    Dancer, Nun, Ghost, Goddess explores the story of the dancers Giō and Hotoke, which first appeared in the fourteenth-century narrative Tale of the Heike. The story of the two love rivals is one of loss, female solidarity, and Buddhist salvation. Since its first appearance, it has inspired a stream of fiction, theatrical plays, and visual art works. These heroines have become the subjects of lavishly illustrated hand scrolls, ghosts on the noh stage, and Buddhist and Shinto goddesses. Physical monuments have been built to honor their memories; they are emblems of local pride and centerpieces of shared identity. Two beloved characters in the Japanese literary imagination, Giō and Hotoke are also models that have instructed generations of women on how to survive in a male-dominated world

    Down syndrome (DS) is the most frequent constitutional form of intellectual disability in humans. In 1958 Jerome Lejeune discovered that children with DS have one extra copy of chromosome 21 (trisomy 21) in their cells. Lejeune considered the trisomic cells as ‘‘drug addicted’’, intoxicated by the excess of gene products that are formed due to the presence of the additional chromosome. If we knew the pathogenetic mechanisms we could devise a more specific therapy. Lejeune, a genial geneticist and biochemist, was also a pediatrician who visited thousands of children with intellectual disability, encouraging their families, highlighting the enrichment in humanity experienced by so many families because of the presence of a person with DS and not only the problems. He dedicated all of his life and so much love to these children, and at the same time he would correct the biochemical errors which hamper the full expression of their human abilities and he used to say: ‘‘Hate the disease, love the patient: that is the practice of medicine’’. To date, no therapy is recognized as being effective in improving the cognitive abilities of persons with DS. Research was slowed by the fact that so many studies have been focused on prenatal diagnosis of the syndrome rather than on its treatment. Following the encounter with Lejeune’s history and work, we have started a systematic study of DS to integrate clinical, biochemical, genetic and bioinformatic data in order to identify novel therapeutic targets for this form of trisomy

    Lo scopo di questa ricerca è la analisi bioinformatica e genetico-molecolare della espressione e dello splicing alternativo dei geni del cromosoma umano 21 (HC21), per comprendere il ruolo dei loro prodotti nella patogenesi della Sindrome di Down (DS). Lo splicing alternativo consiste nella produzione di differenti isoforme di mRNA a partire dallo stesso locus genico, e ha come conseguenza frequente la produzione di proteine strutturalmente e funzionalmente diverse codificate dallo stesso gene. In questo modo la medesima informazione genetica può essere espressa in modalità differenti, con un grande arricchimento della capacità codificante effettiva del genoma. Ricerche svolte negli ultimi anni hanno dimostrato che lo splicing alternativo è un fenomeno molto comune nei geni del genoma umano e in generale nei genomi dei vertebrati, può essere filogeneticamente conservato, è regolato in modo fine e le sue alterazioni sono correlate a svariate patologie. Poiché le differenze tra le varie forme di mRNA prodotte con questo meccanismo possono essere anche molto piccole, si rendono necessarie tecniche basate su una prima elaborazione bioinformatica della struttura delle isoforme e su una successiva analisi in vitro mediante ibridazione o amplificazione progettate in modo specifico per ciascuna isoforma. L'identificazione e l'analisi di tutti i geni di HC21 è al presente al centro dell'interesse negli studi volti a comprendere la patogenesi della DS, e include il problema di una revisione sistematica della modalità di espressione alternativa di tali geni. Gli obiettivi principali della ricerca sono: 1) Identificazione e caratterizzazione sistematica di isoforme di mRNA derivate da splicing alternativo di geni localizzati su HC21. 2) Studio della espressione differenziale delle isoforme di mRNA derivate da splicing alternativo. Saranno studiati con particolare riguardo i membri delle famiglie geniche "Down syndrome critical region 1-like" (DSCR1L) e "Cysteine and tyrosine rich protein 1" (CYYR1), descritte recentemente e ancora poco caratterizzate. 3) Analisi funzionale delle isoforme proteiche derivate da splicing alternativo mediante studio in vitro e impiego di organismi modello. Anche in questo caso saranno studiati con particolare riguardo i membri delle famiglie geniche sopra citate
