    Beliefs about control over gambling among young people, and their relation to problem gambling.

    The aim of this study was to assess the association between beliefs about 2 types of control--(a) illusion of control and (b) internal locus of control--and gambling frequency-problem gambling among young people age 14 to 25 years (435 males, 577 females, and 5 unreported gender). A revised version of the South Oaks Gambling Screen and measures of gambling frequency and gambling beliefs were administered. Results indicated that irrational control beliefs were strongly associated with problem gambling. Young problem gamblers were more likely to believe that they needed money and that gambling would provide it. In addition, young problem gamblers had more faith in their ability to manipulate chance and "beat the system."     Dissertação (mestrado) - Universidade Federal de Santa Catarina, Centro de Ciências da Saúde, Programa de Pós-Graduação em Enfermagem, Florianópolis, 2009.Este estudo trata da elaboração de um modelo que contempla o fenômeno do cuidado em Unidade de Terapia Intensiva Neonatal a partir da compreensão do ser e fazer enquanto enfermeiro neste sistema complexo. Tal compreensão aborda as transformações ocorridas desde a criação das UTINs até a atualidade, vislumbrando sua busca pela integralidade ao oferecer um cuidado sensível e compartilhado. O referencial metodológico da Teoria Fundamentada nos Dados (Grounded theory) associado ao Paradigma da Complexidade de Edgar Morin foi utilizado na análise e na construção do Modelo Teórico: CUIDANDO DO RECÉM-NASCIDO EM UTIN: Convivendo com a fragilidade do viver/sobreviver à luz da complexidade. Participaram da pesquisa 11 sujeitos, entre enfermeiros e mães com bebês internados na UTIN. Utilizou-se a entrevista aberta para coleta dos dados. A análise comparativa dos dados subsidiou o processo de codificação, amostragem, saturação teórica, ordenação e integração dos dados. Os dados foram inseridos no software NVIVO, dando seqüência na organização e agrupamento dos mesmos. A organização dos códigos foi feita de acordo com suas propriedades e, então, agrupados em códigos preliminares, subcategorias e categorias. Através das inter-relações entre categorias, emergente dos dados coletados, identificou-se a categoria central, eixo norteador do Modelo Teórico, e integradora das categorias analíticas denominada: Convivendo com a fragilidade do viver/sobreviver: cuidado altamente complexo, sensível, singular e compartilhado em torno da qual giram as demais categorias: Buscando conhecimentos e competências; Gerenciando o cuidado na UTIN e Vivenciando as singularidades na UTIN. Neste contexto, o cuidado em UTIN se fortalece como uma das áreas da Enfermagem em constante desenvolvimento, visando conciliar os avanços tecnológicos importantes para o sobreviver do bebê prematuro com abordagens que valorizam as inter-relações em seu quotidiano, de modo sistêmico. Busca atuar nas diversas esferas do cuidado complexo em saúde, desde os serviços de apoio da instituição hospitalar e seus gestores até a academia, visando evitar atuações compartimentadas e isoladas, integrando e aplicando conhecimentos científicos, com ganhos para a prática profissional. Oferecer suporte tecnológico já não basta. A pesquisa ressalta que é necessário exercitar as potencialidades já inatas dos profissionais de enfermagem e caminhar para o encontro de outras, um verdadeiro convite a novas percepções de cuidar do neonato, sua família e os membros deste sistema complexo, numa constante inquietação e adaptação para as demandas que surgem. Assim, as indagações norteadoras desta pesquisa convidam para a ampliação de novos horizontes de investigação, de forma que as possibilidades de estudos indicadas permitam o aprofundamento no âmbito das organizações de saúde e da academia.This study deals with the elaboration of a model which contemplates the phenomenon of care in a Neonatal Intensive Care Unit (NICU), based on the comprehension of the human being and human actor as a nurse in this complex system. Such comprehension encompasses the transformations which have occurred since the creation of the NICUs, discerning its search for the entirety to offer sensible and shared care. The methodological reference of the Grounded Theory associated with Edgar Morins Complexity Theory was utilized in analysis and the construction of the Theoretical Model: CARING FOR THE NEWBORN IN THE NICU: Living with the fragility of living/surviving in the light of complexity. The sample was composed of 11 subjects, including nurses and mothers with newborns in the NICU. Open interviews were used in order to collect data. Comparative data analysis subsidized the data codification, sampling, theoretical saturation, ordering, and integration processes. Data was inserted into NVIVO software, providing a sequence in data organization and grouping. Organizing the codes was done according to their properties and thus, grouped in preliminary codes, subcategories, and categories. Through the relationships in these categories which emerged from the data collected, the central category, the guiding axis of the Theoretical Model and the integrating factor of the analytical categories was identified, denominated: Living with the fragility of living/surviving: highly complex, sensitive, singular, and shared care around which are the following categories: Seeking knowledge and competencies; Managing NICU care; and Living the singularities of the NICU. In this context, NICU care is strengthened as one of the Nursing areas in constant development, seeking to conciliate important technological advances towards premature newborn survival with the approaches which systematically value day-to-day relationships. It seeks to enact in the diverse spheres of complex health care, from the support services of the hospital institution and its management to academia, searching to avoid fragmented and isolated care through integrating and applying scientific knowledge with gains for professional practice. Offering technological support is no longer enough. Research highlights that it is necessary to exercise already innate potential in nursing professionals and work towards finding others, a true invitation towards new perceptions for care for the newborn, his/her family, and the members of this complex system in a constant unrest and adaptation of demands which may arise. Thus, the guiding questions of this study invite towards the amplification of new investigative horizons, in such a manner that the possibilities for suggested studies permit more profound in the realm of health care and academic organizations

