    Osteoporose is een aandoening die niet alleen vrouwen maar ook mannen tr eft. Alhoewel de incidentie aan osteoporotische fracturen lager ligt bij mannen, verdient deze aandoening ook bij mannen de nodige aandacht aang ezien ze gepaard gaat met een hogere morbiditeit, mortaliteit en medisch e kost. Het ontstaan van osteoporose wordt niet alleen bepaald door verl ies van botmassa op oudere leeftijd, maar ook door de opbouw van botmass a tijdens de groei. Want hoe hoger de botmassa bereikt op jong volwassen leeftijd, hoe minder kans men maakt om op oudere leeftijd osteoporose t e ontwikkelen. Algemeen bouwen mannen een stevigere en bredere botstruct uur op die aan de buitenzijde gekenmerkt wordt door een bredere cortex e n aan de binnenzijde door een groter trabeculair botvolume. Beiden drage n bij tot de sterkte van het bot. Het ophelderen van de mechanismen betr okken in zowel de opbouw als het behoud van het mannelijk skelet kan daa rom bijdragen tot efficiëntere behandelingswijzen voor osteoporose bij d e man. De productie van geslachtshormonen [androgenen (bijv. testosterone) en o estrogenen (bijv. 17ß-oestradiol)] aan de start van de puberteit spelen een belangrijke rol in de sterke toename van bot tijdens deze periode. M aar ook groeihormoon (GH) en insulin-like growth factor-I (IGF-I), ook w el de GH/IGF-I as genoemd, zijn essentieel. Bovendien zouden beide syste men, die werken via hun respectievelijke receptoren, kunnen interageren tijdens de puberteit om zo een maximale stimulatie van skeletale groei t ot stand te brengen. Naast hun werking tijdens de groei, zijn geslachtsh ormonen eveneens belangrijk voor het behoud van het skelet op volwassen leeftijd. Androgenen worden traditioneel verantwoordelijk geacht voor de opbouw va n een bredere botstructuur bij de man en worden als dé versterkende fact or van het mannelijk bot beschouwd. Maar oefenen androgenen hun werking direct uit door stimulatie van hun receptor? Of kan hun werking ook toeg eschreven worden aan aromatisatie, een proces waarbij androgenen in oest rogenen worden omgezet? Deze vragen werden beantwoord door aan jonge mui zen zonder functionele androgeen receptor, zogenaamde androgeen receptor knockout muizen, ofwel aromatiseerbaar testosterone ofwel niet-aromatis eerbaar dihydrotestosterone toe te dienen. Of de werking van testosteron e kan verhinderd worden door de omzetting tot oestradiol te blokkeren we rd nagegaan door naast testosterone ook een aromatase inhibitor toe te d ienen. Zo blijkt de opbouw van trabeculair bot, dat een soort aaneenscha keling of “netwerk” van botpartikels (de trabekels) is, volledig afhanke lijk te zijn van androgeenwerking die verloopt via de androgeen receptor . Is er een functionele androgeen receptor aanwezig, dan is deze in norm ale omstandigheden dan ook voldoende om een normaal trabeculair netwerk op te bouwen. De groei van de cortex of m.a.w. de breedtegroei van het b ot wordt ook voornamelijk bepaald door androgeenwerking via zijn recepto r. Toch kan de werking van testosterone gereduceerd worden wanneer zijn omzetting tot oestradiol geblokkeerd wordt. Het bot groeit dan minder in de breedte en zal daardoor aan sterkte verliezen. Androgenen zijn dus i nderdaad belangrijk voor de breedtegroei van het bot, maar naast een fun ctionele androgeen receptor is óók aromatisatie én een functionele oestr ogeen receptor essentieel om de breedtegroei van het bot maximaal te sti muleren. De productie van oestradiol en de activatie van de GH/IGF-I as bij de st art van de puberteit zijn, ook bij de man, nauw met elkaar verbonden en beiden worden gerelateerd aan de toename van de groeisnelheid (de groeis purt) tijdens de puberteit. Maar kan oestradiol de groei van het skelet beïnvloeden, ook als er geen functionele GH/IGF-I as aanwezig is? Deze v raag werd beantwoord door toediening van oestradiol aan muizen zonder fu nctionele GH receptor, zogenaamde GH receptor knockout muizen. Bij deze muizen, die dwerggroei vertonen omdat ze geen GH werking hebben en bijge volg ook zeer lage IGF-I spiegels in de bloedbaan hebben, herstelt oestr adiol de groeisnelheid van