mice lack a specific CD169+ macrophage population in the spleen and LNs, suggesting an essential role for the non-canonical NF-κB pathway in the development and maintenance of cells. More interestingly, our research revealed that NIK∆CX3CR1 mice exhibit resistance to EAE, with fewer immune cells infiltrating the CNS and a reduced number of activated microglia. Intriguingly, when NIK is selectively deleted in microglia using the tamoxifen-inducible CX3CR1-CreERt2 line (NIKΔMG), the resistance against EAE diminishes. This suggests that the protective effects observed in NIK∆CX3CR1 mice are not due to the absence of NIK in microglia but rather in other peripheral CX3CR1-positive cells. We further observed a reduction in the priming of T cells in the dLNs before the onset of EAE in the NIK∆CX3CR1 mice. The myeloid cell compartment of NIK∆CX3CR1 mice displayed a dysregulation in genes associated with antigen presentation and migration, along with reduced IL-23 production. Given the crucial role of IL-23 in EAE, we hypothesised that the reduced levels of IL-23 in the NIK∆CX3CR1 mice could be responsible for protection against EAE. Indeed, stimulating cells from MOG-immunised NIK∆CX3CR1 mice with IL-23 before transferring them to RAG-/- mice partly restored the EAE in these mice. Our findings indicate that NIK plays a critical role in the development and function of certain macrophages during steady state and has a crucial role in the progression of EAE by regulating many aspects of myeloid cell activation. The multitude of subtle changes within the myeloid cell compartment, such as alterations in antigen presentation genes, reduced IL-23 production, and upregulation of anti-inflammatory genes, collectively contribute to the significant EAE resistance observed in our mice.     Interleukin-1 signaling in the blood-brain barrier influences the behavioral response to chronic social stress

    Chronic stress is a major risk factor for depression, leading to systemic immune activation and neuroinflammation, both of which contribute to the development of depressive symptoms. The blood-brain barrier (BBB), with endothelial cells as a central component, plays a crucial role in maintaining central nervous system (CNS) homeostasis by regulating the exchange of molecules and immune cells between the blood and CNS. However, the precise impact of chronic stress on BBB function and whether BBB components contribute to the establishment of depression-like behavior following stress remain unclear. Interleukin-1 (IL-1), a key pro-inflammatory cytokine, plays a central role in the development of stress-induced behavioral changes. Its receptor, IL-1R1, is primarily expressed by BBB endothelial cells (BECs) in the CNS, but its expression is restricted to a small subset of these cells under steady-state conditions. Chronic social defeat (CSD) stress has been shown to upregulate IL-1R1 expression on BECs, potentially driving BBB activation during stress. Although systemic endothelial IL 1R1 deletion has been shown to alleviate stress-induced behavioral symptoms, it remains unclear whether IL-1 signaling specifically in BECs mediates these effects. Using the Slco1c1-CreERT2 mouse strain to delete IL-1R1 selectively in BECs, this study aimed to investigate the role of IL-1 signaling in BECs during CSD and its contribution to depression-like behavior. Here, we show that IL-1 signaling in BECs is critical for mediating stress-induced behavioral dysfunction, as conditional deletion of IL-1R1 in BECs increased stress resilience and ameliorated behavioral changes following CSD. We found that IL-1R1⁺ BECs constitute a small, distinctly activated BEC subset under steady-state conditions. CSD stress further increased IL-1R1 expression on BECs, which was essential for their activation during stress. Notably, IL-1 signaling in BECs influenced behavior independently of peripheral immune cell infiltration or neuroinflammation, suggesting that alternative mechanisms are involved. While IL-1 signaling is a well-established key regulator of chronic stress responses, our results demonstrate that this effect is specifically mediated through BECs, highlighting the importance of the BBB in the chronic stress response. Taken together, our findings suggest that targeting IL-1 signaling in BECs could be a promising strategy for mitigating stress-induced neurovascular dysfunction and its associated behavioral consequences.Chronischer Stress ist ein wichtiger Risikofaktor für die Entwicklung von Depressionen. Die Blut-Hirn-Schranke (BHS) mit ihren Endothelzellen als zentralem Bestandteil steuert den Austausch von Molekülen und Immunzellen zwischen Blut und Zentralnervensystem (ZNS) und trägt somit maßgeblich zur Aufrechterhaltung des Gleichgewichts im ZNS bei. Allerdings ist bislang ungeklärt, wie chronischer Stress die Funktion der BHS beeinflusst und inwieweit ihre Bestandteile zur Entstehung depressiven Verhaltens nach Stress beitragen. Interleukin-1 (IL-1) ist ein wichtiges pro-entzündliches Zytokin mit einer zentralen Rolle in der Entstehung stressinduzierter Verhaltensstörungen. Sein Rezeptor, IL-1R1, wird im ZNS unter homöostatischen Bedingungen hauptsächlich von Endothelzellen der BHS exprimiert, jedoch nur von einem kleinen Anteil dieser Zellen. Es wurde gezeigt, dass „chronischer sozialer Niederlage“ (CSD)-Stress die IL-1R1-Expression in den BHS-Endothelzellen erhöht, was zur Aktivierung der BHS während Stress beitragen könnte. Die Deletion des IL-1R1 in Endothelzellen des gesamten Körpers konnte stressinduzierte Verhaltensstörungen teilweise verhindern. Allerdings ist bislang unklar, ob dieser Effekt durch IL-1-Signale speziell in Endothelzellen der BHS vermittelt wird. Zur Klärung dieser Frage verwendeten wir das Slco1c1-CreERT2-Mausmodell, das die gezielte Deletion von IL-1R1 in BHS-Endothelzellen ermöglicht. Unsere Ergebnisse zeigen, dass IL-1-Signale in den Endothelzellen der BHS eine zentrale Rolle in der Entwicklung stressinduzierter Verhaltensstörungen spielen, denn die Deletion von IL-1R1 in diesen Zellen verringerte die CSD-induzierten Verhaltensstörungen signifikant. IL-1R1⁺ Endothelzellen bildeten unter homöostatischen Bedingungen eine kleine Zellpopulation mit aktiviertem Phänotyp. CSD führte zu einer verstärkten IL-1R1-Expression in diesen Zellen, die essenziell für ihre Aktivierung während CSD-Stress war. Interessanterweise beeinflusste der IL-1-Signalweg in BHS-Endothelzellen das Verhalten unabhängig von der Infiltration peripherer Immunzellen oder einer generellen Neuroinflammation, was auf andere involvierte Mechanismen hindeutet. Während die Rolle des IL-1-Signalwegs in chronischem Stress bereits etabliert war, zeigen unsere Ergebnisse, dass diese Effekte unter anderem über BHS-Endothelzellen vermittelt werden und unterstreichen die Bedeutung der BHS in der Regulation der Stressantwort. Unsere Ergebnisse legen nahe, dass die gezielte Modulation von IL-1-Signalen in BHS-Endothelzellen eine vielversprechende Strategie zur Verhinderung stressinduzierter BHS-Dysfunktionen und der damit verbundenen Verhaltensveränderungen darstellen könnte.102 Seiten ; Illustrationen, Diagramm

