    Saprophytic Actinomycetes Promote Nodulation in Medicago sativa-Sinorhizobium meliloti Symbiosis in the Presence of High N

    Saprophytic rhizoactinomycetes isolated from the root nodule surface of the nitrogen-fixing actinorhizal plant Discaria trinervis, Streptomyces MM40, Actinoplanes ME3, and Micromonospora MM18, previously shown to stimulate nodulation in Frankia-Discaria trinervis symbiosis, were assayed as co-inoculants with Sinorhizobium meliloti 2011 on Medicago sativa. When plants were fertilized with a low level of N (0.07 mM), the inoculation of the actinomycetes alone did not show any effect on plant growth. Meanwhile, when actinomycetes were co-inoculated with S. meliloti, nodulation and plant growth were significantly stimulated compared to plants inoculated with only S. meliloti. The analysis of nodulation kinetics of simultaneously or delayed co-inoculations suggests that the effect of the actinomycetes operates in early infection and nodule development counteracting the autoregulation of nodulation by the plant. Because the actinomycete effect was found in the symbiotic nitrogen-fixing state of the plant, we investigated the effects of the actinomycetes, in single inoculation or co-inoculation with S. meliloti, on plants grown under a high level of N (7 mM) that was inhibitory for nodulation by S. meliloti. The inoculation of the actinomycetes alone did not show any effect on plant growth although high N was available. Unexpectedly, the co-inoculation of actinomycetes with S. meliloti on plants grown with high N (7 mM) significantly stimulates nodulation, clearly counteracting the inhibition of nodulation by high N. These results corroborate that the interaction of rhizoactinomycetes would interfere with the autoregulation of nodulation in alfalfa mediated by high N, opening new research lines of potential agronomical applications.Fil: Solans, Mariana.     Infectivity of soilborne Frankia and mycorrhizae in Discaria trinervis along a vegetation gradient in Patagonian soil

    The infective capacities of the nitrogen fixing Actinomycete Frankia and arbuscular mycorrhizal fungi from soils near watercourses, along a vegetation gradient, were studied using plant bioassays. Frankia and arbuscular mycorrhizas capable of infecting Discaria trinervis were found at seventeen sites sampled. More specific enumeration of the infective capacities of both microorganisms in relation to environmental factors was performed in seven representative soils of the analysed vegetation zones (rainforest, xeric forest and steppe) using the most probable number method. The highest nodulation capacities ranged from 340 infective units g–1 soil, in a steppe marsh devoid of actinorhizas, to 61 in a coastal actinorhizal scrub (in xeric forest). The highest number of infective mycorrhizal units – also found in marsh – was 145. In general, rainforest soils had the lowest values for both microorganisms.Infective units of Frankia and arbuscular mycorrhizal fungi in soil were positively correlated (r = 0.89, P < 0.05). Both soilborne symbionts showed the highest infective capacity in semi-arid conditions nearby watercourse and at the valley bottom location. Tripartite symbiosis was effective in plants inoculated with steppe and xeric forest soils and plants inoculated with Frankia BCU110501 and Glomus mosseae.     Polypropylene degrading actinobacteria

