    Le Bundesrat. La protection des intérêts des Länder selon la Loi fondamentale allemande

    After dealing with the West German Bundersrat in a double-chamber system, the author, following the Basic Law of the country, depicts the varied powers of this Chamber. The participation of the Bundersrat in federal legislation is examined through its suspensive veto over every bill adopted by the first chamber — the Bundestag. The Bundersrat also has the power to approve certain categories of Bills. The participation of the Bundersrat in the federal administration, contentious powers and the nomination process are ideas also developed in this article. The author examines the political role of the Chamber and shows that the Bundersrat has a counterbalancing effect between the Bundestag and the federal government. The importance of the roles of the parties in developing the political position of the Bundersrat is also discursed herein. Professor Bothe concludes by saying that the Bundersrat is an important element of West German cooperative federalism and wonders if exporting this institution to Canada would be a wise move

    Tato práce se zabývá praktickým řešením businesového problému v oblasti nízké kontroly a visibility nákladů v rámci dodavatelsko – odběratelského řetězce v kontextu mezinárodní kontejnerové logistiky a implementací self-service business intelligence, aplikací BI nástroje Microsoft Power BI, Power Query jako plnohodnotný ETL nástroj a využití Microsoft O365 nástrojů. Praktickou projektovou metodologií pro implementaci celkového řešení a základními přístupy řešení data governance modelu. Principy návrhu jednotlivých dashboardů a efektivní řešení architektury s důrazem na maximální vzužití stávající infrastruktury a zdrojů. Hlavním cílem je návrh a implementace reportingového a analztického prostředí, které zefektivní process kontroly a reportování logistických nákladů a umožní efektivní využití času jednotlivých uživatelů k analýze a stanovování efektivních kontrolních mechanismů, které povedou k prevenci vzniku nákladů. Nezbytnými dílčími cíli práce je dokumentace datových zdrojů a procesů v dané oblasti, návrh governance procesu a identifikace potenciálních oblastí k větší efektivitě. Výsledkem práce je ověření spokojenosti s dodaným řešením jednotlivými klíčovými uživateli.This paper is focusing on delivering practical solution for solution of business problem with low visibility and cost control in supply chain in context of international container logistics, it provides end-to-end implementation self-service BI environment using Microsoft Power BI tool, Microsoft Power Query and Microsoft O365 tools. It provides practical project methodology for overall project delivery, general approach for implementation of data governance model, approaches to design reports and dashboards and effective set up od data architecture with focus on maximum usage of existing tools and resources. Main goal is to implement reporting and analytic solution that will enable data driven decision making and increase productivity of the users that will enable more focus on root cause analysis and implementation of preventive measures. Partial goals of the project are also to document data sources and processes in the area, set up of governance model proposal and identification of further process improvements.     Diplomová práca sa orientuje na súčasné trendy prístupu k nákladovej alokácii v nástroji IBM Cognos TM1. Koncept, ktorý bol po prvýkrát rozpracovaný v mojej bakalárskej práci, začal v poslednom čase narážať na obmedzenia, spôsobené zvyšujúcimi sa nárokmi na analytické nástroje a informácie, ktoré poskytujú. Cieľom práce je preto analyzovať príčiny vznikajúcich slabín, navrhnúť a implementovať optimalizované riešenie spĺňajúce súčasné požiadavky. Vzhľadom na kvantitatívne zhodnotenie dosiahnutých výsledkov je práca rozšírená o analýzu rámcov a štandardov určených na porovnávanie OLAP nástrojov a ich syntézu do vlastného komplexného modelu. Model sa špecializuje na meranie viacerých OLAP aplikácií cez 4 základné perspektívy obsahujúce výkon, vývoj, použiteľnosť a finančné benefity. Dosiahnuté výsledky potvrdzujú, že inovovaný model je rýchlejší, bohatší na informácie, jednoduchší na použitie a vhodný pre organizácie so štrukturovaným a algoritmickým prístupom k nákladovej alokácií. Druhá časť práce sa zameriava na rozšírenie prezentačnej vrstvy aplikácie do webového rozhrania a vývoj typizovaných vizualizácií pre najrozšírenejšie analytické úlohy. Vzhľadom na absenciu pokročilého aplikačného rozhrania nástroja IBM Cognos TM1 je práca rozšírená o teoretickú analýzu súčasných trendov pri vývoji API a následným návrhom konceptu, umožňujúcim komunikáciu a predávanie dát medzi aplikáciou a TM1 serverov. V záverečnej časti práce je koncept zhmotnený do univerzálnej knižnice vyvinutej v jazyku PHP a aplikovaný na aktualizovaný model alokátora. Využitím knižnice sú následne vyvinuté dve vzorové koncepty rozhraní pre ovládanie a prácu s modelom. Získané poznatky môžu slúžiť ako podklad pre vývoj ďalších komponentov komunikujúcich s TM1 v najrozšírenejšej palete projektov alebo ako teoretický základ pri tvorbe API vo všeobecnosti.Thesis is devoted to current trends in approaching cost allocation developed in IBM Cognos TM1 software. Attained results prove, that reengineered model is faster, data richer, easier to use and appropriate for any organization with structured and algorithmic approach to cost allocation. Second half of the thesis focuses on extending the presentation layer to web browser, designing and developing of custom visualizations for most usual analytic tasks. Considering the absence of advanced application interface in IBM Cognos TM1, the thesis also includes theoretical analysis of current trends in API development and design of concept allowing communication and data transportation between applications and TM1 server. In the concluding section of the thesis, proposed concept is materialized into universal library developed in PHP and applied to novel allocation model. Leveraging the library, two exemplary interfaces for allocator operation and data consumption are implemented. Gained knowledge can serve as basis for development of additional components communicationg with TM1 in variety of projects or theoretical framework for API implementation in general

