Four-laser scanning confocal system for microarray analysis.
We have constructed a confocal scanner suitable for routine microarray analysis from commercially available parts. We have outlined the details that should be considered when designing such an instrument and listed some of the specific components comprising the system [the full list of system components is available on CD from the corresponding author (D.J.G.) at no charge]. Here, we describe the methods used to test the linearity and sensitivity of the instrument. Performance was evaluated with two commonly used dyes, fluorescein and Cy5. While the instrument had a linear correlation between the dye concentration and fluorescence intensity, the observed deviation from a slope of 1.0 underscores the importance of running multipoint calibration experiments to obtain accurate dye quantitation over the full dynamic range of the scanner. This method has utility in testing commercial instruments in addition to the scanner described here. An array with over 300 spots dyed with Cy3 was scanned with our instrument and a high-end commercial instrument. The agreement between the two instruments was very good over a 1000-fold intensity range. Our scanner is a cost-effective alternative to more costly commercial scanners with similar capabilities

Identification of Ultra-hot Jupiters in the PLATO field and perspectives on their characterization
openGli Hot Jupiter (HJ) e gli Ultra-Hot Jupiter (UHJ) rappresentano una classe peculiare di esopianeti: si tratta di giganti gassosi in orbite estremamente ravvicinate alle loro stelle ospiti, con periodi orbitali inferiori ai dieci giorni e temperature superficiali che, nel caso degli UHJ, superano i 2200 K. Questa tesi tratta della fisica che governa gli HJ e UHJ, con particolare attenzione ai meccanismi di formazione, alle conseguenze fisiche delle loro temperature estreme e alla loro composizione atmosferica. Dopo una panoramica introduttiva sugli HJ e sulla loro classificazione, l’analisi si concentra su tre UHJ noti in letteratura: WASP-121b, KELT-25b e HATS-70b; di ciascuno vengono esaminate le proprietà orbitali, atmosferiche e spettroscopiche. I tre pianeti appartengono al primo campo di vista di PLATO (PLAnetary Transits and Oscillations of stars), futura missione spaziale dell'ESA (lancio previsto per fine 2026), che sarà in grado di individuare migliaia di nuovi pianeti, inclusi di tipo terrestre.
Nella tesi sono presentate le principali caratteristiche della missione e il potenziale contributo alla scoperta e caratterizzazione di nuovi HJ e UHJ. L’osservazione sistematica di questi pianeti estremi fornirà informazioni fondamentali per comprendere l’evoluzione dei giganti gassosi in condizioni limite e i processi fisici ad essi legati
