Conclusion 14DoctoralmicroRNAs play an important role in cancer metabolisms. miRNA-181a-5p has decreased expression, particularly in non small cell lung cancer tissues and mesenchymal like lung cancer cells. Epithelial-mesenchymal transition mechanisms and cancer metabolism are regulated by the same signaling pathway. Functional analysis was performed to identify the functional role of miRNA-181a-5p in cancer metabolism and to find the target gene. To define the role of miRNA-181a-5p in cancer metabolism, we performed LDH assay, glucose uptake assay, Oil Red O staining, and mitochondrial ATP synthase inhibitor assay. The target gene of miRNA-181a-5p was determined by luciferase reporter assay, qRT-PCR, and western blot analyses. The Functional effect of miRNA-181a-5p on cancer metastasis was measured by migration and invasion assays. Experimental results showed that overexpression of miRNA-181a-5p reduced aerobic glycolysis and adipocytes and decreased cancer cell invasion and migration. Expression of SIRT1 and ACSL4 was reduced by the binding of miRNA-181a-5p to the 3’-UTR. These results suggest that miRNA-181a-5p might participate in the reprogramming of cancer metabolism

CONCLUSION 23 REFERENCES 24 국문요약 32MasterBackground and Objectives: The EGFR is a member of the erbB family of receptor tyrosine kinase proteins, which also includes HER2/neu (erbB2), HER3 (erbB3), and HER4 (erbB4). High levels of EGFR expression are correlated with poor prognosis and resistance to radiation therapy in a variety of cancers, mostly in head and neck squamous cell carcinoma (HNSCC). So we examined the expression of EGFR in hypopharyngeal cancer and the effect of EGF in progression of hypopharyngeal SCC in the hypopharyngeal SCC cell line (FaDu). Materials and Methods: We performed immunohistochemical stains on 57 specimens of each normal mucosa and hypopharyngeal SCC with EGFR antibody. For Western blot, fresh normal and cancer tissue from the hypopharynx obtained from three patients were used. We performed RT-PCR and Western blot analyses of FaDu cells. Proliferative of the FaDu cells was assayed by counting the number of the cells after treatment by EGF of different concentration of 0, 10, 30 ng/㎖. Dispersion of the cells was observed by measuring the separation and morphologic changes of the cells after colony of FaDu cells was formed in the media and then was treated with EGF of 10ng/㎖ or 30ng/㎖ for 24 hours. Tumor cell invasiveness was assessed by the membrane invasion assay. RT-PCR and zymography were performed to examine the roles of matrix metalloproteinase-2(MMP-2) and MMP-9, as well as the relationship between EGF and MMPs in FaDu invasiveness. Results: The positive rates of EGFR expression in hypopharyngeal SCC were 63.2%. EGFR staining was significantly correlated with pathologic stage (p<0.05). The increased expresssion of EGFR mRNA(RT-PCR) and protein (Western Blot) were detected in hypopharyngeal cancer tissue. Exogenous EGF significantly enhanced the growth of FaDu in a dose-dependent manner 30ng/㎖ (p<0.05). EGF markedly enhanced the invasiveness of cancer cells in a Transwell invasion chamber in a dose-dependent manner (p < 0.05). MMP expression was detected in the FaDu cells, and exogenous EGF enhanced the expression and its activity in the RT-PCR and zymogram analysis. Conclusions: These results suggest that EGF may play an important role in the progression of hypopharyngeal cancer

    Improvement of strength and water absorption of Interlocking Compressed Earth Bricks (ICEB) with addition of Ureolytic Bacteria (UB)

    Interlocking Compressed Earth Brick (ICEB) are cement stabilized soil bricks that allow for dry stacked construction. This characteristic resulted to faster the process of building walls and requires less skilled labour as the bricks are laid dry and lock into place. However there is plenty room for improving the interlocking bricks by increase its durability. Many studies have been conducted in order to improve the durability of bricks by using environmentally method. One of the methods is by introducing bacteria into bricks. Bacteria in brick induced calcite precipitation (calcite crystals) to cover the voids continuously. Ureolytic Bacteria (UB) was used in this study as a partial replacement of limestone water with percentage of 1%, 3% and 5%. Enrichment process was done in soil condition to ensure the survivability of UB in ICEB environment. This paper evaluates the effect of UB in improving the strength and water absorption properties of ICEB and microstructure analysis. The results show that addition of 5% UB in ICEB indicated positive results in improving the ICEB properties by 15.25% in strength, 14.72% in initial water absorption and 14.68% reduction in water absorption. Precipitation of calcium carbonate (CaCo3) in form of calcite can be distinguish clearly in microstructure analysis

    A very pleasing book. The Golden Axe is Aesopic (though never acknowledged here as such) and nicely elaborated and illustrated for this culture. Dokbo lives in a village tucked away in the mountains. The artist misses, I believe, that Dokbo lost the head of his axe. Yunbo does not hear the whole story and so proceeds falsely with the old man water spirit.
