    Relationships Between Supervisor Behaviour and Subordinates' Work Attitudes: The Role of LMX

    This study investigated the relationship between supervisor behaviour and the work experience of subordinate employees and assessed the role of the interpersonal exchange relationship between supervisor and subordinate. Participants completed a questionnaire asking them to rate their supervisor’s behaviour (initiating structure, tolerance of subordinate independence, perceived support, integration and consideration of subordinates), the quality of their leader-member exchange relationship (LMX) with their supervisor, perceived job autonomy, and role ambiguity. To assess subordinates’ work experience, respondents were also asked to report on their level of work engagement, citizenship behaviour, job motivation, commitment to the organisation, and team commitment. Two hundred and fifteen responses were collected and, following a factor analysis, mediation analyses were conducted using the supervisor behaviours as predictors, LMX, autonomy, and ambiguity as mediators and subordinate attitudes and behaviour as outcomes. The results provided support for the proposed mediated relationships with 30 out of 36 indirect relationships being significant. The findings confirmed that 1) supervisory behaviour had indirect effects on subordinate attitudes and behaviour. 2) LMX, job autonomy, and role ambiguity significantly predicted subordinate employees’ work attitudes and behaviour. 3) The behaviours of supervisors helped determine LMX, job autonomy and role ambiguity. These results confirmed that there is a strong relationship between the behaviour of supervisors and the work experience of subordinate employees. Although causation cannot be inferred based on the results of this study, the findings indicate that the supervisor may contribute to the work experience of subordinates. The findings may suggest that if supervisors were to change their role behaviour by the way they deal with subordinates and introduce structure in the workplace this my help reduce subordinates’ role ambiguity, raise perceived job autonomy, lead to a higher quality exchange relationship, and positively influence work outcomes. This would enhance employee work engagement, organisation-directed citizenship behaviour, intrinsic motivation, commitment to organisation and work team benefiting both the organisation and the employees. Longitudinal research into an integrated model of supervisor behaviour and LMX using experimental or observational study designs is recommended in order to develop a model of causal relationships between supervisor behaviour, LMX, and subordinate outcomes. Further investigation of the measurement of supervisor behaviour may be appropriate in order to validate or revise the LBDQ scales, since factor analyses on these scales prompted substantial changes. Additionally, the results of this study indicated that perceived supervisor support and LMX may not be distinct constructs. Further research into the measurement and theoretical grounding of these measures is recommended

    A presente investigação tem por foco de análise conhecer alguns aspectos e comportamentos da profissionalidade dos professores supervisores de estudantes estagiários do EB 1.º Ciclo. Procurámos compreender, não todos os diversos elementos que interagem no processo de supervisão, nem descobrir qual o modelo de profissionalidade do professor supervisor, mas, sobretudo, conhecer os aspectos pedagógicos e didácticos do professor supervisor da actualidade. Na sua centralidade o estudo mostra a necessidade de se continuar a discutir a profissionalidade do professor. Assim, embora não sendo o desenvolvimento profissional a categoria principal explorada neste estudo, procuramos identificar através das tarefas prioritárias a desenvolver junto dos alunos, áreas relevantes no processo de formação do estagiário, ciclo supervisivo e aspectos valorizados no desempenho do estagiário, alguns aspectos do professor supervisor, na convicção de que estes, ao serem conhecidos, vão demarcando uma etapa do processo de desenvolvimento da profissionalidade dos professores supervisores e dos futuros professores do EB 1.º Ciclo. Os dados recolhidos para o enquadramento teórico, fundamentados na revisão bibliográfica e na natureza desta investigação, permitiram analisar a acção do professor supervisor, tendo como base os temas: supervisão pedagógica, ser professor do EB 1.º Ciclo e profissionalidade docente. A definição do conceito de profissionalidade não é fácil, e ao tratar temáticas relacionadas com a mesma, muitas abordagens podem ser consideradas, não obstante interessou-nos alguns aspectos pedagógicos e didácticos. Realizou-se uma investigação, essencialmente, do tipo descritivo, tendo a recolha de dados sido feita através de dez entrevistas e de vinte e um questionários, aplicados a professores supervisores do EB 1.º Ciclo, na Região Autónoma dos Açores – Ilha de São Miguel, no ano de 2007/2008. Tanto nas entrevistas como nos questionários observamos um certo equilíbrio nas respostas dadas. Os resultados indicaram que a acção do professor supervisor do EB 1.º Ciclo se aproxima de um modelo eclético, baseado em várias abordagens, conservando alguns aspectos do modelo tradicional e incorporando aspectos do modelo reflexivo.ABSTRACT: This present research is the analyses the knowing of some aspects of supervisors teachers of interns student EB 1.º Cycle, professional behavior. We have tried to understand, not all the various elements that interact in the process of supervision, or figure out what model of professionalism of the supervisor teacher, but above all, meet the pedagogical aspects of teaching and teacher supervisor of the day. In its center the study shows the need to continue discussing the teacher’s the professionalism. Although, the professional development it’s not the main category explored in this study, we tried to indentify through the priority tasks to be developed with the students in the areas relevant to the trainee, supervised cycle and valued aspects of the intern's performance, some aspects of supervisor teacher, in the belief that they, to be known, will marking a step in the development of teacher’s the professionalism and teacher of supervisors EB 1st. Cycle. Data collected for the theoretical framework, based on the literature and the nature of the research, helped analyze the action of the teacher supervisor, based on the themes: pedagogical supervision, be a teacher of EB 1 and professionalism of teacher. The precise definition of professionalism is not easy, and when dealing with issues relating to it, many approaches can be considered, Therefore some learning and teaching aspects were considered interesting. There was an investigation, essentially, descriptive, where data collection was done through interviews of ten and twenty-one questionnaires administered to teachers supervisors EB 1st Cycle in the Azores - Sao Miguel, in the year 2007/2008.     Determining the competency dimensions of supervisor professor from the nursing students’ viewpoints: A qualitative study

