Synthesis and modification of pamam dendrimers as histone replacement molecules
The primary objective of this work was to synthesise and structurally modify a series of PAMAM dendrimers with a view to investigating and controlling their complexation with, and compaction of, DNA. The Introduction gives an overview to the origins of polymer and dendrimer science, and describes the synthetic approaches and resulting properties of dendrimers together with their impact on biological systems. The preparation of poly(amidoamine) (PAMAM) dendrimers utilising a number of different core materials are reported and characterised in Chapter 2. The conformational motion of the dendritic branches has been studied using 1H spin-lattice relaxation time experiments, and the results have been compared for the differently cored materials. Chapter 3 examines the modification of the dendrimer surfaces with acetyl, poly(ethylene glycol) and fluorescent groups and reports the results of a study of how the degree of DNA condensation affects gene expression by analysing dendrimer-DNA compaction using linear dichroism, dynamic light scattering and competitive binding of ethidium. Experiments designed to investigate modified dendrimer-lipid interactions are also discussed and the results of these are compared to molecular dynamic simulations. The synthesis of novel small molecules based around L-histidine and photo-responsive azobenzene, and the use of these to modify the surface of 4th and 5th generation PAMAM dendrimers is reported in Chapters 4 and 5 respectively. There follows a final chapter containing detailed descriptions of the synthetic procedures and characterisation for each of the compounds studied and the experimental methods used for exploring their behaviou
Co-creating an online TimeBank for inclusive research
Participatory and inclusive approaches to research have become more common as researchers recognize the benefits of enabling the meaningful involvement of representative community users in the development of accessible technologies. One of the major challenges in this context is the how the involvement of community members can be appropriately supported in project-related activities: payment for time and contributions is a particularly difficult and longstanding issue. This paper discusses the inclusive development of an online Timebank involving community members with intellectual disabilities. The TimeBank is conceived as a tool that enables people to contribute their different expertise on the basis of time, rather than monetary reward. The development process of the TimeBank is de-scribed as well as the challenges faced by the research team. There is much potential in the Timebank idea, although considerable further research is needed to establish an accessible, usable, credible and trustworthy resource
Organic Tinkering - Edinburgh Festival - International Festival Collaboration Week workshop on the theme of Art and Technology:Biological tinkering, kludging and modelling
In this project, we are developing a workshop, (August 5th-9th), in collaboration with some of the leading proponents of the Synthetic Biology movement, faculty members at the University of Edinburgh and the Edinburgh College of Art. The workshop will be held in Edinburgh as part of the Art and Technology theme of the Edinburgh Festival – and will bring together faculty from multiple disciplines, including Design, Art, Biology, Engineering, and Computer Science, incubating them together in this week-long workshop. Collaborators on this project are Alan Murray, Head of the School of Design, and Co-Director of Design Informatics; and Alistair Elfick, Director - Synthetic & Systems Biology & School of Engineering, University of Edinburgh; Anna Rabinowicz, Parsons - the New School for Design, New York and RabLabs Design
