Acute myeloid leukaemia:analysis of the expression of P2X7a and P2X7B isoforms
No full textAcute Myeloid Leukaemia (AML) is a clonal disorder originating from the accumulation of myeloid progenitor cells in the bone marrow. An increasing number of reports correlate P2X7 with tumor growth and progression (Adinolfi et al. 2015). Haemopoietic lympho-proliferative disorders, among which AML (Salvestrini et al. 2012) were the first neoplastic models where P2X7 oncopromoting activity was demonstrated (Adinolfi et al., 2002; Chong et al., 2010).The P2X7 receptor for extracellular ATP is a ionotropic, ligand-gated, cation channel and it is expressed by different cell types such as neurons, macrophages, dendritic and microglial cells, fibroblasts, endothelial cells and lymphocytes. Nine different P2X7 splice variants have been detected in humans, among those just two, P2X7A and P2X7B are functional ion channels (Cheewatrakoolpong et al., 2005) both showing growth promoting activity (Adinolfi et al., 2010; Giuliani et al., 2014).
Isoform B is a naturally truncated P2X7 splice variant, widely expressed in several human tissues especially in lymphoid tissue and lymphocytes themselves, often to a much higher level than P2X7A. Considering that a wide characterization of P2X7B is still lacking, we analyzed the differential expression of P2X7A and P2X7B in a cohort of AML patients by Real Time PCR. We obtained a significant correlation between high P2X7B expression and relapse as compared to onset of disease. Moreover, low P2X7A expression associates with favorable karyotype and, although not significant, there is a similar tendency for P2X7B. Futhermore, preliminary data suggest a correlation between age of onset (>65 years) and high P2X7B expression. Although a wider cohort analysis will be required, these data prompt us to postulate that different P2X7A and B expression levels could be involved in AML development, suggesting that P2X7 could be a valuable prognostic marker and therapeutic target for the disease

Aim of our studies was to verify the effect of P2X7 expression and blockade in animal models of cancer. Tumor tested included colon cancer, melanoma
and neuroblastoma modeled either in syngeneic or in xenograft systems. In all this settings P2X7 was increasing tumoral engraftment and growth rate also
affecting blood vessels formation [Adinolfi et al., 2012]. Signaling cascades involved in P2X7 mediated transformation included the HIF-1 alpha-VEGF
and PI3Kinase-GSK3 axes. Of interest for the therapy of cancers that are still lacking a cure, such as advanced stages neuroblastoma and melanoma, was

L'escrit se centra en la idea que el feminisme, com a moviment de masses, demostra el caràcter hegemònic dels interessos de les classes treballadores. Adinolfi va posar en dubte les idees dominants del marxisme ortodox, considerant que el feminisme i la lluita treballadora havien de conformar una unitat d'acció. Per aquesta raó, el seu pensament es va caracteritzar per aportar una visió de la lluita feminista que no coincidia amb la concepció generalitzada del feminisme català de finals del segle XX. L'autora es va alimentar del pensament d'Antonio Gramsci concebent a les dones com a grup subaltern i assenyalant la política de masses com la gran potencialitat del feminisme.En este ensayo se analiza el pensamiento político feminista de Giulia Adinolfi. The writing analyse the idea that feminism, as a mass movement, demonstrates the hegemonic nature of the interests of the working classes. Adinolfi questioned the dominant ideas of orthodox Marxism considering that feminism and the workers struggle should form a unity of action. For this reason, her thought became special by providing a vision of the feminist struggle that did not coincide with the generalized conception of the time. The author fed on Antonio Gramsci's thinking, conceiving women as a subaltern group and pointing out mass politics as the great potential of feminism
