MultiMeta: An R package for meta-analyzing multi-phenotype genome-wide association studies
As new methods for multivariate analysis of genome wide association studies become available, it is important to be able to combine results from different cohorts in a meta-analysis. The R package MultiMeta provides an implementation of the inverse-variance-based method for meta-analysis, generalized to an n-dimensional setting
High Purcell factor photonic crystal cavities for single photon sources
Enhancement of spontaneous emission is of crucial importance in the design of high efficiency single photon sources. This can be achieved by creating defect cavities with both high Q and small effective mode volume, which results in a high Purcell factor. Photonic crystal (PhC) microcavities are very promising candidates in this area and this work is using 3D finite difference time domain modelling to investigate these devices. We have extensive experience in modelling and measuring more conventional pillar microcavities and thus are ideally placed to assess PhC based structures. The structure under consideration is the case for a one missing hole cavity. The structure also shows the strongly confined x directed electric field. The simulated Q factor calculated from observing the ring down of the cavity mode and the use of Fourier transforms are also discussed. As expected for this simple case very small Q factors are obtained. This work will go on to investigate different cavity configurations and calculate effective mode volumes in order to evaluate the Purcell factor in more advanced design

A Framework for Prototyping J2EE Replication Algorithms
In application server systems, such as J2EE, replication is an essential strategy for reliability and efficiency. Many J2EE implementations, both commercial and open-source, provide some replication support. However, the range of possible strategies is wide, and the choice of the best one, depending on the expected application profile, remains an open research question. To support research in this area, we introduce a framework for prototyping J2EE replication algorithms. In effect, it divides replication code into two layers: the framework itself, which is common to all replication algorithms, and a specific replication algorithm, which is "plugged in" to the framework. The division is defined by an API. The framework simplifies development in two ways. First, it keeps much of the complexity of modifying a J2EE implementation within the framework layer, which is implemented only once. Second, through the API, the replication algorithm sees a highly abstracted view of the components in the server. This frees the designer to concentrate on the important issues that are specific to a replication algorithm, such as communication. We have implemented the framework by extending the open-source J2EE server. Compared to an unmodified server, the framework adds a performance cost of about 22%. Aerobic pyruvate metabolism sensitizes cells to ferroptosis primed by GSH depletion
Ferroptosis is a non-accidental, regulated form of cell death operated by lipid peroxidation under strict control of GPx4 activity. This is consistent with the notion that lipid peroxidation is initiated by radicals produced from decomposition of traces of pre-existing lipid hydroperoxides. The question, therefore, emerges about the formation of these traces of lipid hydroperoxides interacting with Fe2+. In the most realistic option, they are produced by oxygen activated species generated during aerobic metabolism. Screening for metabolic sources of superoxide supporting ferroptosis induced by GSH depletion, we failed to detect, in our cell model, a role of respiratory chain. We observed instead that the pyruvate dehydrogenase complex -as other α keto acid dehydrogenases already known as a major source of superoxide in mitochondria- supports ferroptosis. The opposite effect on ferroptosis by silencing either the E1 or the E3 subunit of the pyruvate dehydrogenase complex pointed out the autoxidation of dihydrolipoamide as the source of superoxide. We finally observed that GSH depletion activates superoxide production, seemingly through the inhibition of the specific kinase that inhibits pyruvate dehydrogenase. In summary, this set of data is compatible with a scenario where the more electrophilic status produced by GSH depletion not only activates ferroptosis by preventing GPx4 activity, but also favors the formation of lipid hydroperoxides. In an attractive perspective of tissue homeostasis, it is the activation of energetic metabolism associated to a decreased nucleophilic tone that, besides supporting energy demanding proliferation, also sensitizes cells to a regulated form of death
