PharmDr. Petr Nachtigal, Ph.D. Title of Doctoral Thesis: Possible changes of endoglin expression in endothelial cells Endoglin (Eng) is a co-receptor for ligands of the Transforming Growth Factor β superfamily. Multiple studies have demonstrated that Eng is associated with metabolic syndrome-related pathological conditions, such as hypercholesterolemia, hyperglycemia, hypertension, or obesity. However, the exact role of Eng in these pathologies is still unknown. Therefore, the main purpose of this thesis was to elucidate and summarize the role of membrane and soluble Eng in the selected pathologies associated with metabolic syndrome. We demonstrated that soluble Eng (sEng) levels, together with plasma lipids and markers of inflammation, were reduced after every lipoprotein apheresis in patients with familial hypercholesterolemia. This result, in combination with knowledge acquired by the review article, indicates that increased sEng levels are associated with cardiometabolic disorders. Viaceré štúdie preukázali, že Eng je spojený s patologickými stavmi súvisiacimi s metabolickým syndrómom, ako sú hypercholesterolémia, hyperglykémia, hypertenzia alebo obezita. Avšak presná úloha Eng pri týchto patologických stavoch stále nie je známa. Preto bolo hlavným cieľom tejto práce objasniť a zhrnúť úlohu membránového a solubilného Eng vo vybraných patologických stavoch spojených s metabolickým syndrómom. Preukázali sme, že hladiny solubilného Eng (sEng), spolu s plazmatickými lipidmi a markermi zápalu, sa po každej aferéze lipoproteínov znížili u pacientov s familiárnou hypercholesterolémiou. Possibilities of influence of cardiovascular risk markers with a diet enriched with amaranth flour

Analysis of literary sources suggest that amaranth can meet these properties. Aim: The execution of literary investigation of the issue and experimental verification of lipid- lowering effect of amaranth flour in laboratory conditions. Methods: 50 (Experiment 1) or 40 (Experiment 2) The mice were divided into five (Experiment 1) or four (Experiment 2) groups fed by control mixture (C), high-energy diet (HED), high- energy diet with amaranth (HED+A) and in Experiment 1, by high-energy diet with amaranth flour and simvastatin (HED+A+S) or only with simvastatin (HED+S), and in Experiment 2 a by high-energy diet with squalene (HED+S). Weight gains were measured and TC, LDL and HDL cholesterol determined. Analýza literárních zdrojů naznačuje, že amarant může tyto vlastnosti splňovat. Cíle práce: Provedení literárního šetření dané problematiky a experimentální ověření literaturou avizovaného hypolipidemického působení amarantu v laboratorních podmínkách. Metodika: 50 (Pokus 1) respektive 40 (Pokus 2) myší bylo rozděleno do pěti (Pokus 1) respektive čtyř (Pokus 2) skupin krmených kontrolní směsí (C), vysokoenergetickou dietou (HED), vysokoenergetickou dietou s amarantem (HED+A) a v Pokusu 1 dále vysokoenergetickou dietou s amarantovou moukou a simvastatinem (HED+A+S) či pouze se simvastatinem (HED+S) a v Pokusu 2 vysokoenergetickou dietou se skvalenem (HED+S). Myším byly měřeny hmotnostní přírůstky a stanoven celkový (TC), LDL a HDL cholesterol. PharmDr. Petr Nachtigal, Ph.D. Title of Doctoral Thesis: Pharmacological influence on atherogenesis in experimental animal models of atherosclerosis Atherosclerosis is a slow inflammatory process in arterial walls which is, as a part of cardiovascular diseases, the main source of morbidity and mortality in developed countries. However, searching for the possibilities of atherosclerosis treatment requires detailed knowledge of pathogenesis of the disease itself. The use of mouse models offers one of the possibilities of studying the pathogenesis of atherosclerosis, which enables a number of interventions that would not be acceptable in human medicine. By using the high content lipid diet, we are able to induce the changes of lipid spectrum in mice. Moreover, if a targeted genetic manipulation is used, one can reach considerably advanced plaques in a relatively short time period. It is also possible to interfere in the process of atherogenesis in mouse models by using some hypolipidemic drugs, for example from the statin group that were represented in our studies by atorvastatin. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases.
