    Tutor per le attività di Ricerca svolte dalla Dr. Vita Mariagrazia Vecchio nell’ambito della borsa di ricerca assegnata dal Consorzio Interuniversitario di Ricerca sulla Chimica dei Metalli nei Sistemi Biologici, CIRCMSB, tematica Nuovi Farmaci in Oncologia per la ricerca dal titolo: Studio in vitro della possibile platinazione di acidi nucleici operata da polimerasi e complessi di Pt(II) con basi puriniche. BREVE DESCRIZIONE DEL PROGETTO Tra i farmaci utilizzati nel trattamento antitumorale, i complessi del platino meritano indubbiamente un riguardo particolare soprattutto nel trattamento di tumori solidi dell’apparato urogenitale. Il meccanismo con cui si esplica l’attività antitumorale del cisplatino, non è ancora del tutto noto pur sapendo che è particolarmente importante la capacità di molti complessi di questo metallo di legare le basi puriniche del DNA. Lunghe discussioni sono presenti in letteratura sul come e sul perché il cisplatino e i suoi analoghi di uso clinico, possano raggiungere e platinare il DNA nucleare in cellula. E’ noto anche come la formazione di addotti (mono o bis), soprattutto con basi puriniche del DNA possa interferire con i normali processi di replicazione cellulare, inducendo apoptosi. Per il cisplatino è noto come l’ingresso in cellula attraverso la membrana cellulare possa avvenire sia per diffusione passiva che per fenomeni di trasporto attivo scoperti di recente. Uno stadio chiave per comprendere il meccanismo di platinazione del DNA resta comunque la comprensione dal punto di vista chimico di ciò che avviene nel citoplasma cellulare prima che il DNA possa essere platinato, visto che prima di incontrare il DNA il cisplatino incontra specie ben più reattive (molecole S e N donatrici). In questo contesto si inserisce il nostro progetto di ricerca, cogliendo un aspetto finora non considerato in letteratura che è la possibile ipotizzabile formazione di addotti di varia natura con basi puriniche normalmente presenti nel citoplasma cellulare. Infatti in cellula ovunque sono presenti derivati purinici con funzioni varie tra cui anche la costituzione di una riserva di building blocks per la sintesi di nuovo DNA o RNA in processi di replicazione. Una prima parte del lavoro consisterà nello studio dell’attività citotossica di complessi in cui il platino è legato direttamente a basi puriniche del DNA. Successivamente si valuterà il possibile inserimento nel DNA di nuova sintesi di complessi preventivamente legati alle basi nucleiche del DNA, normalmente presenti in cellula (nucleotide monofosfato, difosfato o trifosfato nelle forme normali o desossi). Infatti pensiamo che non sia giusto escludere a priori un attacco della mono-acquo specie formata in cellula ad esempio dal cisplatino, cis-[(NH3)2(H2O)Cl]+, alle basi puriniche libere, presenti nell’ambiente cellulare, prima che al DNA. Questa idea ci sembra rafforzata dal fatto che, in genere, il DNA è presente in cellula prevalentemente nella forma a doppio filamento poco reattiva e sicuramente meno reattiva delle basi libere, se non altro per problemi di ingombro sterico. E’ questo tipo di considerazioni insieme ad incoraggianti esperimenti preliminari che hanno suggerito di approfondire, lo studio del possibile inserimento di basi già platinate nel DNA di nuova sintesi. Chiaramente l’obiettivo ultimo ed ancora molto lontano di questo lavoro sarà di comprendere se tali processi possano avere un ruolo nell’esplicazione in vivo dell’attività antitumorale dei complessi del platino attualmente in uso clinico. E’ noto che derivati nucleotidici del tipo dNTP sono inglobati nel DNA di nuova sintesi da varie DNA polimerasi. Nello screening preliminare effettuato prima di presentare questo progetto si è scelta una polimerasi procariotica facilmente reperibile e di basso costo come la Taq polimerasi, utilizzando il complesso [Pt(dien)(N7-5’-dGTP)] come sistema modello di complesso con basi puriniche da poter inglobare nel DNA di nuova sintesi. Con tecniche analoghe alla PCR (polymerization chain reaction) si è inserita con successo la base platinata in vari oligonucleotidi con circa 100 bp, confermando l’avvenuta platinazione con tecniche di gel elettroforesi utilizzando gel di poliacrilammide, per separare catene di lunghezza diversa ottenute nel corso dei processi di polimerizzazione, e di assorbimento atomico del platino presente nell’oligo (per individuare la posizione degli oligo sul gel elettroforetico si sono utilizzati marcatori fluorescenti). Ovviamente necessita un’ulteriore conferma di quanto osservato e a questo riguardo pensiamo di ripetere gli esperimenti detti esaminando i prodotti ottenuti anche con tecniche di autoradiografia, marcando con isotopi radioattivi gli oligo, al fine di ottenere una maggiore risoluzione nella determinazione della loro posizione nel gel elettroforetico. Per determinare la presenza effettiva del platino negli oligo purificati per elettroforesi si utilizzeranno tecniche di assorbimento atomico che pensiamo in futuro di abbinare alla spettrometria di massa come l’ESI e la MALDI. Si può prevedere successivamente alla fase di verifica in vitro dei processi di platinazione menzionati anche una fase di verifica, in vivo. Ovviamente obiettivo ultimo di questo progetto è di verificare se il nuovo meccanismo di platinazione descritto possa operare anche in vivo nei mammiferi e nell’uomo. Ciò potrebbe suggerire nuove strategie per la realizzazione di nuovi farmaci antitumorali magari efficaci contro forme tumorali resistenti e dotati di minore tossicità ed effetti collaterali

