    The lost plant fossil and fish locality of “Monte Colle”, near the village of Bolca, Verona province, northern Italy has been considered to be of Eocene age since the middle of the nineteenth century. However, upon re-examination of the plant fossils, especially the specimens of Aularthrophyton Massalongo, which closely resemble the fossil conifer Frenelopsis Schenk, led us to suspect that a Cretaceous age was more probable. Fieldwork to re-evaluate the local stratigraphy, and the identification of the radiolarians Crucella cachensis Pessagno and Patellula helios (Squinabol) within the matrix of surviving hand specimens, all definitively show that the fossil bed actually belongs to the uppermost Cenomanian Bonarelli Level. With this revised age, we properly describe the gross morphology of the surviving plant specimens and reinterpret their identifications and affinities. Frenelopsis petraepurae comb. nov., Geinitzia sp., and a single angiosperm leaf type are described and the consequences for nomenclature outlined.     Investigating RhoA Signaling Defects in Tuberous Sclerosis Complex

    Senior Honors Thesis, Department of Molecular & Cell Biology, University of Wisconsin-MadisonTuberous sclerosis complex (TSC) is an autosomal dominant neurodevelopmental disorder caused by the monoallelic inactivation of TSC1 or TSC2, often resulting in intellectual disabilities, autism spectrum disorders, and epilepsy. The TSC1-TSC2 protein complex is known to negatively regulate mTORC1-mediated protein synthesis and activate mTORC2-mediated cytoskeletal rearrangements, both essential for proper neural development. Previous research from the Gomez lab has found that human forebrain neurons differentiated from patient-derived TSC2+/- induced pluripotent stem cells (iPSCs) exhibit axon outgrowth and guidance cue sensitivity defects independent of mTORC but attributed to diminished RhoA signaling. To further explore this, we examined TSC2+/+ and TSC2-/- iPSC-derived stem cells and neurons and observed significantly decreased RhoA activity in the TSC2-/- lines, indicated by reduced myosin light chain phosphorylation. We also identified morphological differences in TSC2-/- neurons relevant for future rescue experiments. Furthermore, we utilized a Tet-inducible, Halo-tagged TSC2 lentivirus (LV) to restore TSC2 expression in null cells and assessed rescue of both signaling and morphological defects. Future experiments will investigate TSC2 activity and localization in live cells during repulsive guidance cue stimulation, assess whether LV-mediated TSC2 expression can rescue RhoA activity and axon guidance defects, and identify the key TSC2 domains responsible for regulating RhoA. Understanding these mechanisms may reveal therapeutic targets for TSC-related neurodevelopmental defects

    Guido Löhr's recent article makes several insightful and productive suggestions about how to proceed with the empirical study of collective action. However, their critique of the conclusions drawn in Gomez-Lavin & Rachar (2022) is undermined by some issues with the interpretation of the debate and paper. This discussion article clears up those issues, presents new findings from experiments developed in response to Löhr's critiques, reflects on the role of experimental research in the development and refinement of philosophical theories, and adds to Löhr's suggestions about the path forward.Final article published

    Verification and computation in restricted Tile Automata

    Many models of self-assembly have been shown to be capable of performing computation. Tile Automata was recently introduced combining features of both Celluar Automata and the 2-Handed Model of self-assembly both capable of universal computation. In this work we study the complexity of Tile Automata utilizing features inherited from the two models mentioned above. We first present a construction for simulating Turing Machines that performs both covert and fuel efficient computation. We then explore the capabilities of limited Tile Automata systems such as 1-Dimensional systems (all assemblies are of height 1) and freezing Systems (tiles may not repeat states). Using these results we provide a connection between the problem of finding the largest uniquely producible assembly using n states and the busy beaver problem for non-freezing systems and provide a freezing system capable of uniquely assembling an assembly whose length is exponential in the number of states of the system. We finish by exploring the complexity of the Unique Assembly Verification problem in Tile Automata with different limitations such as freezing and systems without the power of detachment

    Complexity of Verification in Self-Assembly with Prebuilt Assemblies

    We analyze the complexity of two fundamental verification problems within a generalization of the two-handed tile self-assembly model (2HAM) where initial system assemblies are not restricted to be singleton tiles, but may be larger pre-built assemblies. Within this model we consider the producibility problem, which asks if a given tile system builds, or produces, a given assembly, and the unique assembly verification (UAV) problem, which asks if a given system uniquely produces a given assembly. We show that producibility is NP-complete and UAV is coNP^{NP}-complete even when the initial assembly size and temperature threshold are both bounded by a constant. This is in stark contrast to results in the standard model with singleton input tiles where producibility is in P and UAV is in coNP for (1) bounded temperature and coNP-complete when temperature is part of the input. We further provide preliminary results for producibility and UAV in the case of 1-dimensional linear assemblies with pre-built assemblies, and provide polynomial time solutions
