    Understanding the mechanism involved in lung epithelial potassium current modulation by ozone

    Given that the ionic balance of the pulmonary alveolar lining fluid is essential for correct gas exchange, our previous studies analyzed the effects of ozone (O3), one of the most harmful pollutants for the respiratory system, on K+ current (IK)in human cultured lung epithelial cells (A549). O3 exposure significantly alters the flow of K+ ions, reducing the outward rectifier current component. O3 does not act directly, but through its byproducts, among which the main ones are 4-hydroxy-2-nonenal (4HNE) and hydrogen peroxide (H2O2). In this study we analyzed the action of 4HNE and H2O2 on IK in human lung cells and verified the protective effect of catalase. Material: A549 cellular line. 30’ exposure: O3: 0.1 ppm; 4HNE: 5, 10 and 20 μM; H2O2: 20 μM. Glucose-oxidase: 10 mU/l, 1.00h exposure, analyzed after 24h. Catalase: 1000 U, 1.30h exposure. Methods: Western blot, immunofluorescence and patch clamp techniques to study the action of the bio-products on IK. 4HNE was able to significantly decrease the IK, but it was less effective than O3. H2O2 produced by the cells after GO administration completely reproduced the O3 effect. Catalase showed its ability in preserving the outward rectifier component depressed by O3, bringing back the current to the control level. We can conclude that 4HNE and H2O2 are responsible for the O3 action on potassium channels. The protective role of catalase confirms the ability of O3 bio-product to modify the cellular redox homeostasis. In perspective it will be interesting to analyze their interaction, and the bio-molecular pathways activated

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Effect of Cu- and Fe- Isolated from Environmental Particulate Matter on Mitochondrial Dynamics in Human Colon CaCo-2 Cells

    Atmospheric particulate matter (PM) is one of the most dangerous air pollutants of anthropogenic origin; it consists of a heterogeneous mixture of inorganic and organic components, including transition metals and polycyclic aromatic hydrocarbons. Although previous studies have focused on the effects of exposure to highly concentrated PM on the respiratory and cardiovascular systems, emerging evidence supports a significant impact of air pollution on the gastrointestinal (GI) tract by linking exposure to external stressors with conditions such as appendicitis, colorectal cancer, and inflammatory bowel disease. In general, it has been hypothesized that the main mechanism involved in PM toxicity consists of an inflammatory response and this has also been suggested for the GI tract. In the present study, we analyzed the effect of specific redox-active PM components, such as copper (Cu) and iron (Fe), in human intestinal cells focusing on ultrastructural integrity, redox homeostasis, and modulation of some mitochondrial-related markers. According to our results, exposure to Cu- and Fe-PM components and their combination induced ultrastructural alterations in the endoplasmic reticulum and in the mitochondria with an additive effect when combined. The increase in ROS and the loss of the mitochondrial mass in the cells exposed to PM indicates that mitochondria are a target of acute metal exposure. Furthermore, the gene expression and the protein levels of mitochondria dynamics markers were affected by the PM exposure. In particular, OPA1 increases at both gene and protein levels in all conditions while Mitofusin1 decreases significantly only in the presence of Fe. The increase in PINK expression is modulated by Fe, while Cu seems to affect mainly Parkin. Finally, a significant decrease in trans-epithelial resistance was also observed. In general, our study can confirm the correlation observed between pollution exposure areas and increased incidence of GI tract conditions

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
