    The modulating role of sex and anabolic-androgenic steroid hormones in cannabinoid sensitivity

    Cannabis is the most commonly used illicit drug worldwide. Although its use is associated with multiple adverse health effects, including the risk of developing addiction, recreational and medical cannabis use is being increasing legalized. In addition, use of synthetic cannabinoid drugs is gaining considerable popularity and is associated with mass poisonings and occasional deaths. Delineating factors involved in cannabis use and addiction therefore becomes increasingly important. Similarly to other drugs of abuse, the prevalence of cannabis use and addiction differs remarkably between males and females, suggesting that sex plays a role in regulating cannabinoid sensitivity. Although it remains unclear how sex may affect the initiation and maintenance of cannabis use in humans, animal studies strongly suggest that endogenous sex hormones modulate cannabinoid sensitivity. In addition, synthetic anabolic-androgenic steroids alter substance use and further support the importance of sex steroids in controlling drug sensitivity. The recent discovery that pregnenolone, the precursor of all steroid hormones, controls cannabinoid receptor activation corroborates the link between steroid hormones and the endocannabinoid system.     Metabolic and mitogenic functions of fibroblast growth factor 1

    Type 2 diabetes (T2D) is a worldwide health problem and its treatment is mostly ineffective, signifying the need to develop new therapies that can improve metabolic health. Fibroblast growth factor 1 (FGF1), a hormone-like protein, has been identified as a potential novel drug canditiate for the treatment of T2D. In diabetic mice, pharmacological administration of recombinant FGF1 protein shows remarkable antidiabetic activity; however, the involved mechanism of action remain largely unclear. Besides, administration of FGF1 also stimulates cell growth, which is a major obstacle for its future therapeutic application. In my thesis, we reported the results of preclinical studies aimed to provide details on FGF1’s mechanism of action and safety. Our results indicate that injected recombinant FGF1 protein activates a glucose-regulatory network in the mouse brain that communicates with the muscle and results in an instant correction of the diabetic phenotype. Regarding the safety of FGF1 administration, we found that drug side effects can be substantially reduced by generating mutant FGF1 proteins. Collectively, these preclinical studies may guide the development of safe and effective FGF1-based biologicals

    The anabolic steroid nandrolone alters cannabinoid self-administration and brain CB1 receptor density and function

    Clinical and pre-clinical observations indicate that anabolic-androgenic steroids can induce neurobiological changes that alter the rewarding effects of drugs of abuse. In this study, we investigated the effect of the anabolic steroid nandrolone on the rewarding properties of the cannabinoid CBI receptor agonist WIN55,212-2 (WIN) in rats. Lister Hooded male rats were treated intramuscularly with nandrolone (15 mg/kg) or vehicle for 14 consecutive days, and then allowed to self-administer WIN (12.5 g/kg/infusion) intravenously. After reaching stable drug intake, self-administration behavior was extinguished to examine drug- and cue-induced reinstatement of cannabinoid-seeking behavior. Other behavioral parameters presumed to influence drug-taking and drug-seeking behaviors were examined to gain more insight into the behavioral specificity of nandrolone treatment. Finally, animals were sacrificed for analysis of 031 receptor density and function in selected brain areas. We found that nandrolone-treated rats self-administered up to 2 times more cannabinoid than vehicle treated rats; but behaved similarly to control rats when tested for drug- and cue-induced reinstatement of cannabinoid-seeking behavior. Enhanced cannabinoid intake by nandrolone-treated rats was not accompanied by changes in locomotor activity, sensorimotor gating, or memory function. However, our molecular data show that after chronic WIN self-administration nandrolone-treated rats display altered CBI receptor density and function in selected brain areas. We hypothesize that increased cannabinoid self-administration in nandrolone-treated rats results from a nandrolone-induced decrease in reward function, which rats seem to compensate by voluntarily increasing their cannabinoid intake. Altogether, our findings corroborate the hypothesis that chronic exposure to anabolic-androgenic steroids induces dysfunction of the reward pathway in rats and might represent a potential risk factor for abuse of cannabis and other drugs in humans.     Ginnasio: Home furniture to support physical exercise