het bot in zowel de lengte als de breedte. Me t andere woorden, oestradiol zet deze dwergmuizen aan tot groeien. Het m echanisme hierachter is vernieuwend en toont aan dat oestradiol rechtstr eeks de synthese van IGF-I in de lever stimuleert. Tot nog toe dacht men dat enkel GH dit kon, maar wij tonen aan dat, in afwezigheid van GH wer king en bij zeer lage IGF-I spiegels, ook oestradiol deze functie kan ve rvullen. De verhoogde IGF-I synthese in de lever resulteert in een stijg ing van IGF-I in de bloedbaan en deze stijging staat in rechtstreeks ver band met het herstel van de groeisnelheid van het bot. Vooral het stimul eren van de breedtegroei springt in het oog aangezien deze zeer sterk be paald wordt door circulerend IGF-I en in grote mate zal bijdragen tot de sterkte van het bot. GH en/of circulerend IGF-I zijn dus zeer belangrijk voor de breedtegroei van het skelet. Maar ook aan androgenen wordt deze belangrijke rol toeg eschreven. Werken beiden samen, onafhankelijk van elkaar of net door int eractie? Is ieders aandeel even groot of heeft één van beide de overhand ? Deze vragen werden beantwoord door aan GH receptor knockout muizen and rogenen (testosterone of dihydrotestosterone) toe te dienen. Zoals voord ien al bewezen, stimuleren androgenen significant de breedtegroei van he t bot. Zij doen dit onafhankelijk, namelijk zonder tussenkomst van GH en /of IGF-I. Toch kunnen androgenen het groeideficit als gevolg van afwezi ge GH werking niet volledig omkeren. GH/IGF-I en androgenen zijn dus bei den onafhankelijk maar samen nodig om een maximale breedtegroei van het bot tot stand te brengen. Wel wordt het merendeel van de breedtegroei be paald door GH en/of IGF-I, aangezien het deficit als gevolg van afwezige GH werking groter is dan het deficit als gevolg van afwezige androgeenw erking. Geslachtshormonen spelen niet alleen een belangrijke rol in de groei van het skelet tijdens de puberteit, maar zijn ook onontbeerlijk voor het b ehoud van het skelet na de groei en dit zowel bij vrouwen als mannen. Ma ar zijn androgenen bij de man relatief belangrijker dan oestrogenen, of zijn ze veeleer equivalent? Deze vragen werden beantwoord door aan manne lijke muizen zonder functionele ER (zogenaamde ER knockout muizen) ofwel dihydrotestosterone ofwel oestradiol toe te dienen. Het trabeculair bot , dat een “netwerk” van botpartikels vormt binnenin het bot en op die ma nier de stevigheid van een bot mee bepaald, kan in dezelfde mate door zo wel androgenen als oestrogenen in stand worden gehouden. De cortex van h et bot lijkt echter eerder “inert” en z’n dimensies veranderen niet onde r invloed van androgeen- of oestrogeenstimulatie. Dit wijst erop de stru ctuur van de cortex voornamelijk bepaald wordt tijdens de actieve period e van groei tijdens de puberteit, terwijl het trabeculair bot responsief blijft aan veranderingen in geslachtshormonen zowel tijdens als na de p uberteit. Oestradiol behoudt wel de botdensiteit, die de hoeveelheid cal cium opgeslagen in een bepaald volume botweefsel weergeeft. In geval van androgeentekort bij de man en daarmee gepaard gaand b otverlies is therapie noodzakelijk. In de zoektocht naar geschikte andro geentherapie is men vooral geïnteresseerd in die androgenen die een guns tig effect hebben op de botmassa en de lichaamssamenstelling (verhouding spier en vet) zonder daarbij nevenwerkingen te vertonen op andere andro geen-gevoelige weefsel, zoals de prostaat. 7α-methyl-19-nortestoste rone is een synthetisch androgeen waarvan nog niet geweten was of het ee n positieve invloed zou kunnen hebben op het bot en de lichaamssamenstel ling zonder daarbij de prostaat te vergroten. In oude ratten werd een pr eklinische evaluatie van dit synthetisch androgeen uitgevoerd en werd aa ngetoond dat vooral lage dosissen interessant zijn, aangezien deze het b ot en de lichaamssamenstelling bewaren zonder de prostaat te vergroten. 7α-methyl-19-nortestosterone kan daarom potentieel belangrijk zijn als therapie bij androgeentekort bij mannen.status: Publishe

    De interactie van androgenen met bot en spier en hun belang voor de piek botmassa opbouw.