    Avaliação da ação citotóxica de cardenolídeos em células tumorais

    Tese (doutorado) - Universidade Federal de Santa Catarina, Centro de Ciências da Saúde, Programa de Pós-Graduação em Farmácia, Florianópolis, 2015.Os cardenolídeos, tais como digoxina e oubaína, são conhecidos por sua eficácia no tratamento da insuficiência cardíaca congestiva e como fármacos antiarrítmicos. Recentemente, foram detectadas novas atividades farmacológicas para esses "antigos" fármacos. Tendo em vista o crescente interesse na pesquisa e desenvolvimento desta classe de compostos como potenciais quimioterápicos, frente a diversas linhagens celulares tumorais, o presente trabalho de tese objetivou avaliar os efeitos citotóxicos em células tumorais de três cardenolídeos (glucoevatromonosídeo, digitoxigenina monodigitoxosídeo e convalotoxina), previamente selecionados pela sua potente ação citotóxica. Inicialmente, eles foram avaliados em linhagens celulares tumorais de diferentes origens e todos demonstraram uma potente ação, em concentrações nanomolares, em todas as linhagens testadas, especialmente nas células de tumor de pulmão (A549). Essa linhagem tumoral foi então selecionada para a continuação dos experimentos para detectar o tipo de morte celular causada por esses três cardenolídeos. O glucoevatromonosídeo (GEV), composto mais promissor, também foi investigado em células U937 (linfoma histiolítico). Todos os três causaram bloqueio da fase G2/M, enquanto que a convalotoxina (CON) aumentou o número de células em subG0 do ciclo celular. O GEV foi capaz de inibir a expressão de importantes proteínas relacionadas ao ciclo celular, como ciclina B1 e p53 em A549. Ainda, esse composto causou bloqueio em subG0 em células U937, demonstrando um efeito dependente do tipo celular. O efeito de morte celular causado pelo GEV também foi tipo celular dependente, já que foi observada morte celular pela ação de caspases nas células U937 e independente da ação das caspases em células A549. A digitoxigenina monodigitoxosídeo não apresentou efeito significativo de morte celular em células A549. A CON aumentou o número de núcleos picnóticos e células Anexina-V positivas, configurando morte celular apoptótica. Alem disso, os três compostos foram capazes de inibir a migração e invasão celulares, em células A549, bem como de reduzir a expressão das proteínas: MMP2, MMP9 e FAK (proteína de adesão focal), que são essenciais no processo de metástase. Além disso, o GEV foi capaz de inibir a expressão de importantes cinases, geralmente super expressas em células tumorais. O conjunto desses dados sugere que estes compostos, especialmente o GEV, podem ser considerados candidatos promissores para o desenvolvimento de uma forma farmacêutica a ser usada no tratamento do câncer de pulmão.Abstract : Cardenolídeos such as ouabain and digoxin are known for their efficacy in treating congestive heart failure as antiarrhythmic drugs. Recently, new pharmacological activities have been found (antiviral and antitumor) to these "old" drugs. Given the growing interest in research and development of this group of compounds, as potential chemotherapeutic agents, this work aimed to evaluate the cytotoxic effects on tumor cells of three cardenolides (glucoevatromonoside, digitoxigenin monodigitoxoside and convallatoxin) previously selected for its potent cytotoxic action. Initially, they were screened in tumor cell lines of different origins and all of them showed potent action at nanomolar concentrations in all cell lines tested, particularly in lung tumor cells (A549). This cell line was then selected for further investigation to detect the kind of cell death caused by these three cardenolides. Glucoevatromonoside (GEV) was the most promising compound, and also investigated in U937 cells (histiocytic lymphoma). All these compounds block G2/M phase, whereas the convallatoxin increased the number of cells in subG0 phase. GEV was able to inhibit the expression of important proteins related to cell cycle, such as Cyclin B1 and p53. Beyond that, this compound caused cell cycle blockage in subG0 phase in U937 cells, demonstrating dependent cell type effect. The effect of cell death caused by GEV was also cell type dependent. In U937 cells, GEV showed a caspase dependent cell death while it is independent of caspase in A549 cells. Digitoxigenin monodigitoxoside did not show a significant percentage of cell death in A549 cells. Convallatoxin increased the number of pyknotic nuclei and Annexin-V positive cells, setting apoptotic cell death in A549 cells. Furthermore, the three compounds are capable of inhibiting cell migration and invasion in A549 cells as well as to reduce the expression of some proteins as MMP2, MMP9 and FAK (focal adhesion protein), which are essential in the process of metastasis. Moreover, GEV was able to inhibit the expression of important kinases, usually over expressed in tumor cells.. Taken together, these data suggest that these compounds, especially GEV can be considered promising candidates for the development of a pharmaceutical form to be used in the treatment of lung cancer

