Population structure of the fungus Sclerotinia sclerotiorum in common bean fields of Argentina
S. sclerotiorum is the causal agent of the white mould disease on common bean crops, one of the most threatening fungal diseases occurring across major bean production regions. Currently, there are no commercial cultivars with high levels of genetic resistance to white mould. The aim of this study was to analyse the population structure of 109 isolates of S. sclerotiorum from six dry bean fields in the main production area of Argentina using nine microsatellite loci. A total of 30 multilocus haplotypes (MLHs) were identified, of which 18 MLHs were unique. The remaining 12 MLHs were constituted by 83% of the isolates, six MLHs of which (composed of 75 isolates) were shared at least between two locations. Population genetic structure analysis and discriminant analysis of principal components (DAPC) identified two genetic clusters (subpopulations). The genetic cluster 1 (GC 1) was composed of 21 isolates and 15 MLHs. Similarly, the genetic cluster 2 (GC 2) was composed of 23 isolates and 15 MLHs. These two genetic clusters were observed in most locations sampled. Low levels of genetic differentiation (ΦST = 0.198; P 1) between genetic clusters were observed. Linkage disequilibrium analysis showed that one of the two genetic subpopulations was under linkage equilibrium (P > 0.001), which is consistent with recombinant populations. These results suggest the occurrence of both modes of reproductive behaviour, clonal and recombining, compromising the durability of management strategies for white mould disease in common bean cultivars.EEA SaltaFil: Aban, Carla Luciana. Characterization of the anthracnose resistance gene present in the Andean cultivar Perla
Biotic stress is a major cause of yield losses in common bean production worldwide. Anthracnose (ANT), caused by the fungus Colletotrichum lindemuthianum (Sacc & Magnus) Briosi & Cavara is among the diseases that affect beans and can cause yield losses of up to 100%, especially when environmental conditions are favorable for the development of the disease (Singh and Schwartz, 2010). The use of disease resistance genes is the most economical and ecological strategy for disease control. Finding and characterization of ANT resistance genes in different materials and available cultivars is important (Mahuku and Riascos, 2004). To date different anthracnose resistance genes were identified in common bean, most of them in Mesoamerican germplasm. Thus, characterization of new Andean sources of resistance is important for bean breeding. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variabilidad genotípica de aislamientos Sclerotinia sclerotiorum en lotes de cultivo de poroto en Salta y Jujuy
En los últimos años el Moho Blanco del poroto, causado por el hongo Sclerotinia sclerotiorum, ha adquirido gran importancia en la región del Noroeste Argentino (NOA), ya que la enfermedad se presenta con mayor severidad y se ha detectado un incremento en el área de prevalencia. Los daños económicos producidos por S. sclerotiorum en el cultivo de poroto pueden llegar al 80% y alcanzar el 100% bajo condiciones ambientales favorables para el desarrollo del patógeno en cultivares sensibles. El objetivo del presente trabajo fue caracterizar la variabilidad genotípica de aislamientos de S. sclerotiorum en las principales áreas de producción de poroto del NOA. Para ello, se realizó un relevamiento en 6 lotes de cultivo de poroto en Salta y Jujuy, obteniéndose un total de 116 aislamientos que fueron analizados mediante técnicas moleculares y de compatibilidad micelial (MCG). Empleando ADN obtenido a partir de micelio los aislamientos se identificaron como S. sclerotiorum analizando las secuencias ITS-ADNr. Además se realizó la amplificación de ADN mediante PCR empleando 11 cebadores URP (Universal Rice Primers) observándose gran variabilidad genética entre los aislamientos evaluados. Por otro lado, se determinaron los MCG contrastando cada aislamiento con sí mismo y contra todos los demás, identificándose un total de 52 MCGs, los cuales fueron únicos para cada lote. Los resultados revelan la existencia de gran variabilidad entre y dentro de los lotes analizados y representan un aporte importante para la búsqueda de fuentes de resistencia a la enfermedad.Fil: Aban, Carla Luciana. Characterization of fungal pathogens and germplasm screening for disease resistance in the main production area of the common bean in Argentina