    Osteoporose is een belangrijk gezondheidsprobleem, niet enkel bij vrouwen, maar ook bij mannen. Ondanks de lagere incidentie aan osteoporotische fracturen bij mannen, gaat osteoporose bij de man gepaard met grotere morbiditeit, mortaliteit en medische kost in vergelijking met postmenopausale osteoporotische vrouwen. Alhoewel osteoporotische fracturen vooral voorkomen bij ouderen, ontstaat osteoporose reeds tijdens de jonge levensjaren door een interactie van zowel genetische als omgevingsfactoren. Geslachtshormonen worden hierbij gezien als belangrijke spelers verantwoordelijk voor het verschil in botsterkte tussen mannen en vrouwen. Een beter inzicht in de rol van mannelijke geslachtshormonen voor de opbouw van het skelet kan daarom leiden tot betere behandelingsmethoden voor osteoporose.Testosteron, het voornaamste androgeen bij mannen, wordt beschouwd als de belangrijkste determinant van mannelijke botgroei tijdens de puberteit. De botgroei kan echter ook verklaard worden door de hogere toename van spiermassa in mannen, waarbij het bot zich aanpast aan de grotere krachten die erop uitgeoefend worden. Bovendien kunnen de effecten van testosteron op bot zowel rechtstreeks gemediëerd worden via de androgeen receptor in botweefsel als onrechtstreeks, via de androgeen receptor aanwezig in spier. Om de rol van de androgeen receptor in deze spier-bot interactie te verduidelijken, werden verschillende experimenten uitgevoerd in mannelijke muizen tijdens de groei. Eerst werd tijdens een longitudinale studie de botmassaopbouw vergeleken in een muismodel voor seniele osteoporose (SAMP6) met een muismodel voor hypogonadale osteoporose (gecastreerde controle SAMR1) van 4 tot 20 weken. Vervolgens werd ook het effect van langdurige dihydrotestosteron (DHT) en oestradiol (E2) substitutie onderzocht in deze beide muismodellen. Aangezien DHT een niet-aromatiseerbaar androgeen is en E2 een oestrogeen, werd op deze manier de rol van zowel de androgeen als de oestrogeen receptor onderzocht (Chapter 4).Een interessante bevinding van deze studie was de significant lagere opbouw van corticale botmassa in SAMP6 muizen in vergelijking met hun controlemuizen, SAMR1. Reeds voor de puberteit (voor de leeftijd van 4 weken) hadden SAMP6 muizen een merkelijk dunnere cortex als gevolg van een verhoogde medullaire botexpansie en gedaalde endocorticale botformatie (-43%). De periostale botformatie was echter toegenomen (+47%) in SAMP6 muizen, wat resulteerde in toegenomen endocorticale en periostale botomtrekken. Dit verschil in corticale botmassa kon echter niet verklaard worden door het gelijke lichaamsgewicht of kleinere spierdiameter in SAMP6 in vergelijking met SAMR1 muizen. In tegenstelling tot deze bevindingen, resulteerde castratie op jonge leeftijd vooral in een verminderde periostale botaanmaak en een verminderde toename in spiermassa tijdens de groei. Bovendien ontstonden de cortical botdefecten voornamelijk tijdens de puberteit (4 tot 8 weken). Castratie resulteerde ook in een verminderde opbouw van trabeculaire botmassa en een hoge botturnover, terwijl de trabeculaire botmassa niet verschillend was tussen SAMP6 en SAMR1.Vervolgens werd ook de werking van verschillende geslachtshormonen onderzocht in gecastreerde SAMP6 en SAMR1 muizen. Zowel DHT als E2 stimuleerde periostale botexpansie in gecastreerde SAMP6 en SAMR1 muizen. Bovendien werd ook de trabeculaire botvorming verbeterd door DHT en E2 toediening. Opmerkelijk hierbij was dat E2 de endocorticale botformatie stimuleerde in SAMP6, waardoor het corticaal fenotype gedeeltelijk hersteld werd. Aangezien ook hier het effect van beide geslachtshormonen zich voornamelijk beperkte tot de puberteit (4 tot 8 weken bij de muis) wordt de de puberteit, en zelfs de periode voor de puberteit, aanzien als een zeer kritische periode voor de corticale botontwikkeling. Bovendien was zowel een niet-aromatiseerbaar androgeen als een oestrogeen in staat om de botmassa te verbeteren bij hypogonadale muizen. Daarom lijkt niet alleen de androgeen receptor maar ook de oestrogeen receptor belangrijk voor de botopbouw bij de man. Ten tweede werd de rol van de androgeen receptor en het effect van vrijwillig lopen onderzocht tijdens de opbouw van botmassa in muizen die androgeen