    Introduction: With regard to the importance of students’ curricular guiding and its effect on their educational process and the quality of university, the prophecy of supervisor professor is very important in students’ professional growth. Thus, this study was accomplished with the purpose of assigning the competency dimensions of supervisor professors from the nursing students’ viewpoints in 2009. Methods: In this qualitative study, the data were gathered through focus group discussion. Fifteen nursing students were selected by purposive sampling and their experiences and viewpoints were surveyed for assigning the competencies of supervisor professors until achieving data saturation. After that, the interviews content was recorded with the participants’ permission. Thematic analysis was performed to identify the categories and subcategories found in the nursing students’ descriptions. Results: According to the analysis of the recorded interviews, 2 major categories were emerged: 1) Competency dimensions and roles of supervisor professor including personal and ethical competency, education and research management competency, and leadership and motivating competency and 2) Barriers of effectiveness of supervisor professor performance were determined. Conclusion: According to the results, being supportive, a good listener, and professional commitment were introduced as personal and ethical competencies of supervisor professor and facilitating the professionalism of nursing students is the important role in this dimension. Supervising on the students’ educational situation, making them familiar with research, and motivating them were discussed as management and leadership competencies. The participants emphasized on the continuous performance appraisal of supervisor professor, determining students’ special empowerments and introducing their responsibility domains in the beginning of entrance to a college, as well as establishing in–service education in order to enhance performance quality of supervisor professor

    A Taxonomy of Network and Computer Attack Methodologies

    Since the invention of computers and networks, people have found various ways to attack them. Attacks over the years have ranged from using a sledge hammer on a computer, to advanced distributed denial of service attacks. This research focuses on computer and network attacks and providing a taxonomy of them. This is to help combat new attacks, improve computer and network security and to provide consistency in language when describing attacks. A wide range of computer and network attacks are examined to provide both a survey of the field and to provide a basis on which to build the proposed taxonomy. The proposed taxonomy consists of four dimensions which provide a holistic taxonomy and to deal with inherent problems in the computer and network attack field. The first dimension covers the attack vector and the main behaviour of the attack. The second dimension allows for classification of the attack targets. Vulnerabilities are classified in the third dimension and payloads in the fourth. The taxonomy is briefly evaluated and is found to work well, with a few areas that could be improved.     O Programa institucional de Bolsa de Iniciação à Docência (PIBID) é uma iniciativa essencial para o desenvolvimento da educação e da formação de professores. Para estas finalidades, os alunos de licenciatura são levados as escolas públicas de ensino básico, sobre a coordenação de um docente de ensino superior, e a supervisão de um professor da escola campo. Assim, buscou-se compreender a importância e contribuições do professor supervisor para a efetivação desse programa, bem como entender como a criação desse “espaço híbrido” entre o ensino superior e o esnino básico contribuíram para o desenvolvimento educacional de todos os envolvidos no processo, focando os estudos principalmente no professor supervisor. Notou-se que esses professores supervisores, a cumprir bem o seu papel, não so ensinavam a prática escolar para os bolsistas do PIBID, como também evoluíam suas metodologias e participavam mais ativamente do ambiente escolar, mostrando que o programa contribui para o desenvolvimento da formação inicial e continuada de professores

