Cyclosporine A loaded SLNs: Evaluation of cellular uptake and corneal cytotoxicity

    No full text
Cyclosporine A (CsA) loaded solid lipid nanoparticles (SLNs) for topical ophthalmic applications were prepared by high shear homogenization and ultrasound method using Compritol 888 ATO, Poloxamer 188 and Tween 80, to investigate the cellular uptake of rabbit corneal epithelial cells (RCE) and to evaluate the cytotoxicity. The size of the optimized formulation was 225.9±5.5nm with a polydispersity index of 0.253±0.05. The zeta potential and entrapment efficiency was detected as −16.9±0.7mV and 95.6%, respectively. The CsA release was found to be enzyme (lipase/co-lipase complex) dependent. SLNs were sterilized at 110 and 121 ◦C. The cytotoxicity was evaluated in vitro by means of RCE cells and was higher at 121 ◦C sterilization temperature, probably due to a supposed leakage of Tween 80 following lipid recrystallization. The permeation and penetration of CsA across/into the corneal cells were evaluated using in vitro and ex vivo experiments. The cellularuptakewas investigated by replacing CsA with the fluorescent dye Rhodamine B. The penetration enhancement properties were supported by confocal laser scanning microscopy analysis. The internalization of SLNs in cornea and in RCE cell lines was confirmed, pointing out the possibility of CsA targeting to the cornea

The relationship between mindfulness and resilience: The mediating role of self compassion and emotion regulation in a sample of underprivileged Turkish adolescents

WOS: 000456764200049The purpose of this study was to examine a mindfulness model of resilience through the mediating effects of self compassion and difficulties in emotion regulation among underprivileged Turkish adolescents. The sample of the study consisted of 752 students (426 female, 326 male) who aged 14-19 (M = 15.82, SD = 0.88). A path analysis was conducted in order to examine the proposed model. The results showed mindfulness to be a positive and significant predictor of self-compassion while negatively and significantly predicting difficulties in emotion regulation. Furthermore, self-compassion and difficulties in emotion regulation were found to be significant predictors of resilience. Indirect paths from mindfulness to resilience (via self-compassion and via difficulties in emotion regulation) were also significant in the model. The hypothesized model explained 21% of the variance in resilience scores.Scientific and Technological Research Council of Turkey (TUBITAK)This study was financially supported by The Scientific and Technological Research Council of Turkey (TUBITAK) under my PhD Thesis process. Objective: The purpose of this research was to translate the Infant Feeding Intentions Scale (IFI) into Turkish, modify it according to the Turkish cultural state, and detect the reliability and validity of the translated version. Materials and Methods: The research data was collected in the pregnancy polyclinic from 128 healthy pregnant women at a university hospital. A personal information form and a Turkish version of the IFI scale (IFI-T) were used for data collection. Language equivalence, content validity, construct validity, and predictive validity were used in the validation research of the IFI-T Scale. In the reliability research, Cronbach's alpha coefficient, item analysis, split-test, and re-test methods were used. The t-test was used for the dependent groups to test the time invariance of the scale. Results: The consistency of specialist opinion on the scale, translated into Turkish and back-translated, was revealed (Kendall W=0.600; p>0.05). The factor loading values resulting from the factor analysis that was directed at the construct validity of the scale were between 0.69 and 0.89 and the items were collected under one factor. The scale total correlation value of all items was over 0.20. In the reliability research, the Cronbach's alpha value was 0.86. The test-retest and split-half test results also supported the reliability of the scales. Conclusion: We found that the IFI-T scale can be used as a valid and reliable tool for evaluating the infant feeding intention in pregnant women