    La acumulación de polímeros sintéticos es un gran problema de contaminación ambiental, ya que no existen métodos eficientes para su eliminación segura; sin embargo, hay evidencias sobre su degradación por actinobacterias. En este trabajo se evaluó la capacidad de degradación de actinobacterias aisladas de suelo del vertedero de Bariloche, sobre tres biopolímeros y un polímero sintético. Por un lado, se estudió la degradación de polímeros naturales (almidón, hemicelulosa, celulosa) en medio sólido, y por otro el otro se evaluó la degradación de láminas de polipropileno inoculadas con los aislamientos y cultivadas en medio líquido durante seis semanas a 37 ºC. Luego se midió la pérdida de peso en las láminas y se estudió la colonización de la superficie del polipropileno mediante microscopia óptica y electrónica. Los datos fueron analizados con un ANOVA de dos vías y la prueba de Tukey. De los 35 aislamientos totales, el 88 % correspondió al género Streptomyces y el 3 % a los demás géneros (Actinomadura, Pseudonocardia, Saccharomonospora y Thermoactinomyces). El 63 % de los aislamientos tuvo la capacidad de degradar almidón, el 86 % hemicelulosa, el 34 % celulosa microcristalina (exoglucanasas) y el 88 % carboximetilcelulosa (endoglucanasas). Se observó colonización de la superficie de polipropileno en todos los tratamientos. Dos de las cepas estudiadas (Streptomyces sp. MP32 y Actinomadura sp. MP5) redujeron el peso de las láminas de polipropileno (p ≤ 0.05). Estos resultados preliminares muestran la capacidad de las actinobacterias para colonizar la superficie del polipropileno, con potencial uso en suelos o ambientes contaminados por plásticos.The accumulation of synthetic polymers is a great environmental contamination issue, since there are no efficient methods to dispose them safely; however, there is evidence about the degradation of plastics by actinobacteria. In this work we tested the degradation capacity of tree biopolymers and a synthetic polymer by actinobacteria isolated from the soil of a landfill in Bariloche. On the one hand, we studied the degradation of natural polymers (starch, hemicellulose, cellulose) in a solid medium, and on the other hand, we studied the degradation of polypropylene sheets inoculated with the isolates and cultured in liquid medium for six weeks at 37 ºC. We then measured weight loss in the sheets and studied colonization of the polypropylene surface using light and electron microscopy. The data were analyzed with a two-way ANOVA and Tukey’s test. Of the 35 total isolates, 88 % corresponded to the Streptomyces genus and 3 % to the rest of genera (Actinomadura, Pseudonocardia, Saccharomonospora and Thermoactinomyces). Of the isolates, 63 % were able to degrade starch, 86 % hemicellulose, 34 % microcrystalline cellulose (exoglucanases), and 88% carboxymethyl cellulose (endoglucanases). Colonization of the polypropylene surface was observed in all treatments. Two of the strains studied, Streptomyces sp. MP32 and Actinomadura sp. MP5 reduced the weight of the polypropylene sheets (p ≤ 0.05). These preliminary results show the ability of actinobacteria to colonize the polypropylene surface, with potential use in soils or environments contaminated by synthetic polymers.Fil: Boenel, Micaela.     La simbiosis actinorrícica en las Rhamnaceas del Parque Reserva Nacional Nahuel Huapi

    En esta tesis se estudian diferentes aspectos de la simbiosis actinorrícica, producida con especies de la familia Rhamnaceae, que crecen en el Parque y Reserva Nacional Nahuel Huapi (PNNH). Se efectuó un relevamiento de la nodulación, en las especies Discaria chacaye (G. Don) Tort., Discaria articulata (Phil.) Miers, Discaria trinervis (Hook. et Arn.) Reiche, y Colletia hystrix Clos., en distintas zonas de vegetación del PNNH. Se halló nodulación en todos los ejemplares colectados, por lo tanto, se verificó que la presencia e infectividad del actinomicete simbiótico Frankia es un factor constante en la región investigada. Los nódulos son del tipo coraloide y sus lóbulos poseen un meristema apical y un cilindro vascular central rodeado por células parenquimáticas, muchas de las cuales de hallan hipertrofiadas con el endófito Frankia en su interior. Los nódulos son del tippo "sp - " (sin esporangios). Se analizó el patrón espacial nodular en plantas de D. trinervis, mediante métodos estadísticos y de arquitectura radical. Se constató una disposición agrupada de nódulos, y una falta de correlación entre le número de nódulos y entrenudos de la raíz principal. Los parámetros de la arquitectura radical solo permitieron determinar la topología de las raíces. Esta indicó que la eficiencia del sistema radical para la adquisición de recursos es intermedia entre los modelos de suelos pobres y los de suelos fértiles. Mediante el análisis del número y área de las células huéspedes de Frankia, se estableció una caracterización anatómica de los nódulos de C. hystrix, D. chacaye y D. trinervis, que indicó los efectos estacionales en la simbiosis. El mayor desarrollo de células huéspedes de Frankia, coincidió con las estaciones de crecimiento activo de las plantas y en invierno se observó la dormición de los nódulos.     Isolation and characterization of an endolichenic actinobacterium from the lichen thallus of Pseudocyphellaria berberina