resistent zijn door een volledige afwezigheid van de androgeen receptor (ARKO). Gedurende een experimentele periode van 11 weken werden 5 weken oude ARKO en wildtype (WT) muizen opgevolgd, waarbij 1 groep ARKO muizen vrijwillig kon lopen in een kooi met een loopwiel (Chapter 5).Hierbij zagen we dat de corticale bottoename (corticale dikte, oppervlakte, omtrek en ‘strength strain index’) verminderd was in muizen zonder androgeen receptor. Bovendien werd er, in tegenstelling tot WT muizen (controle muizen), geen correlatie gevonden tussen de ‘strength strain index’ (een parameter voor botsterkte) en de kuitspierdiameter. Ook de opbouw van trabeculaire botmassa was ernstig verminderd in ARKO muizen ten opzichte van WT muizen. De botturnover parameters waren toegenomen bij de ARKO, een weerspiegeling van hun hoge trabeculaire bot hermodellering. Vrijwillig lopen reduceerde de bot turnovermerkers in ARKO muizen en was in staat om het trabeculair botverlies, maar niet het corticaal botverlies, in ARKO muizen gedeeltelijk tegen te gaan. Bijgevolg suggereerden deze bevindingen dat androgenen niet alleen essentieel zijn om de corticale botmassa te optimaliseren, maar vooral van belang zijn voor de ontwikkeling van het trabeculaire botnetwerk tijdens de groei. Bovendien is de groei een kritische en kwetsbare periode gedurende dewelke androgenen, spiermassa en lichaamsbeweging trabeculair en corticaal bot kunnen beïnvloeden. Zoals hoger aangegeven, stimuleren androgenen niet enkel de botmassa maar ook de spiermassa. Daarom werd ook de rol van de androgeen receptor in de opbouw van de mannelijke spiermassa bestudeerd. Hiervoor werd een spiervezel-specifieke androgeen receptor knockout muis (mARKO) ontwikkeld. In tweede instantie werd het effect van de spiervezel-specifieke androgeen receptor disruptie op spiermassa en -functie ook vergeleken met de effecten in muizen met een globale androgeen receptor knock-out (ARKO) om een beter inzicht te krijgen in de rol van de androgeen receptor in spier (Chapter 6).mARKO muizen vertoonden een belangrijke reductie in androgeen receptor in spier (-60-80%). Dit resulteerde in een lager totaal lichaamsgewicht (-8%) en lager vetvrij lichaamsgewicht (-8%) in mARKO dan WT muizen. Bovendien was ook het gewicht van de androgeen-gevoelige levator ani spier significant verminderd (-46%). Het gewicht van andere perifere skeletspieren was echter slechts weinig of niet gedaald in mARKO muizen. Opmerkelijk was ook dat mARKO muizen minder intra-abdominaal vet hadden dan hun WT controlemuizen. Ook hadden de mARKO muizen, in tegenstelling tot de ARKO muizen, geen corticaal en trabeculair botfenotype, wat aantoont dat de deletie van de androgeen receptor in spiervezels de skeletontwikkeling in de man niet beïnvloedt. De selectieve disruptie van de androgeen receptor in spiervezels resulteerde evenwel in een verschuiving van snelle naar trage spiervezels, echter zonder grote gevolgen op intrinsieke spiersterkte of vermoeidheid zoals gemeten in vitro. Bovendien was het spierfenotype van mARKO muizen gelijkaardig aan dat van muizen met een globale androgeen receptor deletie (ARKO muizen). Ook in ARKO muizen was het gewicht gedaald in sommige, maar niet alle skeletspieren. De levator ani was zelfs compleet afwezig. De levator ani lijkt daarom dus veel gevoeliger voor androgenen dan perifere skeletspieren. In overeenstemming met de veranderingen in mARKO muizen, werd er in de soleus van ARKO muizen ook een hoger percentage aan trage spiervezels gevonden terwijl de intrinsieke spiersterkte en -vermoeibaarheid grotendeels onveranderd waren. Alles bij elkaar genomen, toonden de bevindingen in deze studie aan dat, onder basale omstandigheden, de androgeen receptor in spiervezels bijdraagt tot de ontwikkeling van spiermassa en de spiervezelverdeling, maar niet of nauwelijks tot de intrinsieke spiersterkte of -vermoeibaarheid. Het gewicht van de levator ani bleek de meest gevoelige parameter voor androgeen receptor gemediëerde anabole effecten. Bovendien verandert de specifieke deletie van androgeen receptor in spiervezels de globale lichaamssamenstelling maar niet de botopbouw. Ter conclusie, de androgeen receptor is essentieel is voor een normale trabeculaire en corticale botontwikkeling in het mannelijk skelet. Ook is de androgeen receptor belangrijk voor een normale opbouw van spiermassa, maar minder voor spierfunctie. Bovendien kan een veranderde mechanische belasting door vrijwillig lopen of verandering in spiermassa de opbouw van trabeculaire en corticale botmassa gunstig beïnvloeden.status: Publishe