    Towards NMR analysis of the HIV-1 coreceptor CCR5 and its interaction with RANTES

    CCR5 is a chemokine receptor together with CD4 used by HIV-1 as a primary gate of cell infection. For this reason CCR5 is of great interest for medicine as a target for the anti-HIV-1 therapies. Since the binding site of its endogenous ligand RANTES overlaps with the binding site of viral envelope glycoprotein gp120, a noninflamatory RANTES derivative 5P12-RANTES has been developed as an anti-HIV-1 infection microbicide. The primary aim of this thesis was to establish an NMR-amenable system to study CCR5 and to understand better the interaction with RANTES. For this purpose CCR5 expressed in insect cells was characterized in detail with regards to its secondary structure, oligomeric state, particle size, stability, posttranslational modifications and functionality. In contrast to the previous results, carefully performed detergent screening revealed that FosCholine-12, a detergent which allows high yield purification, does not support CCR5 recognition by 2D7 and cannot be used for studying CCR5 interactions with ligands. Therefore for the functional studies the receptor was solubilized with a milder detergent mixture DDM/CHAPS/CHS, which was shown to support native CCR5 tertiary structure. Using this setup it could be shown by SPR that 5P12-RANTES binds with higher affinity than another potent RANTES variant PSC-RANTES (Morin et al., manuscript in preparation). This explains why 5P12-RANTES, which unlike PSC-RANTES does not cause CCR5 internalization, is an equally effective anti-HIV-1 microbicide. On the other hand, the wild-type RANTES was shown to aggregate on the receptor micelle using a mechanism compatible with the linear oligomerization, a process that is proposed to serve local chemokine preconcentration. To obtain an access to a cost-efficient source of isotope-labeled samples, an E. coli expression system was established for CCR5 (Wiktor et al., 2012, J Biomol NMR, in revision). The expression was facilitated by fusing the N-terminus of CCR5 to well expressing protein domains e.g. thioredoxin. The C-terminal CCR5 truncation and the mutation of cysteines increased the protein yield up to 10 mg/L and improved the sample stability. Due to the engineered thrombin proteolytic site the N-terminal fusion partner i.e. thioredoxin could be quantitatively cleaved and removed by size exclusion chromatography. The FC-12-purified receptor was abundant in alpha-helical secondary structure but could bind RANTES, MIP-1beta and conformation-dependent antibody 2D7 only when solubilized by a DDM/CHAPS/CHS mixture. Using 15N,13C,2H-labeled CCR5 2D and 3D NMR experiments were recorded but only about 80 backbone resonances could be resolved. The spectral quality was jeopardized by large overlap and line-broadening and needs further improvements to allow the assignment and the structural investigation. To study 5P12-RANTES by NMR the backbone assignment was completed. The HSQC spectrum revealed that, unlike wild-type RANTES and other chemokines, 5P12-RANTES does not form dimers. The secondary chemical shift analysis suggest that the overall structure of 5P12-RANTES is similar to the wild-type RANTES, with the exception of the mutated N-terminus, which does not participate in the intermolecular beta-sheet and was shown to be highly flexible. Another important observation was that RANTES secondary structure is perturbed by Fos-Choline detergents, whereas maltosides shift the RANTES monomer:dimmer equilibrium towards its monomeric form. The last part of the thesis present an independent study, where using ubiquitin as an example the mechanism of protein unfolding is studied (Vajpai et al., 2012, Proc Natl Acad Sci USA, in revision) manuscript submitted for publication). The secondary chemical shift analysis showed that the alcohol-denatured ubiquitin structure closely resembles the cold- and pressure denatured structure. This suggests that alcohol, low temperature and pressure unfold proteins by reducing the hydrophobic effect, the cost of exposing hydrophobic residues.
