    Unexpected predictors of antibiotic resistance in housekeeping genes of Staphylococcus aureus

    Methicillin-resistant Staphylococcus aureus (MRSA) is currently the most commonly identified antibiotic-resistant pathogen in US hospitals. Resistance to methicillin is carried by SCCmec genetic elements. Multilocus sequence typing (MLST) covers internal fragments of seven housekeeping genes of S. aureus. In conjunction with mec typing, MLST has been used to create an international nomenclature for S. aureus. MLST sequence types with a single nucleotide polymorphism (SNP) considered distinct. In this work, relationships among MLST SNPs and methicillin/oxacillin resistance or susceptibility were studied, using a public data base, by means of cross-tabulation tests, multivariable (phylogenetic) logistic regression (LR), decision trees, rule bases, and random forests (RF). Model performances were assessed through multiple cross-validation. Hierarchical clustering of SNPs was also employed to analyze mutational covariation. The number of instances with a known methicillin (oxacillin) antibiogram result was 1526 (649), where 63% (54%) was resistant to methicillin (oxacillin). In univariable analysis, several MLST SNPs were found strongly associated with antibiotic resistance/susceptibility. A RF model predicted correctly the resistance/susceptibility to methicillin and oxacillin in 75% and 63% of cases (cross-validated). Results were similar for LR. Hierarchical clustering of the aforementioned SNPs yielded a high level of covariation both within the same and different genes; this suggests strong genetic linkage between SNPs of housekeeping genes and antibiotic resistant associated genes. This finding provides a basis for rapid identification of antibiotic resistant S. arues lineages using a small number of genomic markers. The number of sites could subsequently be increased moderately to increase the sensitivity and specificity of genotypic tests for resistance that do not rely on the direct detection of the resistance marker itself

    Impact of spatial dispersion, evolution, and selection on Ebola Zaire Virus epidemic waves

    Ebola virus Zaire (EBOV) has reemerged in Africa, emphasizing the global importance of this pathogen. Amidst the response to the current epidemic, several gaps in our knowledge of EBOV evolution are evident. Specifically, uncertainty has been raised regarding the potential emergence of more virulent viral variants through amino acid substitutions. Glycoprotein (GP), an essential component of the EBOV genome, is highly variable and a potential site for the occurrence of advantageous mutations. For this study, we reconstructed the evolutionary history of EBOV by analyzing 65 GP sequences from humans and great apes over diverse locations across epidemic waves between 1976 and 2014. We show that, although patterns of spatial dispersion throughout Africa varied, the evolution of the virus has largely been characterized by neutral genetic drift. Therefore, the radical emergence of more transmissible variants is unlikely, a positive finding, which is increasingly important on the verge of vaccine deployment

    Antibiotic resistance is a significant global health concern that affects both human and animal populations. The One Health approach acknowledges the interconnectedness of human health, animal health, and the environment. It emphasizes the importance of collaboration and coordination across these sectors to tackle complex health challenges such as antibiotic resistance. In the context of One Health, antibiotic resistance refers to the ability of bacteria to withstand the efficacy of antibiotics, rendering them less effective or completely ineffective in treating infections. The emergence and spread of antibiotic-resistant bacteria pose a threat to human and animal health, as well as to the effectiveness of medical treatments and veterinary interventions. In particular, One Health recognizes that antibiotic use in human medicine, animal agriculture, and the environment are interconnected factors contributing to the development and spread of antibiotic resistance. For example, the misuse and overuse of antibiotics in human healthcare, including inappropriate prescribing and patient non-compliance, can contribute to the selection and spread of resistant bacteria. Similarly, the use of antibiotics in livestock production for growth promotion and disease prevention can contribute to the development of antibiotic resistance in animals and subsequent transmission to humans through the food chain. Addressing antibiotic resistance requires a collaborative One Health approach that involves multiple participants, including healthcare professionals, veterinarians, researchers, and policymakers

    Fabrication of Solid Oxide Fuel Cells (SOFCs) electrolytes by Electrophoretic Deposition (EPD) was the target of our study. For such purpose, thin layers of Yttria Stabilized Zirconia (YSZ) were electrophoritically deposited on pre-sintered NiO-YSZ pellets. A thin graphite film on each pellet was painted to make it conductive and can act as a working electrode for EPD. In this research acetylacetone-ethanol mixture (1:1 by vol.) was selected as the solvent and by applying different electrical fields (25, 50, 75, 100V/cm) in different deposition times (6, 3, 2, 1.5 min respectively), the obtained layers were examined. It was revealed that although electrical field multiplied deposition time remained constant in our experiments, the thicknesses of the deposited films were increased with increasing the applied electrical field which could not be explained easily by the Hamaker equation. For the next step, sintering of the deposited layers which prepared by applying 75 V/cm electrical field during 2 minutes was investigated. It was found that although the sintering processes were activated at 1250oC, 2 hours soaking time was not enough to produce a non-permeable layer. On the contrary, the layer which sintered at 1400oC for 2 hours was dense and crack-free.     S rostoucí potřebou sociální interakci mezi studenty, zejména mezi zahraničními, se stává online svět stále zajímavějším místem pro budování komunity a sdílení zkušeností. Tato bakalářská práce se zaměřuje na vývoj vícejazyčné webové aplikace pro seznamování studentů vysokých škol. Cílem práce je navrhnout a implementovat aplikaci, která by pomohla studentům snadněji navazovat nové sociální vztahy. Vývoj aplikace byl řízen vodopádovým modelem, který zahrnuje fáze analýzy požadavků, návrhu, implementace, testování a nasazení. Konkrétně jsou v práci použity technologie jako Vue3.js, SASS, TailwindCSS, Axios, Typescript, Vuetify, C#, .NET, Entity Framework a LINQ. Práce obsahuje sedm kapitol, které pokrývají různé aspekty vývoje a implementace aplikace. Každá kapitola se detailně věnuje určitému tématu, včetně analýzy požadavků, návrhu architektury, implementace back-endu a front-endu, testování a nasazení aplikace do provozu. Přímo na návrhu uživatelského rozhraní a marketingové části aplikace autor spolupracoval s externím grafickým designérem, což přispělo k profesionálnějšímu vzhledu a uživatelsky přívětivému prostředí. Výsledkem této práce je plně funkční webová aplikace s názvem studate, dostupná na adrese studate.eu, která umožňuje studentům vysokých škol z různých zemí navazovat nové sociální kontakty prostřednictvím online prostředí. Autor této práce hodlá spustit aplikaci do běžného provozu a propagovat ji mezi různými univerzitami v evropských zemích.With the growing need for social interaction among students, especially international students, the online world is becoming an increasingly interesting place to build community and share experiences. This bachelor thesis focuses on the development of a multilingual web application for introducing university students. The aim of the thesis is to design and implement an application that would help students to establish new social relationships more easily. The development of the application was guided by a waterfall model that includes the phases of requirements analysis, design, implementation, testing and deployment. Specifically, the technologies used in this work include Vue3.js, SASS, TailwindCSS, Axios, Typescript, Vuetify, C#, .NET, Entity Framework, and LINQ. There are seven chapters in the thesis that cover different aspects of application development and implementation. Each chapter covers a specific topic in detail, including requirements analysis, architecture design, back-end and front-end implementation, testing, and deploying the application into production. The author collaborated directly with an external graphic designer on the user interface design and marketing part of the application, which contributed to a professional look and user-friendly environment. The result of this work is a fully functional web application called studate, available at studate.eu, which allows university students from different countries to make new social contacts through an online environment. The author of this thesis intends to launch the application into regular operation and promote it among different universities in European countries