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     Androgeenactie in skeletspier en glucosehomeostase

    Androgens are steroid hormones which are, in men, secreted predominantly by the testes. The major gonadal androgen is testosterone (T). In peripheral tissues, T can be converted by 5α-reductase enzymes into the more potent androgen 5α-dihydrotestosterone (DHT), which, like T, activates the androgen receptor (AR). In addition, the aromatase enzyme complex can convert T into 17β-estradiol (E2), which activates the estrogen receptors α and β (ERα/β). Both AR and ERα/β are ligand-inducible transcription factors that bind to specific DNA sequences to facilitate the transcription of their respective target genes. Androgens are responsible for establishing and maintaining the male phenotype. In addition, they have anabolic effects on several non-reproductive organs as well as metabolic actions. In this thesis, we focused on androgen effects on skeletal muscle and glucose homeostasis. Muscle frailty is a major cause of disability in elderly that greatly impairs their quality of life. Furthermore, loss of muscle mass and function is also observed in immobilized patients and is a common feature of various chronic illnesses. Androgens could potentially be exploited in these patient groups, as the increase in muscle mass and strength in response to T is well-documented. However, the lack of knowledge regarding the exact mechanism of androgen action in muscle, as well as the many effects on other organ systems, currently limits their use in a routine clinical setting. The first part of this project aimed to unravel the androgen response in skeletal muscle and to characterize the signaling pathways involved in their anabolic effects. To this end, we developed a new model to inactivate the AR in the muscle precursor cells, called satellite cells (satARKO = satellite cell-specific knockout of the AR). Total-limb maximal grip strength was decreased in satARKO mice, with soleus muscles containing more type I fibers and less type IIa fibers than the corresponding control littermates. The weight of the perineal levator ani (LA) muscle was markedly reduced. Thus, muscle AR regulates muscle mass, muscle strength, and fiber type distribution. Orchidectomy further decreased LA weight in satARKO mice, suggesting that at least part of the androgen effect on muscle is mediated indirectly via non-muscle AR pathways. In an attempt to identify target genes mediating androgen anabolic action, a transcriptomic analysis was performed in LA muscle of satARKO and control mice. Surprisingly, myostatin (Mstn), a strong inhibitor of skeletal muscle growth, was one of the most androgen-responsive genes (6-fold reduction in satARKO). We identified two conserved androgen response elements in the promotor and in exon 2 of the Mstn gene, pointing towards a direct transcription activation by the AR. As a consequence, muscle hypertrophy in response to androgens was augmented in Mstn knockout mice, suggesting that androgens induce Mstn signaling to restrain their own anabolic actions. Clinically, our data imply that Mstn inhibitors could potentiate the androgen response during the treatment of muscle wasting. Concerns about adverse effects of T on the cardiovascular system as well as prostate hyperplasia have stimulated the development of tissue-selective AR-ligands, the so-called selective AR modulators (SARMs), which are proposed to have desired anabolic effects in muscle and bone while avoiding prostatic side effects by their inability to be converted to DHT due to their non-steroidal structure. The processes by which SARMs induce muscle hypertrophy are, however, not fully elucidated. In the second part of this project, we investigated the mechanism of action of SARMs. We focused on GTx-024, a SARM that was reported to increase muscle mass in elderly men and cancer patient with muscle wasting. In orchidectomized