Educare all'umanità
Mitochondria and metabolism: the role of SAGA complex in budding yeast respiration
Mitochondrion is the energy center of the cell in which different metabolic pathways converge. Cell respiration takes place in this organelle with high production of ATP. Nucleus-mitochondrion communication represents an essential level of cell homeostasis regulation. This crosstalk is strictly related to epigenetic mechanisms, in particular to histone and non-histone proteins acetylation. Budding yeast is a suitable model system to study the interplay between epigenetic and metabolism; in this organism it is possible to analyse fermentative and respiratory pathways varying the carbon source in growth medium. SAGA complex is a transcriptional co-activator with two catalytic domains: the deubiquitination module with DUB Ubp8 and the acetylation domain with HAT Gcn5. In yeast, Gcn5 has a major role in protein acetylation at global and specific lysine; moreover, it is implicated in mitochondrion-nucleus communication. In this work, we found that SAGA complex and its catalytic activities are necessary for cell respiration. Results showed that KAT Gcn5 and DUB Ubp8 are upregulated in respiration at both transcriptional and protein levels. We identify two interactors, the E3 ubiquitin ligase Psh1 and the HDAC Hda1, whose deletion is able to rescue defective respiratory phenotype of strains lacking respectively Ubp8 and Gcn5. We demonstrated that histone residue H3K18 is the preferential target of Hda1 and Gcn5 and its acetylation is needed in respiration. Surprising, we found that Gcn5, Ubp8 and Psh1 localize in both nucleus and mitochondria highlighting a dual role as chromatin modifiers and mitochondrial factors. Moreover, our data showed that Ubp8 expression is regulated by Gcn5, while its cell localization is influenced by Psh1. Finally, results suggest that acetylation is an important mark in respiratory condition

Joint location and pricing within a user-optimized environment
In the design of service facilities, whenever the behaviour of customers is impacted by queueing or congestion, the resulting equilibrium cannot be ignored by a firm that strives to maximize revenue within a competitive environment. In the present work, we address the problem faced by a firm that makes decisions with respect to location, service levels and prices and that takes explicitly into account user behaviour. This situation is modelled as a nonlinear mathematical program with equilibrium constraints that involves both discrete and continuous variables, and for which we propose an efficient algorithm based on an approximation that can be solved for its global optimum
Intra-articular injections for the treatment of osteoarthritis: focus on the clinical use of hyaluronic acid.
Osteoarthritis (OA), also called degenerative joint disease, is the most frequently occurring chronic musculoskeletal disease, particularly affecting the aging population. The use of viscosupplementation, i.e. intra-articular (IA) hyaluronic acid (HA) drug therapy, to treat OA, is growing worldwide, due to important results obtained from several clinical trials, which reported IA HA-related improvements in functional activity and pain management. This review is an update of the IA use of this compound in the treatment of OA, with clinical evidence from the last few years being discussed and used to delineate new trends for the future
Ten years of feasibility pump, and counting
The Feasibility Pump (fp) is probably the best-known primal heuristic for mixed-integer programming. The original work by Fischetti et al. (Math Program 104(1):91–104, 2005), which introduced the heuristic for 0–1 mixed-integer linear programs, has been succeeded by more than twenty follow-up publications which improve the performance of the fp and extend it to other problem classes. Year 2015 was the tenth anniversary of the first fp publication. The present paper provides an overview of the diverse Feasibility Pump literature that has been presented over the last decade
An exact algorithmic framework for a class of mixed-integer programs with equilibrium constraints
In this study, we consider a rich class of mathematical programs with equilibrium constraints (MPECs) involving both integer and continuous variables. Such a class, which subsumes mathematical programs with complementarity constraints, as well as bilevel programs involving lower level convex programs is, in general, extremely hard to solve due to complementarity constraints and integrality requirements. For its solution, we design an (exact) algorithmic framework based on branch-and-bound (B&B) that treats each node of the B&B tree as a separate optimization problem and potentially changes its formulation and solution approach by designing, for example, a separate B&B tree. The framework is implemented and computationally evaluated on a specific instance of MPEC, namely a competitive facility location problem that takes into account the queueing process that determines the equilibrium assignment of users to open facilities, and a generalization of models for which, to date, no exact method has been proposed
The Italian reception of Miguel de Unamuno, between criticism and translations
The aim of this article is to consider the continuity of the presence of the work of Miguel de Unamuno in the Italian cultural system, from the appearance of the first translations and articles on him until today, to investigate whether it is possible to reconstruct an evolution of the interpretive tendencies. Due to the heterogeneous nature of the Italian contributions on the Basque author, a mainly chronological approach has been adopted to consider, decade after decade, what convergences may have been established or what kind of cultural operations may have been carried out. We divide our reconstruction into three stages: (1) from 1901 until Unamuno’s death in 1936, a period in which we can appreciate an almost monolithic appreciation of his figure; (2) from the 1940s to the 1980s, a phase characterized by an initial critical rest and by the greater possibility of historical-literary contextualization of his work as a whole, thanks also to the academic institutionalization of Italian Hispanism; (3) from the 90s to the present day, years in which Unamuno’s critical reception has been marked by a copious amount of studies and translations that confirm the longue durée of the author’s presence in the ranks of the great foreign writers
