The roles of intratumour heterogeneity in the biology and treatment of pancreatic ductal adenocarcinoma
No full textIntratumour heterogeneity (ITH) has become an important focus of cancer research in recent years. ITH describes the cellular variation that enables tumour evolution, including tumour progression, metastasis and resistance to treatment. The selection and expansion of genetically distinct treatment-resistant cancer cell clones provides one explanation for treatment failure. However, tumour cell variation need not be genetically encoded. In pancreatic ductal adenocarcinoma (PDAC) in particular, the complex tumour microenvironment as well as crosstalk between tumour and stromal cells result in exceptionally variable tumour cell phenotypes that are also highly adaptable. In this review we discuss four different types of phenotypic heterogeneity within PDAC, from morphological to metabolic heterogeneity. We suggest that these different types of ITH are not independent, but, rather, can inform one another. Lastly, we highlight recent findings that suggest how therapeutic efforts may halt PDAC progression by constraining cellular heterogeneity
Behrens’ narrative of the discovery of Easter Island:Two editions, two personalities, two realities
No full textThis article is dedicated to the figure of Carl Friedich Behrens, a member of the Dutch expedition led by Jacob Roggeveen, who re-discovered Easter Island in 1722. Behrens, a German soldier serving on one of the ships, left a narrative describing the whole journey. The first edition was published in 1737 followed, among others, by a re-published edition made by German anthropologist Hans Plischke that was published in 1923. The important thing is that this version differs from the original to a great extent and the editor did not account for the changes he had introduced into the text: besides grammar and orthography modernization, he omitted certain portions, misinterpreted other ones and added some comments without marking them as his own. As a result, the narrative gives an impression of having been written by another author; Behrens appears as a person with a different character and attitude, weaker, less convincing and even less trustworthy than he really was. This article presents numerous examples of the distortions as a warning against making a scientific or an anthropological use of unreliable editions of source texts, as this may wield a negative influence upon our view and interpretation of the culture we are analyzing

Can production and trafficking of illicit drugs be reduced or merely shifted ?
No full textThe production of cocaine and heroin, the two most important drugs economically, has been concentrated in a small number of poor nations for 25 years. A slightly larger number of developing nations have been affected by large-scale trafficking in these two drugs. This paper reviews what is known about drug control programs and considers non-traditional options. The usual array of programs for suppressing drug problems, enforcement, treatment, harm reduction and prevention have been assessed almost exclusively in wealthy nations. Although treatment has been shown to be cost-effective, it is of minimal relevance for reducing the drug problems of nations such as Afghanistan, Colombia, Mexico or Tajikistan, which are primarily harmed by production and trafficking rather than consumption. Efforts to reduce drug production and trafficking have not been subject to systematic evaluation but the best interpretation of the available evidence is that they have had minimal effect on the quantities produced or trafficked. It is reasonable to conclude that international drug control efforts can do more to affect where these drugs are produced rather than the quantity. If that is the case, and given that spreading a specific level of production or trafficking to more rather than fewer nations probably decreases global welfare, it may be appropriate to consider a less aggressive stance to current producers and to make strategic decisions about the location of an industry producing a global bad.Crime and Society,Economic Theory&Research,Markets and Market Access,Alcohol and Substance Abuse,Post Conflict Reconstruction