satARKO mice, GTx-024 was still able to restore LA muscle weight, suggesting that GTx-024 action on muscle is, at least in part, indirect via non-muscle AR pathways. The transcript levels of Mstn and Amd1, two genes strongly downregulated in satARKO muscle, were normalized to sham levels in castrated control animals treated with GTx-024, indicating a role for muscle AR in mediating GTx-024 effects. Thus, just like androgens, the SARM GTx-024 has a dual mechanism of action in skeletal muscle, with both muscle AR and non-muscle AR pathways contributing to its anabolic effect. Obesity increases the risk of developing metabolic syndrome and its complications including type 2 diabetes (T2D). Although low T levels are associated with obesity, metabolic syndrome, and T2D, the causality and mechanisms of this association remain unclear. In the third part of this project, we examined the combined effect of androgen deficiency and high fat diet (HFD) on body composition and glucose homeostasis in two different mouse models: orchidectomy and global deletion of the AR. Androgen deficiency led to increased adiposity and serum leptin levels in HFD-fed mice. Fat accumulation was correlated with both increased lipogenesis and decreased lipolysis. Mitochondrial dysfunction in adipocytes may also contribute as reduced mitochondrial DNA content as well as decreased expression of the mitochondrial gene mtNd1 was observed in adipose tissue of castrated HFD-fed animals. Orchidectomy exacerbated HFD-induced glucose intolerance by impairing insulin action in both liver and skeletal muscle, as evidenced by the increased triglyceride and decreased glycogen content in these tissues. Pancreatic insulin secretion was also impaired. The metabolic alterations observed in androgen deficient HFD-fed animals were mainly due to concomitant estrogen deficiency, as T but not DHT supplementation restored the castration effects on body composition, serum leptin levels, glucose tolerance, and pancreatic insulin secretion. Our findings have implications for the prevention and treatment of metabolic disorders in male hypogonadism. Indeed, treatment of hypogonadal men with aromatizable androgens would be preferable to treatment with non-aromatizable androgens, as treatment with DHT or a SARM would not reverse the metabolic adversities due to estrogen deficiency.status: Publishe

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    The major side effects of androgen deprivation are osteoporosis and sarcopenia that both may increase fracture risk. Androgens prevent bone and muscle loss in hypogonadal as well as orchidectomized rodents. However, the potential of androgens to restore hypogonadal osteoporosis has not yet been fully explored. The first aim of the introductory study was therefore to evaluate to what extent androgens (both testosterone (T), the main circulating male androgen, and a synthetic androgen MENT) were able to restore bone mass as well as lean mass in a preclinical animal model of androgen deprivation. Two months following orchidectomy the aged male rats were characterized by cortical and trabecular osteopenia, which was secondary to increased intramedullary bone resorption combined with decreased periosteal bone formation. Both T and MENT not only prevented further bone loss but also stimulated periosteal bone formation in this hypogonadal rat model. Moreover, both androgens also increased lean body mass, which could indirectly also stimulate bone formation. Our further aim was to explore to what extent the AR in bone cells of male mice was involved in the antiresorptive as well as in the osteoanabolic actions of androgens on respectively trabecular and cortical bone compartments. Global disruption of the androgen receptor in male mice was characterized by severe and early trabecular and cortical osteopenia again due to concomitant decreased periosteal bone formation as well as increased intramedullary bone resorption. Although the AR is expressed in different bone cells, the role of AR in these bone cells with respect to the suppressive effects on bone resorption as well as the stimulatory action on bone formation was not established. Using Cre-loxP technology, the AR was selectively and successfully deleted