    As part of a program aimed at metal-catalyzed oxidative transformations of molecules with carbon–metalloid bonds, the synthesis of α-borylated ketones is reported via regioselective TBHP-mediated Wacker-type oxidation of N-methyl­imino­diacetic acid (MIDA)-protected alkenylboronates. The observed regioselectivity correlates with the hemilabile nature of the B–N dative bond in the MIDA boronate functional group, which allows boron to guide selectivity through a neighboring group effect

    Exercise intensity and the protection from postprandial vascular dysfunction in adolescents

The final version is available from American Physiological Society via the DOI in this recordBACKGROUND: Acute exercise transiently improves endothelial function, and protects the vasculature from the deleterious effects of a high fat meal (HFM). We sought to identify whether this response is dependent on exercise intensity in adolescents. METHODS: Twenty adolescents (10 male, 14.3 ± 0.3 y) completed three 1-day trials: 1) rest (CON); 2) 8x1 min cycling at 90% peak power with 75s recovery (high-intensity interval exercise; HIIE); 3) cycling at 90% of the gas exchange threshold (moderate-intensity exercise; MIE) one hour before consuming a HFM (1.50 g∙kg(-1) fat). Macrovascular and microvascular endothelial function were assessed before and immediately after exercise, and three hours after the HFM by flow mediated dilation (FMD) and laser Doppler imaging (peak reactive hyperaemia; PRH). RESULTS: FMD and PRH increased one hour after HIIE (P<0.001, ES=1.20 and P=0.048, ES=0.56) but were unchanged after MIE. FMD and PRH were attenuated three hours after the HFM in CON (P<0.001, ES=1.78 and P=0.02, ES=0.59). FMD remained greater three hours after the HFM in HIIE compared to MIE (P<0.001, ES=1.47) and CON (P<0.001, ES=2.54), and in MIE compared to CON (P<0.001, ES=1.40). Compared to CON, PRH was greater three hours after the HFM in HIIE (P=0.02, ES=0.71) and MIE (P=0.02, ES=0.84), with no differences between HIIE and MIE (P=0.72, ES=0.16). Plasma [triacylglycerol] and [total antioxidant status] were not different between trials. CONCLUSIONS: Exercise intensity plays an important role in protecting the vasculature from the deleterious effects of a HFM. Performing HIIE may provide superior vascular benefits than MIE in adolescent groups