    Recent research has shown that lichenized fungi, which constitute a symbiotic association between a fungus and an algae and/or cyanobacteria, could be a source of biotechnologically interesting microorganisms, including actinobacteria. Studies conducted on lichenized microecosystems in the Andean region (Patagonia, Argentina) showed the isolation of twenty-three strains of actinobacteria, belonging to different genera, coming from the lichen Pseudocyphellaria berberina (Ascomycota). The aim of this work was to complement these studies by means of a polyphasic approach (morpho-physiological, chemical and molecular characterization) in order to describe a new endolichenic Pseudonocardia (Actinobacteria) strain BCRU MITI 22 (= DSM 46833). The study of cultivable actinobacteria isolated from lichen could contribute to a better interpretation of the physiological functions of these organisms within the “lichen microbiome”.Fil: Vobis, Gernot.     Potential biocontrol actinobacteria: Rhizospheric isolates from the Argentine Pampas lowlands legumes

    Control of fungal plant diseases by using naturally occurring non-pathogenic microorganisms represents a promising approach to biocontrol agents. This study reports the isolation, characterization, and fungal antagonistic activity of actinobacteria from forage soils in the Flooding Pampa, Argentina. A total of 32 saprophytic strains of actinobacteria were obtained by different isolation methods from rhizospheric soil of Lotus tenuis growing in the Salado River Basin. Based on physiological traits, eight isolates were selected for their biocontrol-related activities such as production of lytic extracellular enzymes, siderophores, hydrogen cyanide (HCN), and antagonistic activity against Cercospora sojina, Macrophomia phaseolina, Phomopsis sp., Fusarium oxysporum, and Fusarium verticilloides. These actinobacteria strains were characterized morphologically, physiologically, and identified by using molecular techniques. The characterization of biocontrol-related activities in vitro showed positive results for exoprotease, phospholipase, fungal growth inhibition, and siderophore production. However, none of the strains was positive for the production of hydrogen cyanide (HCN). Streptomyces sp. MM140 presented the highest index for biocontrol, and appear to be promising pathogenic fungi biocontrol agents. These results show the potential capacity of actinobacteria isolated from forage soils in the Argentine Pampas lowlands as promising biocontrol agents, and their future agronomic applications.Fil: Solans, Mariana.     Se aislaron veintitrés (23) cepas de actinomicetes cultivables de la especie corticícola Pseudocyphellaria berberina presente en la selva templada valdiviana, Argentina, veitiún (21) fueron endoliquénicas y dos (2) ectoliquénicas. Se identificaton diecisiete (17) cepas de los géneros, Actinoplanes (2), Dactylosporangium (1), aff. Pseudonocardia (1), Micromonospora (6), aff. Micromonospora (1), Streptomyces (5) y Streptosporangium (1). Este es el primer registro para la Argentina de actinomicetes aislados de hongos liquenizados.Cultivable actinomycetes associated to lichenized microecosystems from the temperate Valdivian rainforests, Argentina. Twenty-three (23) cultivable strains of actinomycetes were isolated from the corticolous lichen species Pseudocyphellaria berberina present in the Valdivian temperate rainforest, Argentina, being twenty-one (21) endolichenic and two (2) ectolichenic. Seventeen (17) strains were identified as belonging to genera Actinoplanes (2), Dactylosporangium (1), aff. Pseudonocardia (1), Micromonospora (6), aff.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
