Synthesis and biological evaluation of hAMCase inhibitors and anomeric equilibrium of gluco and xylopyranose derivatives in D2O
Asthma is considered a serious health and socioeconomic issue all over the world, affecting more than 300 million individuals. It is a chronic inflammatory disease characterized by airway inflammation, mucus hypersecretion and airway hyperresponsiveness. One of the causes of this disorder is the result of an abnormal Th2-dependent response to environmental antigens. In particular, Th2 cells produce cytokines, such as interleukin (IL)-4, IL-5, IL-9, and especially IL-13, in the airway wall, associated with the asthma symptoms. Acidic Mammalian Chitinase (AMCase) is a chitinolytic enzyme belonging to glycosyl hydrolases family. AMCase seems to be involved in Th2 inflammatory diseases, such as asthma. It has been demonstrated that during Th2-dependent inflammation IL-13 induces the AMCase expression in the airways epithelial cells and alveolar macrophages, which stimulates the release of chemokines, recruiters of proinflammatory agents. In conclusion, AMCase contributes to the pathogenesis of asthma and represents a potential therapeutic target for the treatment of this disease and other forms of Th2-mediated inflammation. Prof. Botta research group, after a careful study, designed and synthesized an innovative AMCase inhibitor, BM22 (Ki = 13.2 μM), characterized by a macrocyclic structure. The aim of this work was the study of BM22 chemical space in order to improve its activity with the exploration of the macrocyclic portion, the linker length and substitution the amidinourea terminal moiety with a diamino-triazole group. Some of these compounds have been tested against AMCase enzyme and showed a better activity compared to that of BM22.
The last chapter of this thesis deals with the study of the anomeric equilibrium of xylo and glucopyranoses in D2O at room temperature. The anomeric effect is an effect that determines the increased preference of the axial position of the C1 substituent in a pyranose ring, rather than the sterically preferred equatorial position. Two models were proposed to explain this effect: Electrostatic model and Hyperconjugation model. The anomeric ratio was established by qNMR, which involved integration of resolved signals in 1H-NMR spectrum. Interesting results have been obtained, as all compounds showed a preference for α-anomer, including, surprisingly, the electron donating methoxy group as well
Dislipidemie aterogene: geni coinvolti nella sindrome da basse HDL
Le dislipidemie aterogene sono alterazioni del metabolismo lipidico associate ad elevato rischio cardiovascolare. Esse sono caratterizzate da bassi valori di HDL ed elevati livelli di trigliceridi, con numerosi geni potenzialmente coinvolti. Ciò rende particolarmente complessa l’analisi mutazionale.
In una casistica pediatrica di 55 bambini con valori di colesterolo HDL 150 mg/dl, abbiamo effettuato l’analisi mutazionale di 3 geni candidati, quello per l’apolipoproteina AI (APOA1), quello per la lipoproteinlipasi (LPL) e quello per la lecitina:colesterolo aciltransferasi (LCAT). Come popolazione di controllo è stata utilizzata una popolazione costituita da 174 bambini sani. Questo è stato il primo obiettivo del progetto del dottorato.
Nel gene APOA1 sono state identificate le seguenti variazioni di sequenza: 1 nel promotore (-75G>A) e 19 introniche (8 delle quali nuove).
Nel gene LPL sono state identificate le seguenti variazioni di sequenza: 1 al 5’UTR (-281T>G), 8 esoniche (N291S, L365V, D9N, S45N (nuova), V108V, E118E, T106T, S447X) e 14 introniche. La N291S, L365V, -281T>G e la D9N sono state precedentemente descritte come varianti associate a fenotipi iperlipidemici e bassi livelli di HDL.
Le varianti identificate nel gene LPL sono state ricercate tramite saggio SNaPshot nella popolazione di controllo. La variante N291S è associata al fenotipo di dislipidemia aterogena (p=0.01).
L’identificazione delle lesioni molecolari nei geni candidati e la loro caratterizzazione risultano indispensabili al miglioramento dell’abilità diagnostica, prognostica e terapeutica di tale sindrome.
Il secondo obiettivo di questo progetto di dottorato, è stato quello di identificare mutazioni nel gene ABCA1 in un probando adulto caratterizzato da livelli molto bassi di colesterolo HDL (HDL-C: 1 mg/dl) al fine di confermare la diagnosi clinica di malattia di Tangier. L’analisi mutazionale e d’espressione del gene ha identificato due varianti introniche (1195-27 G>A; 1510-1 G>A, nuova) che provocano un difetto di splicing e portano alla produzione di una proteina tronca. Il probando è risultato essere eterozigote composto per le due varianti introniche che, quindi, possono essere associate alla malattia di Tangier
Recycle of Industrial Heritage: is it still life?
Today as in the past decades, a growing number of industrial and military sites are being abandoned and disused. This number in Europe is estimated to go from a few hundred in small Member States with a modest industrial legacy to a few hundred thousand in the larger Member States with an important industrial past (Cabernet, 2006).
To date in Italy no overall census has yet been achieved on abandoned industrial sites, if not at the level of regions or local authorities but, according to Istat surveys of 2012, it seems that they represent 3% of the National territory, an area of about 9 thousand square kilometres, equal to that of Umbria, of which 30% located in the urban area.
The interpretation of this phenomenon of decommissioning, as a central issue of architectural debate on the contemporary city, is therefore extremely urgent due to the intrinsic potential linked to regeneration projects and new uses of this considerable heritage.
This research work, exemplified by a case study, wants to meditate to find tools to give a new meaning and a renewed use to pre-existence in our urban and environmental territory and to give new life to what is rejected or abandoned, cancelling waste processes as much as possible (Bocchi, 2013).
The spaces of production, designed and realized according to precise functionalist and technical logics, have offered and offer themselves today to various potential uses: spaces conceived from a rationalized and delimited productive logic, expression of a precise historical period, evidence of social transformation, symbol of redemption and economic health of the working class, but also places of pride for technological advancement, aggregation and daily life. They may become fertile ground for design transformations that bring out potential of new uses.

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Caregiver decisions along the Alzheimer's disease trajectory
Available online 29 November 2018Despite the rising prevalence of Alzheimer's disease (AD), there is limited systematic evidence about disease specific decisions. The aim of this qualitative descriptive study was to identify decisions across the AD trajectory using focus groups of past and present caregivers. Qualitative content analysis revealed three main categories with corresponding categories and sub-categories. Main Category One-Decisions pertaining to self-yielded two categories: decision pertaining to the offering of self and care for the caregiver. Main Category Two-Decisions pertaining to the patient-yielded three categories: decisions about care and treatment, living arrangements, and protecting the patient from harm. Main Category Three-Communication and relationships in decisions-yielded two categories: navigation and negotiations. The results of this study will inform healthcare providers and caregivers as they work together to anticipate, prepare, and plan for care management decisions over the AD trajectory.Marie Truglio-Londrigan, Jason T.