    In toxic demyelination oligodendroglial cell death occurs early and is FAS independent

    Oligodendroglial cell death is a frequent phenomenon of many neurological diseases, e.g. in demyelinating diseases such as multiple sclerosis (MS). The underlying mechanisms are largely unknown. Here, we demonstrate that in the toxic demyelination cuprizone model, oligodendroglial cell death and downregulation of myelin genes start days after initiation of the cuprizone diet and weeks before demyelination is obvious. In early - but not in later - stages, dying oligodendrocytes express activated caspase 3, suggesting a switch from classical apoptotic pathways to caspase 3-independent mechanisms during the course of the cuprizone diet. The expression level of FAS in the corpus callosum, a cell death receptor crucial for oligodendroglial cell death in experimental autoimmune encephalomyelitis (EAE), correlates with the expression of activated caspase 3 in oligodendrocytes. However, mice lacking FAS in oligodendrocytes are not protected against cuprizone-induced oligodendroglial cell death, showing that FAS is dispensable for oligodendroglial cell death in the cuprizone model.     B cells modulate the homeostasis of splenic marginal zone antigen-presenting cells to promote anti-viral CD8+ T cell responses

    CMV gehört zur Spezies Familie der β-Herpesviridae, die eine chronische, asymptomatische Infektionen verursachen können. Eine Rekonstitution von CD8+ T-Lymphozyten korreliert dabei mit der Genesung von HCMV-Infektionen. Ebenso bietet die Transplantation von virusspezifischen CD8+ T-Zellen Schutz vor CMV-Infektionen bei immungeschwächten Mäusen. Im Gegensatz zu den T-Zellen ist die Rolle der B-Zellen bei der CMV-Infektionen jedoch deutlich weniger erforscht. B-Zellen spielen eine entscheidende Rolle bei der Sekretion von Antikörpern, tragen maßgeblich zur humoralen Immunantwort bei und dienen somit dem Schutz gegen virale Infektionen. Darüber hinaus sind B-Zellen entscheidend für die Entwicklung des funktionalen Aufbaus der Milz, insbesondere für die Aufrechterhaltung der sogenannten Marginalzone. In der MZ interagieren angeborene und adaptive Immunzellen mit durch Blut übertragenen Antigenen, um eine Immunreaktion unter anderem gegen virale Pathogene einzuleiten. Auch milz-residente APCs wie CD169+ marginale metallophile Makrophagen, SIGN-R1+ MZ-Makrophagen und cDC spielen eine wichtige Rolle in der Abwehr von Virusinfektionen. Diese Zellen internalisieren Antigene aus dem Blutkreislauf, degradieren sie und initialisieren somit eine adaptive Immunantwort. Veröffentlichungen zufolge sind marginale metallophile CD169+ Makrophagen der Zelltyp, der am meisten von Viren infiziert wird. Marginale metallophile CD169+ Makrophagen übertragen Antigene über das CD169-Molekül auf CD8+ cDCs, die darauf folgend eine wirksame CD8+ T-Zell-Antwort auslösen können39. cDC werden in die Untergruppen cDC1- und cDC2 unterteilt, können aber auch in weitere cDC-Untergruppen unterteilt werden, mit jeweils unterschiedlichen immunmodulatorischen Funktionen haben. Unter den CD8+ cDC1-Subpopulationen sind Langerin+cDC1, die sich vorwiegend in der milzspezifischen MZ befinden, die am stärksten im Auslösen einer CD8+-T-Zell-Immunantwort spezialisierte kreuzpräsentierende. Jedoch ist noch nicht bekannt, ob B-Zellen, über die in der MZ vorhandenen APC eine Rolle bei antiviralen CD8+ T-Zellantworten modulieren. Durch diese Studie stellte sich heraus, dass B-Zellen die virusspezifischen CD8+ T-Zell-Antworten begünstigen. Interessanterweise bietet die verringerte Antigenpräsentationskapazität der APC in Mäusen ohne B-Zellen eine vernünftige mechanistische Erklärung für dieses Phänomen. Wir zeigenen, dass