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    New organometallic Platinum containing biomimetic Vitamin E analogues.

    Bioorganometallic chemistry is an emerging area of organometallic chemistry. The presence of a metal in a biomimetic molecule may be useful for the possibility to deliver, a particular metal, to a specific target, for therapeutic and/or imaging purposes. In this work, we report the synthesis of new Pt(II) containing organometallic species, having the same structural core of Vitamin E, a remarkable family of biologically active chromane derivatives. Such compounds are characterized by substitution of the carbon atom in 2 position within the chromane pseudo-pyran heterocycle with a square planar Pt(II) centre, which carries also a chelate dinitrogen ligand. Cytotoxicity and Pt uptake measurements, performed on health or cancer cells, show interesting structure-activity relationships for the new metalla-chromanes

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used

    New chemistry of olefin complexes of platinum(II) unravelled by basic conditions: Synthesis and properties of elusive cationic species

    The evolution in basic medium ([RO-] = 1 M in methanol, R = H or Me) of five-coordinate platinum(II) compounds, [PtCl2(η2-C2H4)(N-N)], 2a-c, (N-N = N,N,N′,N′-tetramethyl-1,2-ethanediamine, a; 2,2′-bipyridyl, b; 1,10-phenanthroline, c) leads to the formation of [PtCl(η1-CH2CH2-OCH3)(N-N)], 5a-c. The analogous compound 5d (N-N = 2,9-dimethyl-1,10-phenanthroline, d) can also be prepared, but not via transformation of the five-coordinate species 2d in basic medium where it is quite stable. 5d can instead be prepared by reaction of d with a strongly basic methanol solution of Zeise's anion [PtCl3(η 2-C2H4)]-, 1. In such a medium the di-anionic trans-[PtCl2(OR)(η1-CH2CH2-OCH3)]2- species (1′′) reacts with d to form exclusively 5d. Hydrolysis of 5a-c with acids bearing weakly coordinating anions leads to [PtCl(η2-C2H4)(N-N)] +, 3a-c, as stable cations; upon the same treatment 5d does not generate 3d, but it reacts with HCl to give 2d in almost quantitative yield. Cationic complexes 3b, 3c, here reported for the first time, were reacted with some nucleophiles and their behaviour compared with that of the already known 3a. In 3b, 3c the metal centre competes with the coordinated ethene for binding to nucleophiles; therefore the acetylacetonate anion can either add to the olefin (affording compounds 6b, 6c) or to the metal ion replacing the ethene ligand (yielding compounds 7b, 7c). Under similar conditions, 3a gives exclusively 6a. Secondary amines readily add to ethene in 3b, 3c, affording the addition products 8b, 8c, which undergo a ready cyclization to an azaplatinacyclobutane ring (9b, 9c). The remarkable ease of the four-membered ring formation has been related to the high electrophilic character of the metal core in 3b, 3c