in osteocytes, which are the most abundant bone cells with the highest AR expression. For this purpose DMP1 (Dentin Matrix Acidic Phosphoprotein 1) Cre targeted expression was used which is expressed in terminally differentiated osteocytes. Bone phenotype of the ocy-ARKO mice -in analogy with other studies in late and early osteoblast differentiation stages- was only characterized by a late and moderate trabecular osteopenia without affecting cortical bone formation and/or excessive bone resorption. In order to further explore whether AR in osteoclasts could directly explain its suppressive effect, a selective ocl-ARKO was created with similar Cre-loxP technology using the cathepsin K Cre, which is expressed in mature osteoclasts. AR expression osteoclasts was however extremely low. The ocl-ARKO showed no bone phenotype and responded normally to androgen deprivation as well as replacement. Therefore, AR in mature osteoclasts did not appear to suppress bone resorption. Nevertheless, osteoclasts were increased in global ARKO mice. Moreover, bone marrow cell cultures of the global ARKO mice -in comparison with WT mice- also showed increased osteoclast generation as well as activity. To further explore the potential suppressive effect of AR in osteoblasts and in bone marrow cells on osteoclast generation and activity co-culture experiments were performed. Co-cultures of osteoblasts with bone marrow cells from respectively WT and ARKO showed increased osteoclastogenesis as well as osteoclast activity. These data indicated that bone marrow cells, and not osteoblasts, were essential for osteoclast generation and activity. Bone marrow however consists of hematopoietic cells, which can differentiate towards osteoclasts, and of stromal cells, which can differentiate into osteoblasts. AR mRNA expression was high in these stromal cells. Further gene expression analyses revealed no differences in RANKL/OPG ratio, which is the main determinant of bone resorption, in osteoblasts or stromal cells between ARKO and WT. Therefore the enhanced bone resorption observed in the global ARKO could not be due to higher RANKL expression and therefore more stimulation of osteoclastogenesis. Interestingly ARKO stromal cells exhibited higher expression of Runx2 mRNA compared to WT. Therefore, the AR might prevent early recruitment of bone marrow cells into the bone-remodeling cascade with minimal effect at later stages of osteoblast or osteoclast differentiation. Our in vitro observations might also explain the discordance of the modest bone phenotype characterized by late trabecular osteopenia in current selective bone ARKO models versus the severe early cortical as well as trabecular osteopenia in global ARKO. The final aim of this study was to investigate to what extent androgens increased the sensitivity to mechanical loading and hereby indirectly stimulated bone formation. As shown earlier, androgens indeed stimulated also muscle and hereby loading which could explain their stimulatory action on bone formation. In a mice tibia loading model the osteoanabolic response following mechanical loading was evaluated in mice who were androgen deprivated and mice who received androgen replacement (aromatizable testosterone or non-aromatizable dihydrotestosterone) immediately after orchidectomy. This osteoanabolic response was enhanced by androgen deprivation and decreased by androgen replacement hereby refuting the hypothesis that androgens increased mechanical sensitivity. On the contrary, mechanical sensitivity was reduced by androgens. Interestingly, such action was associated with changes in bone sclerostin expression, which is the major regulator of bone formation and which may open further therapeutic perspectives. Moreover, both androgens and loading, however independently also decreased bone resorption as well as RANKL-OPG expression. In conclusion, the osteoanabolic action of androgens could not be explained by AR in bone cells nor by enhancement of mechanosensitivity. The antiresorptive action was also not fully explained by its presence in bone cells but may be related to its action on bone marrow stromal cells, which orchestrate bone remodeling.status: Publishe

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