B-Zellen durch die Expression von LTβ in der Lage sind, die Präsenz von marginalen metallophilen CD169+ Makrophagen in der Milz aufrechtzuerhalten. Darüber hinaus haben wir mit Hilfe der hochdimensionalen Durchflusszytometrie gezeigt, dass die Abwesenheit von B-Zellen und folglich eine Verringerung der marginalen metallophilen CD169+ Makrophagenpopulation zu einer Reduktion von Langerin+XCR1+ cDC1s führt, welche für das Auslösen einer MCMV-spezifischen CD8+ T-Zell-Antworten erantwortlich sind. Zusammenfassend lässt sich sagen, dass die hier vorgestellten Studien neue Erkenntnisse über die Funktion von B-Zellen im Kontext der antiviralen Immunität liefern. Neben dem Beitrag zur Virusbeseitigung durch die Produktion von Antikörpern und Zytokinen zeigen unsere Ergebnisse, dass B-Zellen in der Lage sind, virusspezifische CD8+ T-Zellantworte aufrechtzuerhalten, indem sie APC wie Makrophagen und dendritische Zellen modulieren kontrollieren. Wir hoffen, dass unsere Erkenntnisse tragen somit zum weiteren Verständnis der Funktion von B-Zellen in der virusspezifischen Immunantwort bei und bilden demnach Grundlagen für neue Therapieansätze zur Behandlung von Erkrankungen.IX, 88 Seiten ; Illustrationen, Diagramm

    Synthetic diets exacerbate auto-immunity in mice : EAE stage resolved proteomic immune cell characterization

    Western diet (WD)-associated non-communicable diseases (NCDs), including metabolic syndrome, and neurodegenerative conditions like multiple sclerosis (MS), are leading causes of death in the Western world. Although our knowledge of MS etiology has advanced, further insights into host-diet interactions are crucial to unravel the complex mechanisms driving MS. Additionally, our understanding of time-resolved specific contributions of myeloid cells during neurodegeneration is limited. We study how dietary intervention impacts experimental autoimmune encephalomyelitis (EAE) pathophysiology in mice. Intriguingly, our data show that synthetic Western diet (WD) or synthetic control diet (CD) exposure exacerbates EAE severity, compared to chow diet-fed mice. Rather than weight gain, we identify reduced fiber availability and consequently systemically reduced short-chain fatty acid (SCFA) levels as primary drivers of disease exacerbation. Pathophysiologically this is reflected by increased infiltration of Th17 and GM-CSF+ T cells into the spinal cord of synthetic diet-fed mice, along with decreased circulating Ly6Chi monocytes and a marked expansion of microglia and macrophages at the peak of EAE symptoms. Additionally, synthetic diets diminish the number of Tregs in the spinal cord during peak EAE. Supplementation with the SCFA butyrate significantly reduces EAE scores in WD-fed and, particularly, CD-fed mice. Finally, we use high-sensitivity liquid chromatography-tandem mass spectrometry (LC-MS/MS) to map the trajectory of myeloid cell-type specific protein expression throughout EAE. We identify Ly6Chi monocytes to acquire an EAE stage-specific proteome only upon spinal cord infiltration. Using lineage tracing to separate tissue-resident macrophages from invading, we reveal that microglia engage reparative metabolic pathways, while monocyte derived macrophages maintain inflammatory signaling even after clinical peak. We show the retention of myelin as well as lipid and cholesterol associated proteins in tissue-resident and peripheral phagocytes, and the acquisition of a foam-cell associated proteome. Treatment of mice with the cholesterol efflux transporter cyclodextrin reduces EAE severity. Consequently, our study highlights both cyclodextrin and butyrate as promising therapeutic candidates with potential translational significance for human health