The common bean (Phaseolus vulgaris L.) is the most important grain legume in the human diet, mainly in Africa and Latin America. Argentina is one of the five major producers of the common bean in the world, and the main cultivation areas are concentrated in the northwestern provinces of this country. Crop production of the common bean is often affected by biotic factors like some endemic fungal diseases, which exert a major economic impact on the region. The most important fungal diseases affecting the common bean in Argentina are white mold caused by Sclerotinia sclerotiorum, angular leaf spot caused by Pseudocercospora griseola, web blight and root rot caused by Rhizoctonia solani, which can cause production losses of up to 100% in the region. At the present, the most effective strategy for controlling these diseases is the use of genetic resistance. In this sense, population study and characterization of fungal pathogens are essential for developing cultivars with durable resistance. In this review we report diversity studies carried out on these three fungal pathogens affecting the common bean in northwestern Argentina, analyzing more than 200 isolates by means of molecular, morphological and pathogenic approaches. Also, the screening of physiological resistance in several common bean commercial lines and wild native germplasm is reviewed. This review contributes to the development of sustainable management strategies and cultural practices in bean production aimed to minimize yield losses due to fungal diseases in the common bean.Fil: Taboada, Gisel María. Variabilidad genética en germoplasma de tabaco Tipo Virginia empleando marcadores moleculares
Dentro de la actividad agraria de la provincia de Salta, el cultivo de tabaco Nicotiana tabacum, tiene una apreciable participación en la economía regional, tanto desde el punto de vista de la producción y exportaciones provinciales, como por su relevancia en la generación de empleo. El objetivo del trabajo fue conocer la variabilidad genética existente entre líneas de tabaco Virginia conservadas en el Banco de germoplasma del NOA (BANOA) y variedades comerciales para aportar nuevos materiales a los programas de mejoramiento. Se analizaron 5 entradas de tabaco Virginia del BANOA y 12 variedades comerciales de importancia para la región. La extracción de ADN se realizó empleando un protocolo de CTAB modificado a partir de hojas de plántulas de tabaco mantenidas en almácigo flotante. A partir del ADN extraído se realizó la amplificación mediante PCR empleando marcadores microsatélites distribuidos en todo el genoma de tabaco. Los productos de amplificación se visualizaron en geles de poliacrilamida 10% y se tiñeron con GelRed. Se analizó un total de 24 primers de los cuales el 33% mostró polimorfismo entre las líneas y variedades. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Molecular versus morphological markers to describe variability in sugar cane (Saccharum officinarum) for germplasm management and conservation = Marcadores moleculares y morfológicos para la descripción de variabilidad en caña de azúcar (Saccharum officinarum) con fines de manejo y conservación de germoplasma
Sugarcane is one of the most important industrial crops in tropical and subtropical regions. INTA (Argentina) administrates a Sugarcane Germplasm Bank and carries out a breeding program. The current study was designed to assess the phenotypic and genetic diversity among 65 sugarcane accessions selected from the INTA. Clustering and ordination methods based on quantitative and qualitative morphological traits and SSR data were applied. Generalized Procrustes Analysis allowed evaluating the correlation between relationships established with both markers A good of fit between dendrograms and similarity matrices were revealed by high cophenetic coefficients (r=0.82, p<0.0001; r=0.73, p<0.0001; r=0.82, p<0.0001 for phenotypic quantitative, phenotypic qualitative and molecular data respectively). The presence of different reliable population structure was observed when considering different data sources. Procrustes allowed finding those accessions that should have been responsible for the low correlation found between the individual configurations (73%). Both morphologic and molecular markers resulted discriminative enough to differentiate among accessions. It was not possible, however, to correlate associations of markers with the origin of materials. Phenotypic and genetic distances based on morphology and molecular information serves to assist conservation and organization of collection of materials, and the choice of parent combinations for breeding purposes.La caña de azúcar es uno de los cultivos industriales más importantes de regiones tropicales y subtropicales. El INTA (Argentina) administra un Banco de Germoplasma de caña de azúcar y lleva a cabo un programa de mejora. El presente trabajo fue diseñado para estimar la variabilidad fenotípica y genética entre 65 accesiones de caña de azúcar seleccionadas del INTA. Se aplicaron métodos de clasificación y ordenamiento en el análisis de datos morfológicos y de SSR. EL Análisis de Procrustes Generalizado permitió evaluar la correlación entre las relaciones establecidas a partir de ambos tipos de marca-dores. Un buen ajuste entre los dendrogramas y las matrices de similitud fue soportado por un alto coeficiente de correlación cofenética (r=0,82, p<0,0001; r=0,73, p<0,0001; r=0,82, p<0,0001 para datos cuantitativos, cualitativos y moleculares respectivamente). La presencia de una estructura poblacional fue reconocida cuando se consideraron los diferentes tipos de datos. El Procrustes permitió detectar aquellas accesiones que serían responsables de la baja correlación detectada entre configuraciones individuales (73%). Tanto los marcadores morfológicos como los moleculares resultaron lo suficientemente discriminativos para diferenciar accesiones. No obstante, no fue posible correlacionar las asociaciones establecidas por los marcadores con el origen de los materiales.