    This graduation project is a demonstration of the potentialities of a theory-based approach to design for mood regulation, which is based on the belief that design can influence mood by enabling and stimulating people to engage in effective mood-regulating activities.For this project, the western home bathroom was chosen as a context of research, due to the personal interest of the author in the relaxing properties of bathing, and bathing related activities such as the Finnish sauna, and the Turkish steam bath.The goal of the project was kept very broad in the beginning and gradually narrowed down during the analysis phase. A significant contribution to the creation of a well-defined design goal was given by the realization that, while relaxing practices are usually well accommodated in the domestic environment, little attention is paid to activities that influence people’s mood towards a high energy state such as exercising. Eventually, the project’s objective became to design a piece of furniture, in which physical exercise-supporting features coexist with more relax-oriented attributes. This product would aim to make high energy state inducing activities an integral part of the everyday domestic routine.Three concepts were generated by following an incremental process in which insights from one idea served as a starting point for the next. The third and last concept was then developed using a broad variety of techniques such as sketching, collage making, digital visualization, prototyping, and testing.The final result is a piece of furniture to exercise and to relax on, which form stands somewhere in between a bench and a chaise longue. Its name is Ginnasio, a reference to the building that, in Ancient Greece, functioned as a training facility, but also as a place to relax, socialize and engage in philosophical discussion

To be a leader must go through various stages so as to be a good leader not born but made, the process of making someone a leader is called the science of leadership. Leadership can not be separated by discipline, because it is a very close relationship. This thesis is an exposition, the research to explain and explain the object under study. The data used is data that has been collected at the time of the author carry out the practice screen for one year. The data obtained will be processed and arranged systematically and regularly. In solving the problem, the authors argue that all officers on board must apply good leadership characteristics and instill discipline in them first. A good boss must also understand leadership based on management theory, and make experience as learning. The frequent misunderstanding over the MV. Pan Global when ABK received orders from officers. This is due to the large number of officers who do not use the approach or give a firm assignment that causes saturation and even a rebellious nature of the order.     In the light of the massive economic and social changes which have affected the North East of England over the last 25 years, the author assesses the vitality of the indigenous culture and reflects upon current cultural trends and the North East’s future, particularly in relation to a regional Europe. He traces the folk-tradition of the region and looks at ways in which this can be drawn upon to develop a meaningful link between past and present. He looks closely at the changing nature of class-relationships in the North East and reflects upon how a valid local culture can survive in a multi-cultural society. He draws upon his own extensive experience in Community Arts, looking at definitions of the term in the new political climate and arguing for its positive contribution to the cultural debate. He dwells on the issue of regionalism and devolution in a new Europe, comparing the situation in the North East of England with political and cultural changes in Scotland and other parts of the United Kingdom

    As one of the most popular musical instruments, drum kits' design has not changed a lot since it first emerged. However, the product contains some problems which significantly affect user experience. Not to improve the design, this project aims to dig deeper into the world of drum kit and drummers to identify their emotional values and related functional needs, hence to find new design possibilities for this classic musical instrument. Trend research, user research, and market research were conducted. The result was finalized into the fundamental emotion and related functional needs. The design vision was established, and the design was completed. The final design – Collector, is a conceptual drum kit with a new design language, which allows drummers to create their own sound library and is flexible to use in different scenarios. The design was evaluated with real drummers and was proved to be both practical and attractive. The concept shows enormous potential to be further developed into a commercial product.Integrated Product Desig

    Enabling high-quality plastic recycling with the Virtual Chemist: A device that analyses the composition of plastic waste

    In the past 40 years, an incomprehensible 8,3 billion metric tons of plastic has been produced and is primarily being discarded and incinerated, instead of being recycled. Only 10 per cent of this total amount has been recycled to date, therefore it was decided to focus on recycling of plastic waste to reduce plastic pollution which negatively impacts life on a global scale. Through an extensive analysis of the recycling context it was found that the biggest problem in plastic recycling is the inability to achieve a constant production of high-quality recyclate. With insufficient knowledge to address this found problem, it was decided to do an internship at Polytential BV, a start-up company located in Delft, the Netherlands. Their innovative product the Virtual Chemist, a device that analyses the composition of plastic waste up to 98,5 per cent accuracy, enables mechanical recyclers and compounders to analyse large volumes of plastic waste.The solution will facilitate a transition towards a data-driven recycling industry, increasing profitability in processing plastic waste by providing the industry with accurate and representative composition data, as processing large volumes is currently not possible. However, The Virtual Chemist was still in an early development stage (MVP) and required further development to be presented to clients as a sellable product (MMP). This was done by addressing the user needs and facilitating the right user experience, achieved through an analysis of the context, user, current system, and workflow to define a baseline for this project. Additionally, each step was completed by focussing on the user interaction and experience. The analysis results were used to formulate a design brief, which defined the goal and direction for the ideation and conceptualisation of the product. This report presents a summary of the completed design process, research findings, and concludes with recommendations that focus on further development of the Virtual Chemist.Design for Interactio