    Thesis is focused on one of the most discussed area of Management Information Systems currently -- Corporate Performance Management. It does not aim on detail explanation of theme itself, but orientate to specific problematic of setting key solution of managerial accounting -- cost allocation, to the Performance Management systems. Thesis is divided into theoretical and practical part. First part is devoted to Corporate Performance Management definition, its placement in architecture of Business Intelligence devices as well as reasoning for its implementation considering current economical situation. From the main three pillars of Performance Management is major attention dedicated to analytical part enabling execution of what-if analyses. Part devoted to cost allocation identifies contributions of its implementation in optimization and minimization of expenditures as well as offers reasons for using them in bank sphere. Practical case study documents particular project phases of implementation of cost allocation analytical solution in fictive bank, while major section is dedicated to architecture and design part of the calculation engine. Project is concluded with description and explanation of advanced implementation techniques and methods in IBM Cognos TM1 software

    The molecular setup of the avian head mesoderm and its implication for craniofacial myogenesis

    The head mesoderm is the mesodermal tissue on either side of the brain, from forebrain to hindbrain levels, and gives rise to the genuine head muscles. Its relatedness to the more posterior paraxial mesoderm, the somites, which generate the muscles of the trunk, is conversely debated. To gain insight into the molecular setup of the head mesoderm, its similarity or dissimilarity to the somitic mesoderm, and the implications of its setup for the progress of muscle formation, we investigated the expression of markers (1) for mesoderm segmentation and boundary formation, (2) for regional specification and somitogenesis and (3) for the positive and negative control of myogenic differentiation. We show that the head mesoderm is molecularly distinct from somites. It is not segmented; even the boundary to the first somite is ill-defined. Importantly, the head mesoderm lacks the transcription factors driving muscle differentiation while genes suppressing differentiation and promoting cell proliferation are expressed. These factors show anteroposteriorly and dorsoventrally regionalised but overlapping expression. Notably, expression extends into the areas that actively contribute to the heart, overlapping with the expression of cardiac markers

    Carboranyl bisglycosides for the treatment of cancer by boron neutron capture therapy

    Boron neutron capture therapy is a special type of radiotherapy for the treatment of cancer by using boron compounds. Problems often arise from the: low water solubility of these compounds, their unselective uptake into the cancer cells, and their toxicity Here we describe the novel water-soluble ortho-carboranyl bisglycosides 7 and 10 containing either lactose or glucose and the mixed bisglycosides 1 and 28 containing glucose mannose, and galactose. The carboranyl bisglycosides show almost no toxicity toward bronchial carcinoma cells of line A549 up to a concentration of 0.50 mM As anticipated, these compounds exhibit nearly no uptake into C6 glioma cells; they can therefore be used for a selective delivery into malignant cells:by using conjugates of glycohydrolases and monoclonal antibodies which bind to tumor-associated antigens, since by enzymatic hydrolysis the bisglycosides are transformed into lipophilic compounds

    Modelování, analýza a počítačové simulace heterogenní katalýzy v mikroreaktorech

    Tato prace se zabývá nelineárním systémem parciálních diferenciální rovnic, konkrétně kombinací tzv. reakce-difuze, resp. konvekce difuze. Tato situace odpovidá popisu heterogenní catalýzy, kdy zkoumame je proudění objemových částic buzené sta- cionarním rychlostním polem. Absorptční mechanismy na stenach jsou popsané pomocí Langmir-Hinschelwoodovy absoprční kinetiky a jednosměrné povrchové reakce odpoví- dají zákonu zachování objemu všech částic. Tyto fyzikální jevy se přirozeně vyskytují u mnoha aplikací heterogenní katalýzy. V první části práce se zaměřujeme na analýzu sys- tému. Dokážeme existenci a jednoznačnost tzv. "mild" řěšení využitím teorie nelineárních semigrup. Ve druhé části práce presentujeme numerické metody, které jsme použili pro výpočet numerického řešení, a které splňují všechna analytická a fyzikalní kritéria, tudíž jsou vhodným nastrojem k výpočtu modelu popisujících heterogenní katalýzu v reálném prumyslovém využití. Klíčová slova: heterogenní katalýza, sparovaný reakce-difuze a konvekce-difuze system, teorie nelineárních semigrup, bio-diesel mikroreaktor 1We investigate a nonlinear reaction-diffusion system coupled with convection- diffusion system. This combined system corresponds to physical description of heteroge- neous catalysis when the flow of bulk-constituents is driven by a given stationary velocity field; diverse mechanisms between bulk- and surface-parts of the model-domain are de- scribed by Langmuir-Hinshelwood absorption kinetics; and the irreversible reactions on the catalytic walls meets the law of mass action with quadratic rate. The first part of the thesis is focused on analytical results; in Chapter 2 we prove existence and unique- ness of a mild solution for so-called near-by problem using nonlinear semigroup theory; in Chapter 3 we investigate the weak formulation of the problem. We prove an existence of a weak solution for little modified problem which, under an assumption, coincides with the original problem. In the second part of the thesis (Chapter 4) we numerically investigate the evolution of the bio-diesel microreactor. We compute numerical solutions using several methods and we test the results by analytical and physical conditions; with the aim to find the most efficient way to compute precise and physically correct solution.
